A simple, genotype-independent and efficient method for plant regeneration using shoot tip explants of pearl millet (Pennisetum glaucum) was established. Linsmaier and Skoog (LS) medium supplemented with 2,4-dichloro-phenoxyacetic acid (2.5 mg l(-1)) and kinetin (0. 2 mg l(-1)) was used for induction of embryogenic calli. Development of numerous somatic embryos was observed within 10 days after transferring onto Murashige and Skoog (MS) medium supplemented with 6-benzyl aminopurine (2 mg l(-1)) and indole 3-butyric acid (0. 5 mg l(-1)) under light (16 hr photoperiod). Histological observations confirmed the origin of somatic pro-embryoids and globular embryoids. Regenerated plants established in soil, grew normally and produced fertile seeds. RAPD analysis also revealed genetic uniformity of the regenerants. The short duration of time taken for regeneration (30-35 days) and its high frequency (78-87%) makes this system highly suitable for applications such as genetic transformation. 相似文献

The sordarins are fungal specific inhibitors of the translation factor eEF2, which catalyzes the translocation of tRNA and mRNA after peptide bond formation. We have determined the crystal structures of eEF2 in complex with two novel sordarin derivatives. In both structures, the three domains of eEF2 that form the ligand-binding pocket are oriented in a different manner relative to the rest of eEF2 compared with our previous structure of eEF2 in complex with the parent natural product sordarin. Yeast eEF2 is also shown to bind adenylic nucleotides, which can be displaced by sordarin, suggesting that ADP or ATP also bind to the three C-terminal domains of eEF2. Fusidic acid is a universal inhibitor of translation that targets EF-G or eEF2 and is widely used as an antibiotic against Gram-positive bacteria. Based on mutations conferring resistance to fusidic acid, cryo-EM reconstructions, and x-ray structures of eEF2, EF-G, and an EF-G homolog, we suggest that the conformation of EF-G stalled on the 70 S ribosome by fusidic acid is similar to that of eEF2 trapped on the 80 S ribosome by sordarin. 相似文献

subculturing to fresh medium. Protecting the medium from light by covering the surface with sterile aluminum foil significantly
improved leaf color and chlorophyll content by 50–60%. Raising the concentration of FeNa2EDTA (Fe-EDTA) in the light-exposed medium from the normal level (100 μM) to 150 or 200 μM significantly increased the leaf chlorophyll two- to threefold, suggesting that the light-induced effect was mainly attributable
to photo-oxidation of Fe-EDTA. Fe-EDTA supplementation also resulted in better expansion and lobe formation of upper leaves,
a reduction in internode length, an increase in the number of nodes, and a significant improvement in propagation rate. There
was also a reduction in axillary sprout growth at elevated Fe-EDTA levels, indicating a more prominent apical dominance effect
and a reduction in the number and vigor of tendrils. Increasing the Fe-EDTA level in the medium to 150 μM is suggested for routine micropropagation of triploid watermelon, in order to compensate for the photo-oxidative loss of

P elements are widely used as insertional mutagens to tag genes, facilitating molecular cloning and analyses. We modified a P element so that it carried two copies of the suppressor of Hairy-wing [su(Hw)] binding regions isolated from the gypsy transposable element. This transposon was mobilized, and the genetic consequences of its insertion were analyzed. Gene expression can be altered by the su(Hw) protein as a result of blocking the interaction between enhancer/silencer elements and their promoter. These effects can occur over long distances and are general. Therefore, a composite transposon (SUPor-P for suppressor-P element) combines the mutagenic efficacy of the gypsy element with the controllable transposition of P elements. We show that, compared to standard P elements, this composite transposon causes an expanded repertoire of mutations and produces alleles that are suppressed by su(Hw) mutations. The large number of heterochromatic insertions obtained is unusual compared to other insertional mutagenesis procedures, indicating that the SUPor-P transposon may be useful for studying the structural and functional properties of heterochromatin. 相似文献

Gametocyte maturation in Plasmodium falciparum is a critical step in the transmission of malaria. While the majority of parasites proliferate asexually in red blood cells, a small fraction of parasites undergo sexual conversion and mature over 2 weeks to become competent for transmission to a mosquito vector. Immature gametocytes sequester in deep tissues while mature stages must be able to circulate, pass the spleen and present themselves to the mosquito vector in order to complete transmission. Sequestration of asexual red blood cell stage parasites has been investigated in great detail. These studies have demonstrated that induction of cytoadherence properties through specific receptor-ligand interactions coincides with a significant increase in host cell stiffness. In contrast, the adherence and biophysical properties of gametocyte-infected red blood cells have not been studied systematically. Utilizing a transgenic line for 3D live imaging, in vitro capillary assays and 3D finite element whole cell modelling, we studied the role of cellular deformability in determining the circulatory characteristics of gametocytes. Our analysis shows that the red blood cell deformability of immature gametocytes displays an overall decrease followed by rapid restoration in mature gametocytes. Intriguingly, simulations suggest that along with deformability variations, the morphological changes of the parasite may play an important role in tissue distribution in vivo. Taken together, we present a model, which suggests that mature but not immature gametocytes circulate in the peripheral blood for uptake in the mosquito blood meal and transmission to another human host thus ensuring long-term survival of the parasite. 相似文献
