Abstract

P22 phage >105 PFU ml?1 could be used to inhibit Salmonella Typhimurium biofilm formation by 55–80%. Concentrations of EDTA >1.25?mM and concentrations of nisin >1,200?µg ml?1 were also highly effective in reducing S. Typhimurium biofilm formation (≥96% and ≥95% reductions were observed, respectively). A synergistic effect was observed when EDTA and nisin were combined whereas P22 phage in combination with nisin had no synergistic impact on biofilm formation. Triple combination of P22 phage, EDTA and nisin could be also used to inhibit biofilm formation (≥93.2%) at a low phage titer (102 PFU ml?1), and low EDTA (1.25?mM) and nisin (9.375?µg ml?1) concentrations. A reduction of 70% in the mature biofilm was possible when 107 PFU ml?1 of P22 phage, 20?mM of EDTA and 150?μg ml?1 of nisin were used in combination. This study revealed that it could be possible to reduce biofilm formation by S. Typhimurium by the use of P22 phage, EDTA and nisin, either alone or in combination. Although, removal of the mature biofilm was more difficult, the triple combination could be successfully used for mature biofilm of S. Typhimurium.  相似文献

A second order kinetic model was developed to predict the rate and extent of NH(4)(+) removal as struvite from anaerobic digester effluents. Alternative to this, NH(4)(+) can be recovered from struvite and the remaining Mg(2+) and PO(4)(3-) can be recycled back to the wastewater to fix more NH(4)(+). The NH(4)(+) solution was retained and the remaining Mg(2+) and PO(4)(3-) were returned back to be mixed with wastewater. In a five-step process, NH(4)(+) recovery was initially 92% and progressively decreased to 77% in the fifth stage, due to loss of Mg(2+) and PO(4)(3-) at each step in the supernatant. Finally, economic analysis of recycling nutrients was performed and compared to the one step process. The cost of NH(4)(+) recovery was calculated as $0.36/kgNH(4)-N which is lower than $7.7/kgNH(4)-N the cost of one step process without considering the market value of struvite obtained in one step process.  相似文献

J. Appl. Physiol.83(1): 189-195, 1997.Fish oil supplementation and physicalexercise may induce oxidative stress. We tested the effects of 8 wk of-tocopherol (vitamin E) and fish oil (FO) supplementation on resting and exercise-induced oxidative stress. Rats(n = 80) were divided into groupssupplemented with FO, FO and vitamin E (FOVE), soy oil (SO), and SO andvitamin E (SOVE), and for FOVE and SOVE they were dividedinto corresponding exercise groups (FOVE-Ex and SOVE-Ex). Lipidperoxidation [thiobarbituric acid-reacting substances(TBARS)] was 33% higher in FO compared with SO in the liver, butoxidative protein damage (carbonyl levels) remained similar in bothliver and red gastrocnemius muscle (RG). Vitamin E supplementation,compared with FO and SO, markedly decreased liver and RG TBARS, butliver TBARS remained 32% higher in FOVE vs. SOVE. Vitamin E alsomarkedly decreased liver and RG protein carbonyl levels, althoughlevels in FOVE and SOVE were similar. Exercise increased liver and RGTBARS and RG protein carbonyl levels markedly, with similar levels inFOVE-Ex and SOVE-Ex. FO increased lipid peroxidation but not proteinoxidation in a tissue-specific manner. Vitamin E markedly decreasedlipid peroxidation and protein oxidation in both FOVE and SOVE,although liver lipid peroxidation remained higher in FOVE. Despitehigher levels of hepatic lipid peroxidation at rest in FOVE comparedwith SOVE, liver appeared to be relatively less susceptible toexercise-induced oxidative stress in FOVE.

This article examines the major political debates in post-Soviet Azerbaijan vis à vis the very assumptions of individual autonomy, equality, national culture and citizenship, and universalism upon which modern nation-states have historically been based. The information presented in this article is based on personal interviews conducted with the leading and influential members of the Azerbaijani political elite in 1998. The interviews were based on two broad themes. The first relates to the perceptions of the Soviet period and on what grounds the Azerbaijanis differentiate the new Republic of Azerbaijan from the former Soviet Azerbaijan. The second relates to their perceptions of both the outside world and themselves with regards to differing understandings of nationalism, national culture, and national/ethnic or local identities. The study of the Azerbaijani example of post-Soviet political culture may help us to understand the dynamics of nation-building on the basis of the major political debates and of conflicting national, ethnic and local identities in Azerbaijan.  相似文献

In this study, the distribution, morphology and relative abundance of Sulfate Reducing Bacterial (SRB) and Methanogenic Archaeal (MA) populations in the Black Sea sediments were investigated by using in situ hybridization with fluorescently labeled rRNA-targeted oligonucleotide probes. Results were discussed with respect to the characteristics of sampling points. MA and SRB showed a great diversity in all sediment samples. Higher abundance of MA (20–30%) and SRB (30–35%) populations were observed within the sediments from deeper parts of the Black Sea than the shallower parts (10–11% MA and 13–14% SRB). Desulfobotulus, Desulfosarcina and Desulfococcus groups were the most commonly detected SRB groups in the Black Sea sediments. Relative percentage of these SRB groups within sediments from deeper parts of the Black Sea was in a range of 17–21% whereas that of was in a range of 4–5% within the sediments from the shallower parts. Order Methanococcales were the dominant methanogenic group in all samples. Relative percentages of order Methanococcales were in a range of 8–12% and 4–5% within sediments from deeper parts and the coastal parts of the Black Sea, respectively.  相似文献

Zn2+‐homoeostasis including free Zn2+ ([Zn2+]i) is regulated through Zn2+‐transporters and their comprehensive understanding may be important due to their contributions to cardiac dysfunction. Herein, we aimed to examine a possible role of Zn2+‐transporters in the development of heart failure (HF) via induction of ER stress. We first showed localizations of ZIP8, ZIP14 and ZnT8 to both sarcolemma and S(E)R in ventricular cardiomyocytes (H9c2 cells) using confocal together with calculated Pearson's coefficients. The expressions of ZIP14 and ZnT8 were significantly increased with decreased ZIP8 level in HF. Moreover, [Zn2+]i was significantly high in doxorubicin‐treated H9c2 cells compared to their controls. We found elevated levels of ER stress markers, GRP78 and CHOP/Gadd153, confirming the existence of ER stress. Furthermore, we measured markedly increased total PKC and PKCα expression and PKCα‐phosphorylation in HF. A PKC inhibition induced significant decrease in expressions of these ER stress markers compared to controls. Interestingly, direct increase in [Zn2+]i using zinc‐ionophore induced significant increase in these markers. On the other hand, when we induced ER stress directly with tunicamycin, we could not observe any effect on expression levels of these Zn2+ transporters. Additionally, increased [Zn2+]i could induce marked activation of PKCα. Moreover, we observed marked decrease in [Zn2+]i under PKC inhibition in H9c2 cells. Overall, our present data suggest possible role of Zn2+ transporters on an intersection pathway with increased [Zn2+]i and PKCα activation and induction of HF, most probably via development of ER stress. Therefore, our present data provide novel information how a well‐controlled [Zn2+]i via Zn2+ transporters and PKCα can be important therapeutic approach in prevention/treatment of HF.  相似文献

We report a study of the role of putative exopolysaccharide gene clusters in the formation and stability of Pseudomonas putida KT2440 biofilm. Two novel putative exopolysaccharide gene clusters, pea and peb, were identified, and evidence is provided that they encode products that stabilize P. putida KT2440 biofilm. The gene clusters alg and bcs, which code for proteins mediating alginate and cellulose biosynthesis, were found to play minor roles in P. putida KT2440 biofilm formation and stability under the conditions tested. A P. putida KT2440 derivative devoid of any identifiable exopolysaccharide genes was found to form biofilm with a structure similar to wild-type biofilm, but with a stability lower than that of wild-type biofilm. Based on our data, we suggest that the formation of structured P. putida KT2440 biofilm can occur in the absence of exopolysaccharides; however, exopolysaccharides play a role as structural stabilizers.  相似文献
