The cytochrome c oxidase complex (CcO) catalyzes the four-electron reduction of dioxygen to water by using electrons from ferrocytochrome c. Redox free energy released in this highly exergonic process is utilized to drive the translocation of protons across a transmembrane electrochemical gradient. Although numerous chemical models of proton pumping have been developed, few attempts have been made to explain the stepwise transfer of energy in the context of proposed protein conformational changes. A model is described that seeks to clarify the thermodynamics of the proton pumping function of CcO and that illustrates the importance of electron and proton gating to prevent the occurrence of the more exergonic electron leak and proton slip reactions. The redox energy of the CcO-membrane system is formulated in terms of a multidimensional energy surface projected into two dimensions, a nuclear coordinate associated with electron transfer and a nuclear coordinate associated with elements of the proton pump. This model provides an understanding of how a transmembrane electrochemical gradient affects the efficiency of the proton pumping process. Specifically, electron leak and proton slip reactions become kinetically viable as a result of the greater energy barriers that develop for the desired reactions in the presence of a transmembrane potential. 相似文献

The group A streptococcal emm-like genes, which encode the cell-surface M and M-like proteins, are divided into distinct mrp, emm and enn subdivisions and are clustered together in a region of the chromosome called the vir regulon. In order to understand the mechanisms involved in the evolution of emm-like genes, a 180bp fragment of the 5 variable region of the enn gene was characterized in 31 strains for which emm sequences and multilocus enzyme electrophoretic profiles have been previously determined. The results demonstrate that nucleotide polymorphisms at the enn locus are generated predominantly by point mutations and short deletions or insertions, and that variation among enn and emm genes has arisen by similar mechanisms. However, diversity at the enn locus is restricted in comparison to the emm locus. Moreover, there is strong evidence for intragenic recombination at the enn locus and the pattern of distribution of emm and enn alleles among strains suggests that these genes may be independently acquired by horizontal transfer and recombination from distinct donor strains, thereby generating a mosaic structure for the vir regulon. The results add to a growing body of evidence that horizontal gene transfer has played a major role in the evolution of Streptococcus pyogenes vir regulons. 相似文献

A total of 188 Bordetella strains were characterized by the electrophoretic mobilities of 15 metabolic enzymes and the distribution and variation in positions and copy numbers of three insertion sequences (IS). The presence or absence of IS elements within certain lineages was congruent with estimates of overall genetic relationships as revealed by multilocus enzyme electrophoresis. Bordetella pertussis and ovine B. parapertussis each formed separate clusters, while human B. parapertussis was most closely related to IS1001-containing B. bronchiseptica isolates. The results of the analysis provide support for the hypothesis that the population structure of Bordetella is predominantly clonal, with relatively little effective horizontal gene flow. Only a few examples of putative recombinational exchange of an IS element were detected. Based on the results of this study, we tried to reconstruct the evolutionary history of different host-adapted lineages. 相似文献

Drosophila melanogaster belongs to a closely related group of eight species
collectively known as the melanogaster subgroup; all are native to
sub-Saharan Africa and islands off the east coast of Africa. The
phylogenetic relationships of most species in this subgroup have been well
documented; however, the three most closely related species, D. simulans,
D. sechellia, and D. mauritiana, have remained problematic from a
phylogenetic standpoint as no data set has unambiguously resolved them. We
present new DNA sequence data on the nullo and Serendipity-alpha genes and
combine them with all available nuclear DNA sequence data; the total data
encompass 12 genes and the ITS of rDNA. A methodological problem arose
because nine of the genes had information on intraspecific polymorphisms in
at least one species. We explored the effect of inclusion/exclusion of
polymorphic sites and found that it had very little effect on phylogenetic
inferences, due largely to the fact that 82% of polymorphisms are
autapomorphies (unique to one species). We have also reanalyzed our
previous DNA-DNA hybridization data with a bootstrap procedure. The
combined sequence data set and the DNA-DNA hybridization data strongly
support the sister status of the two island species, D. sechellia and D.
mauritiana. This at least partially resolves what had been a paradox of
parallel evolution in these two species.
