We have previously determined that larval feeding experience with a feeding/oviposition deterrent modified the feeding responses of larvae and oviposition responses of subsequent moths. These behavioural changes were attributed to learning, but the possibility of chemical legacy could not be ruled out. In the present study, we have topically applied a feeding/oviposition deterrent plant extract from Hoodia gordonii (Masson) Sweet ex Decne (Asclepiadaceae) to larvae, pupae, and adults of Trichoplusia ni (Hübner) (Lepidoptera: Noctuidae) to determine whether the feeding response of larvae and oviposition response of subsequent female moths is similarly modified by chemicals applied to the external surface of the insect. Our results indicate that traces of the extract that may be present internally or externally on the larvae do not reduce the feeding deterrent response of larvae. Furthermore, traces of the extract in or on larvae, pupae, or adult moths did not alter oviposition choice of female moths, leading us to discount the role of experience through topical application in this study. The fact that feeding/oviposition choice was only influenced by prior feeding experience of the larvae and not by topical administration suggests that habituation via sensory stimulation through mouthpart chemosensilla is likely a central phenomenon. Continuous exposure of adult moths to the extract over a period of 7 days did not affect the oviposition response of the female moths, ruling out the role of adult experience on host-plant selection in T. ni . To the best of our knowledge, this is the first study to examine the role of experience via topical application of chemicals onto all life stages of the insect except the egg. Chemical legacy may not be playing a role in influencing the oviposition choices of female T. ni moths.  相似文献

Previous work has indicated that loss of ability of DNA dependent RNA polymerase, from stationary phase cultures of B. subtilis, to transcribe phage øe DNA was a sine qua non for sporulation. To ascertain if this change in template specificity was sporulation-specific, we repeated these experiments using a defined sporulation medium. The changes observed previously did not occur in the defined medium although sporulation was normal. The ability of the enzyme to transcribe other DNA templates was also examined. Similar studies were carried out using a polymerase from a rifamycin-resistant, sporulation conditional mutant. The significance of these findings with regard to the regulation of sporulation in B. subtilis is discussed.  相似文献

Phenological shifts are the most widely reported ecological responses to climate change, but the requirements to distinguish their causes (i.e. phenotypic plasticity vs. microevolution) are rarely met. To do so, we analysed almost two decades of parturition data from a wild population of North American red squirrels (Tamiasciurus hudsonicus). Although an observed advance in parturition date during the first decade provided putative support for climate change‐driven microevolution, a closer look revealed a more complex pattern. Parturition date was heritable [h2 = 0.14 (0.07–0.21 (HPD interval)] and under phenotypic selection [β = ?0.14 ± 0.06 (SE)] across the full study duration. However, the early advance reversed in the second decade. Further, selection did not act on the genetic contribution to variation in parturition date, and observed changes in predicted breeding values did not exceed those expected due to genetic drift. Instead, individuals responded plastically to environmental variation, and high food [white spruce (Picea glauca) seed] production in the first decade appears to have produced a plastic advance. In addition, there was little evidence of climate change affecting the advance, as there was neither a significant influence of spring temperature on parturition date or evidence of a change in spring temperatures across the study duration. Heritable traits not responding to selection in accordance with quantitative genetic predictions have long presented a puzzle to evolutionary ecologists. Our results on red squirrels provide empirical support for one potential solution: phenotypic selection arising from an environmental, as opposed to genetic, covariance between the phenotypic trait and annual fitness.  相似文献

During endometrial inflammation, bovine endometrium responds by increasing the production of pro-inflammatory mediators, such as interleukin 1 beta (IL-1β), tumor necrosis factor alpha (TNFα), and eicosanoids. The purpose of this study was to establish and characterize an in vitro model of endometrial inflammation using bovine endometrial epithelial (bEEL) and stromal (bCSC) cell lines. We evaluated the effects of the infectious agent (bacterial lipopolysaccharide; LPS) and pro-inflammatory mediators (IL-1β and TNFα) on eicosanoid biosynthesis pathway gene expression and production by bEEL and bCSC cells. Based on concentration-response experiments, the optimal concentrations for responses were 1?μg/mL LPS, 10?ng/mL IL-1β and 50?ng/mL TNFα. Real-time PCR results show that there was an upregulation of relative mRNA expression of PTGS2 when bEEL and bCSC were treated with LPS, IL-1β and TNFα. An increase in PTGES3 expression was observed when bEEL cells were treated with LPS and IL-1β and PTGES2 when treated with IL-1β. In bCSC cells, FAAH relative mRNA was decreased upon treatments. Rate of production of PGE2, PGF, PGE2-EA and PGF-EA were also determined using liquid chromatography tandem mass spectrometry. Our results show that eicosanoid production was increased in both cell lines in response to LPS, IL-1β, and TNFα. We suggest that the characteristics of bEEL and bCSC cell lines mimic the physiological responses found in mammals with endometrial infection, making them excellent in vitro models for intrauterine environment studies.  相似文献
