Survivin is an inhibitor of apoptosis overexpressed in different types of tumors and undetectable in most terminally differentiated normal tissues. In the current study, we sought to evaluate the in vitro theranostic properties of a molecular beacon-oligodeoxynucleotide (MB) that targets survivin mRNA. We used laser scanning confocal microscopy to study MB delivery in living cells and real-time PCR and western blot to assess selective survivin-targeting in human malignant melanoma cells. We further assess the pro-apoptotic effect of MB by measuring internucleosomal DNA fragmentation, dissipation of mitochondrial membrane potential (MMP) and changes in nuclear morphology. Transfection of MB into A375 and 501 Mel cells generated high signal intensity from the cytoplasm, while no signal was detected in the extracellular environment and in survivin-negative cells (i.e., human melanocytes and monocytes). MB time dependently decreased survivin mRNA and protein expression in melanoma cells with the maximum effect reached at 72 h. Treatment of melanoma cells with MB induced apoptosis by significant changes in MMP, accumulation of histone-complexed DNA fragments in the cytoplasm and nuclear condensation. MB also enhanced the pro-apoptotic effect of standard chemotherapeutic drugs tested at clinically relevant concentrations. The MB tested in the current study conjugates the ability of imaging with the pharmacological silencing activity against survivin mRNA in human melanoma cells and may represent an innovative approach for cancer diagnosis and treatment.  相似文献

This study aimed to analyze the impact of winter weather conditions on young children’s behavior and affective states by examining a group of 61 children attending day-care centers in Florence (Italy). Participants were 33 males, 28 females and their 11 teachers. The mean age of the children at the beginning of the observation period was 24.1 months. The day-care teachers observed the children’s behavioral and emotional states during the morning before their sleeping time and filled in a questionnaire for each baby five times over a winter period of 3 weeks. Air temperature, relative humidity, air pressure and solar radiation data were collected every 15 min from a weather station located in the city center of Florence. At the same time, air temperature and relative humidity data were collected in the classroom and in the garden of each day-care center. We used multilevel linear models to evaluate the extent to which children’s emotional and behavioral states could be predicted by weather conditions, controlling for child characteristics (gender and age). The data showed that relative humidity and solar radiation were the main predictors of the children’s emotional and behavioral states. The outdoor humidity had a significant positive effect on frustration, sadness and aggression; solar radiation had a significant negative effect only on sadness, suggesting that a sunny winter day makes children more cheerful. The results are discussed in term of implications for parents and teachers to improve children’s ecological environment.  相似文献

Annual production of age-1 and age-2 crayfish of three populations in the New River, West Virginia, was quantified and then compared to previously reported harvest of crayfish by a bait fishery. Sampling of crayfish within circular plots and age and growth analyses aided estimation of crayfish production. Orconectes sanbornii sanbornii has a two-year life cycle, and Cambarus sciotensis and Orconectes virilis had three-year life cycles. Production by age-1 and age-2 crayfish (all species) was about 7.0 g live weight m−2 yr−1; almost half of this production was by C. sciotensis. Greater crayfish density and therefore production occurred in riffles than in pools. Annual crayfish harvest by anglers and commercial catchers was equivalent to about 5% of annual crayfish production.  相似文献

Branchipus visnyai is presently known from only one location, in Italy, on Monte Catabio (central Appennine, Latium, 1590 m a.s.l). It lives in a large astatic pond and in a small temporary pool close to the pond. Typically two generations appear in the pond (in spring after snowmelt and in autumn after dilution by rainfall), unless an exceptionally dry season occurs. Yet, due to shallowness and small size, filling and duration of the pool is erratic and depends on the annual amount of snow and rainfall. Consequently, B. visnyai does not always appear in the pool.Field studies suggest that either a rise in temperature or a drop in conductivity may trigger cyst hatching in the two biotopes. Also, the species exhibits a high tolerance to change in environmental factors like water temperature. Hatching occurs within a range of 10–22 °C, adults survive at temperatures from 1 to 27 °C. The optimum seems to correspond to 10–20 °C.  相似文献

The association between the variability of phytoplankton biomass and community structure and the distribution of water masses around the Antarctic Peninsula were examined during austral summer 1993. Phytoplankton biomass showed high variability, and was dominated by an autotrophic flagellate (Cryptomonas sp.) that represented, on average, 91% of total phytoplankton biomass. The lowest phytoplankton biomasses were associated with the strongly mixed, saline, cold waters characteristic of the Weddell Sea water mass, and with the waters influenced by ice melt from the Bellingshausen Sea. The highest biomasses were found in the confluence of these water masses, where a front develops. Community composition also differed among water masses, with eukariotic picoplankton and diatoms having their highest relative contribution to community biomass in stations with Bellingshausen Sea and Weddell Sea water masses, whereas the abundance of Cryptomonas sp. was highest at the confluence of these waters. These results indicate that mesoscale processes, that determine water mass distribution, are of paramount importance in controlling the time and space variability of Antarctic phytoplankton.  相似文献
