Chronic airway inflammation is a hallmark of asthma, an immune-based disease with great societal impact. Honokiol (HNK), a phenolic neurotransmitter receptor (γ-aminobutyric acid type A) agonist purified from magnolia, has anti-inflammatory properties, including stabilization of inflammation in experimentally induced arthritis. The present study tested the prediction that HNK could inhibit the chronic inflammatory component of allergic asthma. C57BL/6 mice sensitized to and challenged with OVA had increased airway hyperresponsiveness to methacholine challenge and eosinophilia compared with naive controls. HNK-treated mice showed a reduction in airway hyperresponsiveness as well as a significant decrease in lung eosinophilia. Histopathology studies revealed a marked drop in lung inflammation, goblet cell hyperplasia, and collagen deposition with HNK treatment. Ag recall responses from HNK-treated mice showed decreased proinflammatory cytokines in response to OVA, including TNF-α-, IL-6-, Th1-, and Th17-type cytokines, despite an increase in Th2-type cytokines. Regulatory cytokines IL-10 and TGF-β were also increased. Assessment of lung homogenates revealed a similar pattern of cytokines, with a noted increase in the number of FoxP3(+) cells in the lung. HNK was able to alter B and T lymphocyte cytokine secretion in a γ-aminobutyric acid type A-dependent manner. These results indicate that symptoms and pathology of asthma can be alleviated even in the presence of increased Th2 cytokines and that neurotransmitter agonists such as HNK have promise as a novel class of anti-inflammatory agents in the treatment of chronic asthma. 相似文献

Hexavalent chromium Cr(VI) is a well known human carcinogen. This genotoxic metal induces DNA strand breaks and chromosome damage. However, the relationship between these lesions is uncertain. Our study focused on examining the role of XRCC1 in sodium chromate-induced cytotoxicity and chromosomal aberrations in Chinese Hamster Ovary (CHO) cells. Three different cell lines were used: AA8 (parental), EM9 (XRCC1 mutant) and H9T3 (EM9 complemented with human XRCC1 gene). Results show that concentration-dependent decreases in relative survival are similar in all three cell lines, indicating that XRCC1 is not crucial for protecting cells from sodium chromate-induced cytotoxicity. Similarly the frequency of damaged metaphase cells was not affected by XRCC1 deficiency. However, the total number of Cr(VI)-induced chromosome aberrations was exacerbated by XRCC1 deficiency and the spectrum of chromosome damage changed dramatically. Specifically, chromatid and isochromatid lesions were the most prominent aberrations induced in the cell lines and XRCC1 was essential to reduce the formation of chromatid lesions. In addition, XRCC1 deficiency caused a dramatic increase in the number of chromatid exchanges indicating that it is involved in protection from Cr(VI)-induced chromosome instability. 相似文献

Although parasite habitat preference is well studied, it is rarely rigorously evaluated statistically because of many zero intensities. Attachment-site preference and intensities of 2 macroectoparasite species ( Caligus elongatus and Calliobdella vivida ) of Atlantic sturgeon, Acipenser oxyrinchus Mitchill, in Minas Basin, Bay of Fundy, Canada, were characterized with the use of zero-inflated negative binomial statistical models that included a fork-length offset to control for body size. Three other parasites were encountered, sometimes in high numbers on various body sites, but too few counts overall prevented construction of meaningful statistical models. Of 26 sturgeons, prevalence of (1) C. elongatus (Copepoda) was 85%, mainly on caudal fins and nonfin body sites; (2) C. vivida (Hirudinea) was 81%, mainly on the pelvic and pectoral fins, and dorsal and ventral-lateral body sites; (3) Dichelesthium oblongum (Copepoda) was 31% within the gills or burrowed into the musculature at the base of fins; (4) Argulus stizostethii (Crustacea: Branchiura) was 8%; and (5) Nitzschia sturionis (Monogenea) was 12%. Only D. oblongum was associated with visible damage, mainly as lesions on gills and soft tissues. Forest management practices in the eastern United States directly impact large parcels of land that serve as habitat for timber rattlesnakes (Crotalus horridus). We assessed the behavioral response of timber rattlesnakes to commercial logging activities and the impact of such activities on a timber rattlesnake population in northcentral Pennsylvania. We radiotelemetrically monitored 67 individual snakes over periods of up to 4 years, marked and recaptured 306 snakes, and conducted search and survey efforts before, during, and after commercial logging operations on 3 timber sale parcels (totaling 154.2 ha). Location and timing of timber sales created the maximum opportunity for interaction of snakes with logging operations and with altered habitat. Observed logging-related mortality of snakes was low (<2% of the population/yr), but potential mortality could have reached 7%. Logging activity and resulting habitat changes did not alter behavior or movement patterns of telemetrically monitored snakes. Snakes with established activity ranges in timber sale areas continued to use these areas both during and after logging operations. Similarly, snakes with activity patterns that did not include timber sale areas did not alter their movement patterns to include such sites in the short-term. Timbering increased structural diversity of the habitat and, concurrently, diversity of habitat used by timber rattlesnakes increased. Our results suggest that the opportunity exists to develop forest management practices that provide timber products while limiting impacts on behavior and habitat use of timber rattlesnakes. To further reduce impacts to timber rattlesnake populations we recommend that management agencies require commercial logging contractors, sub-contractors, and field employees to adhere strictly to a policy that prohibits the intentional killing of rattlesnakes encountered during logging activities. © 2010 The Wildlife Society. 相似文献

Background
Tuberculosis is a major health problem in São Paulo, Brazil, which is the most populous and one of the most cosmopolitan cities in South America. To characterize the genetic diversity of Mycobacterium tuberculosis in the population of this city, the genotyping techniques of spoligotyping and MIRU were applied to 93 isolates collected in two consecutive years from 93 different tuberculosis patients residing in São Paulo city and attending the Clemente Ferreira Institute (the reference clinic for the treatment of tuberculosis).Findings
Spoligotyping generated 53 different spoligotype patterns. Fifty-one isolates (54.8%) were grouped into 13 spoligotyping clusters. Seventy- two strains (77.4%) showed spoligotypes described in the international databases (SpolDB4, SITVIT), and 21 (22.6%) showed unidentified patterns. The most frequent spoligotype families were Latin American Mediterranean (LAM) (26 isolates), followed by the T family (24 isolates) and Haarlem (H) (11 isolates), which together accounted for 65.4% of all the isolates. These three families represent the major genotypes found in Africa, Central America, South America and Europe. Six Spoligo-International-types (designated SITs by the database) comprised 51.8% (37/72) of all the identified spoligotypes (SIT53, SIT50, SIT42, SIT60, SIT17 and SIT1). Other SITs found in this study indicated the great genetic diversity of M. tuberculosis, reflecting the remarkable ethnic diversity of São Paulo city inhabitants. The MIRU technique was more discriminatory and did not identify any genetic clusters with 100% similarity among the 93 isolates. The allelic analysis showed that MIRU loci 26, 40, 23 and 10 were the most discriminatory. When MIRU and spoligotyping techniques were combined, all isolates grouped in the 13 spoligotyping clusters were separated.Conclusions
Our data indicated the genomic stability of over 50% of spoligotypes identified in São Paulo and the great genetic diversity of M. tuberculosis isolates in the remaining SITs, reflecting the large ethnic mix of the São Paulo city inhabitants. The results also indicated that in this city, M. tuberculosis isolates acquired drug resistance independently of genotype and that resistance was more dependent on the selective pressure of treatment failure and the environmental circumstances of patients.87.
