Electron microscopy (EM) in combination with image analysis is a powerful technique to study protein structure at low- and high resolution. Since electron micrographs of biological objects are very noisy, substantial improvement of image quality can be obtained by averaging individual projections. Crystallographic and noncrystallographic averaging methods are available and have been applied to study projections of the large protein complexes embedded in photosynthetic membranes from cyanobacteria and higher plants. Results of EM on monomeric and trimeric Photosystem I complexes, on monomeric and dimeric Photosystem II complexes, and on the monomeric cytochromeb6/f complex are discussed.  相似文献

BackgroundThe protoporphyrin IX-triplet state lifetime technique (PpIX-TSLT) is proposed as a potential clinical non-invasive tool to monitor mitochondrial function. This technique has been evaluated in several animal studies. Mitochondrial respirometry allows measurement in vivo of mitochondrial oxygen tension (mitoPO2) and mitochondrial oxygen consumption (mitoVO2) in skin. This study describes the first use of a clinical prototype in skin of humans.MethodsThe clinical prototype was tested in 30 healthy volunteers. A self-adhesive patch containing 2 mg 5-aminolevulinic acid (ALA) was applied on the skin of the anterior chest wall (sternal) for induction of mitochondrial protoporphyrin IX and was protected from light for 5 h. MitoPO2 was measured by means of oxygen-dependent delayed fluorescence of protoporphyrin IX. MitoVO2 was determined by dynamic mitoPO2 measurements on the primed skin, while locally blocking oxygen supply by applying local pressure with the measurement probe. MitoPO2 was recorded before and during a 60-s period of compression of the microcirculation, at an interval of 1 Hz. Oxygen consumption (i.e. the local oxygen disappearance rate) was calculated from the decay of the mitoPO2 slope.ResultsOxygen-dependent delayed fluorescence measurements were successfully performed in the skin of 27 volunteers. The average value (± SD) of mitoPO2 was 44 ± 17 mmHg and mean mitoVO2 values were 5.8 ± 2.3 and 6.1 ± 1.6 mmHg s-1 at a skin temperature of 34°C and 40°C, respectively. No major discomfort during measurement and no long-term dermatological abnormalities were reported in a survey performed 1 month after measurements.ConclusionThese results show that the clinical prototype allows measurement of mitochondrial oxygenation and oxygen consumption in humans. The development of this clinically applicable device offers opportunities for further evaluation of the technique in humans and the start of first clinical studies.  相似文献

Rapid, accurate, and automated measurement of soil matric potential is desirable. Evidence suggested that the Watermark resistance block might be an appropriate and inexpensive tool, so we conducted an evaluation of its relevant characteristics. A number of these blocks were calibrated under laboratory conditions to determine their individual and aggregate responses to soil matric potential, soil type, and temperature. We found that the temperature response could be expressed as a single equation, valid for all tested blocks, but comparison against matric potential revealed that each block had a characteristic response. Furthermore, block responses were different in two soils and, for a given soil, not necessarily reproducible. Given these limitations, these sensors are probably useful only as relative indicators of soil water status.  相似文献

Summary Mixing ability analyses, adapted from combining ability analyses used in plant breeding, were performed on yield and stripe rust (Puccinia striiformis) severity data for two-way mixtures among either four or five club wheat (Triticum aesitivum) cultivars grown in five environments. Initially, two statistics were calculated for each trait: general mixing ability (GMA), the average performance of a cultivar over all of the mixtures, and specific mixing ability (SMA), the deviation of a mixture from the estimated performance of the pair based on its average performance in mixtures. General mixing ability was further divided into two components: genotype performing ability (GPA), the innate ability of a cultivar to yield and resist disease in pure stand, and true general mixing ability (TGMA), the average ability of a cultivar to influence yield and disease when mixed with other cultivars. Significant mean squares for genotypes, GMA, SMA, and TGMA were found for all of the traits in most environments. Examination of TGMA and SMA revealed cultivars and cultivar combinations that were statistically better mixers than the others. Some of the significant effects were probably due to the use of cultivars that differed in height and stripe rust resistance, but for other combinations there was no apparent explanation for enhanced mixing ability.Paper No. 9132 of the Oregon Agricultural Experiment Station. Diatoms possess fucoxanthin chlorophyll proteins (FCP) as light-harvesting systems. These membrane intrinsic proteins bind fucoxanthin as major carotenoid and Chl c as accessory chlorophyll. The relatively high sequence homology to higher plant light-harvesting complex II gave rise to the assumption of a similar overall structure. From centric diatoms like Cyclotella meneghiniana, however, two major FCP complexes can be isolated. FCPa, composed of Fcp2 and Fcp6 subunits, was demonstrated to be trimeric, whereas FCPb, known to contain Fcp5 polypeptides, is of higher oligomeric state. No molecular structure of either complex is available so far. Here we used electron microscopy and single particle analysis to elucidate the overall architecture of FCPb. The complexes are built from trimers as basic unit, assembling into nonameric moieties. The trimer itself is smaller, i.e. more compact than LHCII, but the main structural features are conserved.  相似文献

In this study we investigated the spatial and temporal variation in soil solution chemistry and of water and ion fluxes through the soil in a forest ecosystem. Our aim was to evaluate the relevance of these variations for the accuracy of average areal soil solution concentrations and ion fluxes with seepage at 90 cm depth.Twenty spatially distinct subcompartments of approximately 1 m2 were established within a mature stand of Norway spruce and ceramic suction lysimeters were installed at depths of 20, 35 and 90 cm. A tensiometer was placed close to each suction lysimeter, and one throughfall sampler was established for each subcompartment.Soil solution samples were analysed for major ions (H+, Na+, K+, Ca2+, Mg2+, Mn2+, Fe3+, Al3+, Cl-, NO 3 - , SO 4 2- . We calculated water fluxes for each subcompartment separately by a numeric simulation of the soil water flux close to the lysimeters. The ion fluxes at each lysimeter were calculated by multiplying the simulated water fluxes with the ion concentrations on a fortnightly base. Averaging these 20 independent ion fluxes gave the areal average flux and an estimate of its statistical accuracy. The spatial variation of ion concentrations in the soil solution was high with coefficients of variance ranging from 5% to 128%. Part of the spatial variation was related to stem distance. Temporal variation of the concentrations was less than spatial for most ions. The spatial variation of water and ion fluxes with seepage was also substantial; for example the fluxes of SO 4 2- -S calculated for each subcompartment ranged from 21 to 119 kg ha-1 yr-1, with an arithmetic average of 47 kg ha-1 yr-1. For H2O, Mg2+, Cl-, and SO 4 2- , the spatial heterogeneity of seepage fluxes was largely explained by the heterogeneity of throughfall fluxes. No such relationship was found for nitrogen.Despite using 20 replicates, the 95% confidence intervals of the average annual areal fluxes with seepage were found to be 20–30% for most ions.  相似文献
