Recently, the widespread distribution of pesticides detected in the hive has raised serious concerns about pesticide exposure on honey bee (Apis mellifera L.) health. A larval rearing method was adapted to assess the chronic oral toxicity to honey bee larvae of the four most common pesticides detected in pollen and wax - fluvalinate, coumaphos, chlorothalonil, and chloropyrifos - tested alone and in all combinations. All pesticides at hive-residue levels triggered a significant increase in larval mortality compared to untreated larvae by over two fold, with a strong increase after 3 days of exposure. Among these four pesticides, honey bee larvae were most sensitive to chlorothalonil compared to adults. Synergistic toxicity was observed in the binary mixture of chlorothalonil with fluvalinate at the concentrations of 34 mg/L and 3 mg/L, respectively; whereas, when diluted by 10 fold, the interaction switched to antagonism. Chlorothalonil at 34 mg/L was also found to synergize the miticide coumaphos at 8 mg/L. The addition of coumaphos significantly reduced the toxicity of the fluvalinate and chlorothalonil mixture, the only significant non-additive effect in all tested ternary mixtures. We also tested the common ‘inert’ ingredient N-methyl-2-pyrrolidone at seven concentrations, and documented its high toxicity to larval bees. We have shown that chronic dietary exposure to a fungicide, pesticide mixtures, and a formulation solvent have the potential to impact honey bee populations, and warrants further investigation. We suggest that pesticide mixtures in pollen be evaluated by adding their toxicities together, until complete data on interactions can be accumulated.  相似文献

The introduction of plasmid DNA into germinating spores of an industrially improved strain of Saccharopolyspora erythraea was accomplished by electroporation. Various parameters affecting the efficiency of electroporation were examined. The most critical factor was the extent of spore germination. Electrocompetence was limited to a 4-h period following the initial emergence of the germ tube. Electroporation efficiencies as high as 2 × 105 CFU μg−1 of plasmid DNA were obtained using electrocompetent germlings. The optimal field strength was 12–14 kV cm−1 with a pulse duration of 15–20 ms. Electrocompetent germlings were stored at −80°C without a significant decrease in transformation efficiency. The utility of this protocol was demonstrated by isolating a propionyl-CoA carboxylase mutant through targeted gene disruption and replacement. Oxidation of the branched-chain amino acid leucine was studied in 22 male Sprague-Dawley rats (70-90 g) over 3 days following the ingestion on Day 1 of a mixed diet containing a tracer dose (10 muCi) of L-[1-14C]Leu. One group (E) completed 1 hr exercise at 80% VO2 max immediately after a 2-hr feeding period on all 3 days, while a second group served as a control. Rats from group E were sacrificed immediately after the 2 hr feeding on Day 1, following exercise on Days 1 and 3, and at the end of Day 3. The following were determined: (1) continuous 14CO2 production, (2) radioactivity remaining in the gastrointestinal tract, and (3) distribution of free vs protein bound 14C in muscle and liver. The results indicated that (1) 14CO2 production increased during exercise on all 3 days (P less than 0.01), (2) 14CO2 production also increased (P less than 0.05) following food intake (unlabeled diet), (3) 14CO2 production due to exercise was greater than that due to food intake (P less than 0.05), (4) absolute 14CO2 production decreased dramatically by 15 hr of Day 1 (P less than 0.01) with little change thereafter (except with exercise and food intake on Days 2 and 3), (5) greater than 98% of the labeled diet was absorbed from the GIT 51 hr postingestion, and (6) 14C in the free pool of muscle and liver could account for less than 15% of the total 14CO2 production. These results suggest that protein bound 14C in addition to free 14C may be responsible for a significant proportion of the observed increased 14CO2 production during exercise.  相似文献

Two new series of potent and selective dual EGFR/ErbB-2 kinase inhibitors derived from novel thienopyrimidine cores have been identified. Isomeric thienopyrimidine cores were evaluated as isosteres for a 4-anilinoquinazoline core and several analogs containing the thieno[3,2-d]pyrimidine core showed anti-proliferative activity with IC50 values less than 1 μM against human tumor cells in vitro.  相似文献

The Gulf of Mexico is a subtropical marginal sea of the western North Atlantic Ocean with a diverse cetacean community. Ship-based, line-transect abundance surveys were conducted in oceanic waters (>200 m deep) of the northern Gulf within U. S. waters (380,432 km2) during spring from 1996 to 1997 and from 1999 to 2001. Data from these five surveys were pooled and minimum abundance estimates were based on 12,162 km of effort and 512 sightings of at least 19 species. The most commonly sighted species (number of groups) were pantropical spotted dolphin, Stenella attenuata (164); sperm whale, Physeter macrocephalus (67); dwarf/pygmy sperm whale, Kogia simalbreviceps (58); Risso's dolphin, Grampus griseus (38); and bottlenose dolphin, Tursiops truncatus (24). The most abundant species (number of individuals; coefficient of variation) were S. attenuata (91,321; 0.16); Clymene dolphin, S. clymene (17,355; 0.65); spinner dolphin, S. longirostris (11,971; 0.71); and striped dolphin, S. coeruleoalba (6,505; 0.43). The only large whales sighted were P. macrocephalus (1,349; 0.23) and Bryde's whale, Balaenopteraedeni (40; 0.61). Abundances for other species or genera ranged from 95 to 2,388 animals. Cetaceans were sighted throughout the oceanic northern Gulf and, whereas many species were widely distributed, some had more regional distributions.  相似文献
