Rat testicular microsomal membrane fraction contains both Mg+2-dependent and Mg+2-independent Ca+2-ATPase activity. The latter activity is about two times higher than the former. Calcium ion required for maximum activation of Mg+2-independent Ca+2-ATPase in 3.0 mM, whereas for the dependent one it is 2.5 mM. Both the enzymes are resistant to cold shock upto seven days. Histidine and imidazole buffers are found to be the most suitable for dependent and independent enzyme activities, respectively. The pH optima for dependent one is 7.5, whereas for the independent one it is 8.5. Temperature optima for the former is 37°C and for latter one it is 40°C. Among all the nuclestides tested, ATP is found to be the best substrate for both the enzymes. The optimum concentration of ATP for dependent and independent enzyme activities are 3.0 mM and 1.5 mM respectively. Divalent metal ions like Zn+2, Ba+2 and Mn+2 have been found to inhibit Mg+2-dependent Ca+2-ATPase activity whereas Mg+2-independent Ca+2-ATPase activity is inhibited by the divalent ions except zinc which is found to stimulate the enzyme activity. Both the enzymes are inhibited by vanadata, EDTA and EGTA. I50, for vanadate is 0.05 and 0.125 mM for dependent and independent activities, respectively. Sulfhydral groups modifying agents e.g., NEM, DTNB and chlorpromazine are found to affect the enzyme activities in different ways. Thus NEM and chlorpromazine are found to inhibit and DTNB stimulate the enzyme activities in both the cases.  相似文献

Host macrophage infection by the parasite Leishmania donovani is heterogeneous, but it is not clear which factors are responsible for parasite recognition within the macrophages. One possible factor may be the alteration of the microviscosity of the macrophage membrane. This in turn may affect receptor expression and hence parasite infection. In this paper we describe alteration of the lipid composition and hence the microviscosity of the macrophage membrane in a controlled manner using liposome fusion technique. At a higher macrophage membrane microviscosity a larger number of parasites have been found to adhere to the macrophage surface. However, the proportion of parasites finally internalized when compared to parasites adhering to macrophages is inversely correlated with the artificially altered macrophage membrane microviscosity. The process of endocytosis has been examined in both native and lipid modified macrophages in the presence of several sugar antagonists. The results indicate (i) glucose and mannose are specifically involved in the binding process, and (ii) the microviscosity has a key role in controlling the macrophage parasite interaction. The results obtained so far support a model of endocytosis where expression of the receptor is a critical initial process dependent on the microviscosity of the membrane.  相似文献

The peroxidase activity of mouse submaxillary gland was found to be elevated by about 128% at 22 hr. after the administration of reserpine (0.5 mg/kg). This effect of reserpine was observed in rat also. Neither pretreatment with 6-hydroxy dopamine (6-OH dopamine) nor surgical sympathetic denervation could abolish the increase of the peroxidase activity elicited by reserpine. Also, treatment with propranolol, dibenamine or atropine sulfate failed to reverse the effect of reserpine. These results suggest that neither catecholamine nor acetyl choline is involved in this reserpine action.  相似文献

Aggregation and fusion of unilamellar vesicles by poly(ethylene glycol)   总被引:5,自引:0,他引:5  
Various aspects of the interaction between the fusogen, poly(ethylene glycol) and phospholipids were examined. The aggregation and fusion of small unilamellar vesicles of egg phosphatidylcholine (PC), bovine brain phosphatidylserine (PS) and dimyristoylphosphatidylcholine (DMPC) were studied by dynamic light scattering, electron microscopy and NMR. The fusion efficiency of Dextran, glycerol, sucrose and poly(ethylene glycol) of different molecular weights were compared. Lower molecular weight poly(ethylene glycol) are less efficient with respect to both aggregation and fusion. The purity of poly(ethylene glycol) does not affect its fusion efficiency. Dehydrating agents, such as Dextran, glycerol and sucrose, do not induce fusion. 31P-NMR results revealed a restriction in the phospholipid motion by poly(ethylene glycol) greater than that by glycerol and Dextran of similar viscosity and dehydrating capacity. This may be associated with the binding of poly(ethylene glycol) to egg PC, with a binding capacity of 1 mol of poly(ethylene glycol) to 12 mol of lipid. Fusion is greatly enhanced below the phase transition for DMPC, with extensive fusion occurring below 6% poly(ethylene glycol). Fusion of PS small unilamellar vesicles depends critically on the presence of cations. Large unilamellar vesicles were found to fuse less readily than small unilamellar vesicles. The results suggest that defects in the bilayer plays an important role in membrane fusion, and the 'rigidization' of the phospholipid molecules facilitates fusion possibly through the creation of defects along domain boundaries. Vesicle aggregation caused by dehydration and surface charge neutralization is a necessary but not a sufficient condition for fusion.  相似文献

Phytoremediation, popularly known as ‘green technology’ has been employed in the present investigation to examine the potential of fluoride removal from water by some aquatic plants. Fluoride contamination in drinking water is very much prevalent in different parts of the world including India. Batch studies were conducted using some aquatic plants e.g., Pistia stratiotes, Eichhornia crassipes, and Spirodela polyrhiza which profusely grow in natural water bodies. The experimental data exhibited that all the above three aquatic floating macrophytes could remove fluoride to some relative degree of efficiency corresponding to initial concentration of fluoride 3, 5, 10, 20 mg/l after 10 days exposure time. Result showed that at lower concentration level i.e., 3 mg/L removal efficiency of Pistia stratiotes (19.87%) and Spirodela polyrhiza (19.23%) was found to be better as compared to Eichhornia crassipes (12.71%). Some of the physiological stress induced parameters such as chlorophyll a, chlorophyll b, total chlorophyll, carotenoid, total protein, catalase, and peroxidase were also studied to explore relative damage within the cell. A marginal stress was imparted among all the plants for lower concentration values (3 mg/L), whereas at 20 mg/l, maximum damage was observed.  相似文献

While the extremes in organic photovoltaic bulk heterojunction morphology (finely mixed or large pure domains) are easily understood and known to be unfavorable, efficient devices often exhibit a complex multi‐length scale, multi‐phase morphology. The impact of such multiple length scales and their respective purities and volume fractions on device performance remains unclear. Here, the average spatial composition variations, i.e., volume‐average purities, are quantified at multiple size scales to elucidate their effect on charge creation and recombination in a complex, multi‐length scale polymer:fullerene system (PBDTTPD:PC71BM). The apparent domain size as observed in TEM is not a causative parameter. Instead, a linear relationship is found between average purity at length scales <50 nm and device fill‐factor. Our findings show that a high volume fraction of pure phases at the smallest length scales is required in multi‐length scale systems to aid charge creation and diminish recombination in polymer:fullerene solar cells.  相似文献

  • Endophytic microbes isolated from plants growing in nutrient‐deficient environments often possess properties that improve nutrition of agriculturally important plants. A consortium of non‐rhizobial endophytic microbes isolated from a macrophyte Typha angustifolia growing in the marginal wetlands associated with a Uranium mine was characterized for their beneficial effect on rice and the mechanisms of growth promotion were investigated.
  • The microbes were identified and characterized for their potential plant growth promoting (PGP) properties. Effect of these microbes on nitrogen (N)‐metabolism of rice was tested as Typha endophytes were predominantly (N)‐fixing. Relative N‐use efficiency and expression of genes involved in N‐uptake and assimilation were investigated in treated plants.
  • Evidence of horizontal gene transfer (HGT) of dinitrogen reductase gene was observed within the consortium from a Pseudomonas stutzeri strain. The consortium behaved as plant probiotic and showed substantial growth benefits to Typha, their natural host as well as to rice. Typha endophytes colonized rice endosphere significantly increasing biomass, shoot length and chlorophyll content in rice plants both under N‐sufficient and N‐deficient conditions. N‐uptake and assimilation genes were upregulated in plants treated with the endophytes even after three weeks post infection.
  • Our results suggested, HGT of nitrogen‐fixation trait to be highly prevalent among endophytes isolated from nutrient‐poor habitats of the uranium mine. A long‐term nitrogen deficiency response in the treated plants was elicited by the consortium improving N‐uptake, assimilation and relative N‐use efficiency of rice plants. This appeared to be at least one of the main strategies of plant growth promotion.

Delivering diclofenac diethylamine transdermally by means of a hydrogel is an approach to reduce or avoid systemic toxicity of the drug while providing local action for a prolonged period. In the present investigation, a process was developed to produce nanosize particles (about 10 nm) of diclofenac diethylamine in situ during the development of hydrogel, using simple mixing technique. Hydrogel was developed with polyvinyl alcohol (PVA) (5.8% w/w) and carbopol 71G (1.5% w/w). The formulations were evaluated on the basis of field emission scanning electron microscopy, texture analysis, and the assessment of various physiochemical properties. Viscosity (163–165 cps for hydrogel containing microsize drug particles and 171–173 cps for hydrogel containing nanosize drug particles, respectively) and swelling index (varied between 0.62 and 0.68) data favor the hydrogels for satisfactory topical applications. The measured hardness of the different hydrogels was uniform indicating a uniform spreadability. Data of in vitro skin (cadaver) permeation for 10 h showed that the enhancement ratios of the flux of the formulation containing nanosize drug (without the permeation enhancer) were 9.72 and 1.30 compared to the formulation containing microsized drug and the marketed formulations, respectively. In vivo plasma level of the drug increased predominantly for the hydrogel containing nanosize drug-clusters. The study depicts a simple technique for preparing hydrogel containing nanosize diclofenac diethylamine particles in situ, which can be commercially viable. The study also shows the advantage of the experimental transdermal hydrogel with nanosize drug particles over the hydrogel with microsize drug particles.  相似文献

Desulfurization of dibenzothiophene (DBT) and alkylated DBT derivatives present in transport fuel through specific cleavage of carbon-sulfur (C-S) bonds by a newly isolated bacterium Chelatococcus sp. is reported for the first time. Gas chromatography-mass spectrometry (GC-MS) analysis of the products of DBT degradation by Chelatococcus sp. showed the transient formation of 2-hydroxybiphenyl (2-HBP) which was subsequently converted to 2-methoxybiphenyl (2-MBP) by methylation at the hydroxyl group of 2-HBP. The relative ratio of 2-HBP and 2-MBP formed after 96 h of bacterial growth was determined at 4:1 suggesting partial conversion of 2-HBP or rapid degradation of 2-MBP. Nevertheless, the enzyme involved in this conversion process remains to be identified. This production of 2-MBP rather than 2-HBP from DBT desulfurization has a significant metabolic advantage for enhancing the growth and sulfur utilization from DBT by Chelatococcus sp. and it also reduces the environmental pollution by 2-HBP. Furthermore, desulfurization of DBT derivatives such as 4-M-DBT and 4, 6-DM-DBT by Chelatococcus sp. resulted in formation of 2-hydroxy-3-methyl-biphenyl and 2-hydroxy –3, 3/- dimethyl-biphenyl, respectively as end product. The GC and X-ray fluorescence studies revealed that Chelatococcus sp. after 24 h of treatment at 37°C reduced the total sulfur content of diesel fuel by 12% by per gram resting cells, without compromising the quality of fuel. The LC-MS/MS analysis of tryptic digested intracellular proteins of Chelatococcus sp. when grown in DBT demonstrated the biosynthesis of 4S pathway desulfurizing enzymes viz. monoxygenases (DszC, DszA), desulfinase (DszB), and an NADH-dependent flavin reductase (DszD). Besides, several other intracellular proteins of Chelatococcus sp. having diverse biological functions were also identified by LC-MS/MS analysis. Many of these enzymes are directly involved with desulfurization process whereas the other enzymes/proteins support growth of bacteria at an expense of DBT. These combined results suggest that Chelatococcus sp. prefers sulfur-specific extended 4S pathway for deep-desulphurization which may have an advantage for its intended future application as a promising biodesulfurizing agent.  相似文献
