Expanded polystyrene (EPS) beads have been shown to be an effective tool for controlling immature stages of mosquitoes, as well as preventing oviposition by adults. Polystyrene does not biodegrade quickly, resulting in some concerns about its effect on the environment. A potential solution is the use of biodegradable materials that cover the surface of mosquito breeding sites in the same way as EPS beads. Two candidates are polylactic acid (PLA) beads and corn starch shreds. Larval mortality and adult emergence of Culex quinquefasciatus Say (Diptera: Culicidae) were monitored in bowls with each of four treatments: EPS beads, PLA beads, corn starch shreds and a control. The PLA beads were as effective as EPS beads at preventing mosquito emergence, whereas the shredded corn starch treatment resulted in significantly higher rates of emergence to the control. Similarly, EPS and PLA beads resulted in 100% mortality after 10 days, while there was low mortality of larvae in the corn starch (9%) and control treatments (20%). PLA beads provided similar levels of mortality and reduction in adult emergence as EPS beads. However, the production requirements of PLA beads may limit its use in field conditions.  相似文献

The metabolic composition of fecal extracts provides a window for elucidating the complex metabolic interplay between mammals and their intestinal ecosystems, and these metabolite profiles can yield information on a range of gut diseases. Here, the metabolites present in aqueous fecal extracts of humans, mice and rats were characterized using high-resolution (1)H NMR spectroscopy coupled with multivariate pattern recognition techniques. Additionally, the effects of sample storage and preparation methods were evaluated in order to assess the stability of fecal metabolite profiles, and to optimize information recovery from fecal samples. Finally, variations in metabolite profiles were investigated in healthy mice as a function of time. Interspecies variation was found to be greater than the variation due to either time or sample preparation. Although many fecal metabolites were common to the three species, such as short chain fatty acids and branched chain amino acids, each species generated a unique profile. Relatively higher levels of uracil, hypoxanthine, phenylacetic acid, glucose, glycine, and tyrosine amino acids were present in the rat, with beta-alanine being unique to the rat, and glycerol and malonate being unique to the human. Human fecal extracts showed a greater interindividual variation than the two rodent species, reflecting the natural genetic and environmental diversity in human populations. Fecal composition in healthy mice was found to change over time, which might be explained by altered gut microbial presence or activity. The systematic characterization of fecal composition across humans, mice, and rats, together with the evaluation of inherent variation, provides a benchmark for future studies seeking to determine fecal biomarkers of disease and/or response to dietary or therapeutic interventions.  相似文献

The jointless mutation of tomato results in the formation of flower pedicels that lack an abscission zone and inflorescence meristems that revert to vegetative growth. We have analyzed periclinal chimeras and mericlinal sectors of jointless and wild-type tissue to determine how cells in different meristem layers (L1, L2, and L3) and their derivatives interact during these two developmental processes. Cells in the inner meristem layer, L3, alone determined whether the meristem maintained the inflorescence state or reverted to vegetative growth. Moreover, L3 derivatives determined whether a functional pedicel abscission zone formed. Limited and disorganized autonomous development of wild-type L2-derived cells occurred when they overlay mutant tissue. Adjacent mutant and wild-type L3-derived tissues in pedicels developed autonomously, indicating little or no lateral communication. Only the outermost L3-derived cells within the pedicel were capable of orchestrating normal pedicel development in overlying tissues, revealing the special status of those cells as coordinators of development for L1- and L2-derived cells, whereas the innermost L3-derived cells developed autonomously but did not influence the development of other cells.  相似文献

Natural products constitute and significantly impact many current anti-cancer medical interventions. A subset of natural products induces injury processes in malignant cells that recruit and activate host immune cells to produce an adaptive anti-cancer immune response, a process known as immunogenic cell death. However, a challenge in the field is to delineate forms of cell death and injury that best promote durable antitumor immunity. Addressing this with a single-cell chemical biology natural product discovery platform, like multiplex activity metabolomics, would be especially valuable in human leukemia, where cancer cells are heterogeneous and may react differently to the same compounds. Herein, a new ten-color, fluorescent cell barcoding-compatible module measuring six immunogenic cell injury signaling readouts are as follows: DNA damage response (γH2AX), apoptosis (cCAS3), necroptosis (p-MLKL), mitosis (p-Histone H3), autophagy (LC3), and the unfolded protein response (p-EIF2α). A proof-of-concept screen was performed to validate functional changes in single cells induced by secondary metabolites with known mechanisms within bacterial extracts. This assay was then applied in multiplexed activity metabolomics to reveal an unexpected mammalian cell injury profile induced by the natural product narbomycin. Finally, the functional consequences of injury pathways on immunogenicity were compared with three canonical assays for immunogenic hallmarks, ATP, HMGB1, and calreticulin, to correlate secondary metabolite-induced cell injury profiles with canonical markers of immunogenic cell death. In total, this work demonstrated a new phenotypic screen for discovery of natural products that modulate injury response pathways that can contribute to cancer immunogenicity.  相似文献

Lingual surface attrition of the maxillary anterior teeth (LSA-MAT) has been found in additional prehistoric Latin American Indian skeletons. LSAMAT was first observed in crania from an Archaic Brazilian site. This second finding occurs in teeth from Venado Beach, a late prehistoric site in Panama. LSAMAT is also present in some fragmentary specimens from the Archaic Cerro Mangote site in Panama. LSAMAT at Venado Beach is present in 57% of 28 adult crania. As in the Brazilian study, LSAMAT is associated with a high caries rate (82% of 50 adults; 11.7% of 852 permanent teeth). As first suggested, eating and processing of some type of abrasive carbohydrate food, such as manioc, is the possible cause of LSAMAT. However, other possible causes relating to habitation on or near marine coasts cannot be totally ruled out.  相似文献

Hartley guinea pigs were sensitized to 2,4-dinitrofluorobenzene, Mycobacterium tuberculosis H37RA, and allogenic skin. During the development of these sensitivities, serum alpha globulin changes were detected by polyacrilamide gel disc electrophoresis. Electrophoretic region 4 (alpha globulins) increased, then subsided, concomitant with the development of cellular immunity. Skin testing did not seem to influence alpha globulin levels in sensitized animals. However, high alpha globulin levels decreased skin test responsiveness. It is suggested that the increased alpha globulin noted here is immunoregulative alpha globulin (IRA).  相似文献
