Summary This paper describes a practical new use of 3-mercaptopropionic acid as a highly versatile multidetachable linker for solid-phase synthesis. Our approach is based on the stability of the alkylthioester functionality to optimized Boc-SPPS protocols and HF treatment, as well as on the mild activation of the thioester functionality toward nucleophilic or reductive displacement. This allows several C-terminal modifications to be introduced into a synthetic molecule during the cleavage step. We have shown that unprotected peptides can be efficiently cleaved from a propyl thioester-polyethylene glycol-poly-(N,N-dimethylacrylamide) copolymer resin using a great variety of nucleophiles to give the corresponding C-terminally modified peptides (esters, thioesters, carboxylic acids, thioacids, amides, hydroxamic acids, hydrazides, alcohols). The nucleophilic cleavage reaction is both rapid and exceptionally clean in all the cases tested. The indole-glucosinolate, glucobrassicin, which is found in large amounts in members of the genus Brassica, was subjected to tests in the standard Avena curvature bioassay. Both the tetramethylammonium salt of glucobrassicin and the non-crystallizted material induced curvature at a concentration of 5 × l0?4M. The curvature from the salt was, however, only half of that induced by the acid form. Bioassays of chromatograms of fresh methanolic extracts of savoy cabbage indicated the presence of indole-3-acetic acid as well as indole-3-acetonitrile in small amounts along with large amounts of glucobrassicin. Segments of green hypocotyl tissue of savoy cabbage respond with limited growth to high concentrations of glucobrassicin. Segments of etiolated hypocotyls or green epicotyls give no growth response to glucobrassicin. Pea stem segments which do not respond to indoleacetonitrile responded with similar growth to glucobrassicin and indoleacetic acid. Nitrilase activity was found in savoy hypocotyl and epicotyl tissue by chromatography of incubation media. The growth regulation in savoy cabbage in conjunction with data on the distribution of glucobrassicin in the plant, leads to the conclusion that glucobrassicin is a special, inactive storage and transport form of the active auxin in cabbage.  相似文献

We compared predictive success in two common algorithms for modeling species' ecological niches, GARP and Maxent, in a situation that challenged the algorithms to be general – that is, to be able to predict the species' distributions in broad unsampled regions, here termed transferability. The results were strikingly different between the two algorithms – Maxent models reconstructed the overall distributions of the species at low thresholds, but higher predictive levels of Maxent predictions reflected overfitting to the input data; GARP models, on the other hand, succeeded in anticipating most of the species' distributional potential, at the cost of increased (apparent, at least) commission error. Receiver operating characteristic (ROC) tests were weak in discerning models able to predict into broad unsampled areas from those that were not. Such transferability is clearly a novel challenge for modeling algorithms, and requires different qualities than does predicting within densely sampled landscapes – in this case, Maxent was transferable only at very low thresholds, and biases and gaps in input data may frequently affect results based on higher Maxent thresholds, requiring careful interpretation of model results.  相似文献

We report a quantitative method by which a single microculture can be examined for (i) cell morphology; (ii) cell number; (iii) DNA synthesis; and (iv) expression of cell antigens. This method first involves measuring by enzyme-linked immunosorbent assay (ELISA) the total bromodeoxyuridine (BrdU) incorporation into DNA by monolayer microcultures. The BrdU-ELISA measurement was followed by simultaneous immunostaining for BrdU-positive nuclei and for a cytoplasmic antigen. The method was applied to the measurement of mitogen-induced proliferation of rat sciatic nerve Schwann cell and cerebral astroglia microcultures. The ELISA measurement of BrdU incorporation compares favorably with measurements of tritiated thymidine incorporation and offers the additional advantages that the same microculture can subsequently be examined for cell number, for cell morphology, and for the percentage of cells having BrdU-labeled nuclei and other antigens.  相似文献

Physiological responses to dehydration in amphibians are reasonably well documented, although little work has addressed this problem in hibernating animals. We investigated osmotic and metabolic responses to experimental manipulation of hydration state in the wood frog (Rana sylvatica), a terrestrial hibernator that encounters low environmental water potential during autumn and winter. In winter-conditioned frogs, plasma osmolality varied inversely with body water content (range 69–79%, fresh mass) primarily due to increases in sodium and chloride concentrations, as well as accumulation of glucose and urea. Decreased hydration was accompanied by a marked reduction in the resting rate of oxygen consumption, which was inversely correlated with plasma osmolality and urea concentration. In a separate experiment, resting rates of oxygen consumption in fully hydrated frogs receiving injections of saline or saline containing urea did not differ initially; however, upon dehydration, metabolic rates decreased sooner in the urea-loaded frogs than in control frogs. Our findings suggest an important role for urea, acting in concert with dehydration, in the metabolic regulation and energy conservation of hibernating R. sylvatica.  相似文献

In six normal supine subjects epinephrine infusion produced a greater leukocytosis with smaller changes in heart rate and blood pressure than did norepinephrine or isoproterenol. Upright exercise in those subjects produced a greater leukocytosis than supine exercise at the same work load. To determine the lung's participation in these events, indium-labeled neutrophils (PMN) were given to four of the subjects. We found that 20-25% were retained in the first pass through the lung when compared with technetium-labeled erythrocytes. The number of labeled PMN in the lung gradually decreased and the number in the spleen and the liver increased. Exercise and catecholamine infusion caused an acceleration in the release of labeled cells from the lung, an increase in both labeled and unlabeled cells in the peripheral blood, and an increase in the number of labeled cells in the liver and spleen. This suggests that increased perfusion of low-flow areas in the lung may contribute to the increased leukocytosis seen in association with both exercise and catecholamine infusion.  相似文献

Chemistry-driven strategies for modifying, controlling and monitoring protein function in vitro and in vivo have attracted widespread interest among chemists in recent years. Several strategies have now emerged that complement standard genetics-based approaches, and they are being increasingly adopted by biologists to address issues in relevant contexts from cells to animals. With the development of these chemical biology tools, we might be approaching a time when detailed quantitative analysis of protein function, to a degree previously available only in reconstituted systems, is attainable in an in vivo setting.  相似文献

We recently reported that daily dietary supplementation with 100?μg selenium (a dose exceeding a rat's nutritional requirement by about 33-fold) initiated immediately after total-body irradiation (TBI) and maintained for 21?weeks mitigates radiation nephropathy in a rat model as indicated by blood urea nitrogen (BUN) levels and histopathological criteria (Radiat Res. 2009; 17:368-73). In this follow-up study, we explored the risks and benefits of delaying the onset of supplementation, shortening periods of supplementation, and escalating selenium supplementation beyond 100?μg/day. Supplementation with 200?μg selenium/day (as selenite or seleno-l-methionine) substantially improved the mitigation of radiation nephropathy by lowering BUN levels at 4?months after TBI from 115 to as low as 34?mg/dl and by proportionally lowering the incidence of histopathological abnormalities. Shortening the period of supplementation to 3 or 2?months did not compromise efficacy. Delaying the onset of supplementation for 1?week reduced but did not abrogate the mitigation of radiation nephropathy. Supplementation with 300?μg/day mitigated radiation nephropathy less effectively than 200?μg and was poorly tolerated. Rats that had been given 10?Gy of TBI were less tolerant of high-dose selenium than nonirradiated rats. This reduced tolerance of high-dose selenium would need to be taken into consideration when selenium is used for the mitigation of radiation injury in victims of nuclear accidents or acts of radiological terrorism. The high dose requirements, the pronounced threshold effect, and the superior performance of selenite suggest that the mitigation of radiation nephropathy involves mechanisms that go beyond the induction of selenoproteins.  相似文献
