Engineering microalgae has opened a new era for plant biologists and biotechnologists. Microalgae had been proved as a promising candidate for the production of biopharmaceuticals, nutraceuticals, antioxidants, antimicrobial and antiviral compounds, in dyeing and food industry as well for biofuel production. Genetic transformation of some important microalgae has been successful, but several other potential microalgae species still need scientific attention. The success of the genetic transformation depends mainly on the utilization of the selectable and screenable markers. Like for other higher crop plants, several useful markers have been reported for microalgae transformation. In this follow-up, we compared different marker genes for genetic engineering of approximately all the industrially important microalgae. We have discussed the expression host, the targeted genome, appropriate selection agent, as well as the transformation method. Genetic transformation is an expensive and labor intensive process and this review will aid to shorten the time span by providing a database of appropriate markers for microalgae research which could serve as a guide for those involved in the genetic engineering of microalgae.  相似文献

Apoptosis is a programed cell death that is vital for tissue homeostasis. However, embryo apoptosis had been known to be related to embryo fragmentation which should be avoided in in vitro fertilization (IVF). The purpose of this study was to evaluate the relationship of embryo apoptosis with the grade of immature oocytes and cleavage stage of in vitro produced (IVP) cattle embryos. This study consisted of 345 oocytes collected through ovary slicing. Immature oocytes were graded as A, B and C. This grading was based on cumulus cell thickness and compactness. All oocytes then underwent an in vitro maturation (IVM) procedure. An IVF was done 24 h after IVM culture. Prior to staining, stage of cleaved embryos was determined and classified as either 2, 4, 8 or >8-cell embryo stage. Apoptosis status of cleaved IVP embryos was determined by using annexin V-FITC staining technique at 48 and 72 h post insemination (hpi). Apoptosis status for each embryo was classified as either early or late. The result showed that there was no significant difference (p > 0.05) of apoptosis status among grade A, B and C embryos. All grades of oocytes showed embryo apoptosis where 1.5% late apoptosis for grade A, 4.5% and 10.4% of early and late apoptosis for grade B and grade C. Early apoptosis was not seen in grade A embryo. We also noted no significant difference (p > 0.05) of apoptosis status between 2, 4, 8 and >8-cell embryo stage. Early apoptosis was also not seen in >8-cell stage. Even though there were no differences in apoptosis expression between the three classes, the cleavage rate of grade A oocytes was significantly higher (p < 0.01) than grade B and grade C. Two glutathione peroxidase isoenzymes were purified from 24-day old embryos of the camel tick Hyalomma dromedarii and designated tick embryo glutathione peroxidase 1 and 2 (TEGPx1 and TEGPx2). The purification procedure involved ammonium sulfate precipitation, as well as ion exchange and gel filtration column chromatography. Glutathione peroxidase isoenzymes subunit molecular mass was determined by SDS-PAGE to be 36 ± 2 kDa and 59 ± 1.5 kDa for TEGPx1 and TEGPx2, respectively. TEGPx1 isoenzyme exhibited a dimeric structure with native molecular mass of 72 kDa while TEGPx2 was a monomeric protein. TEGPx1 and TEGPx2 displayed their pH optima at 7.6 and 8.2. Both isoenzymes cleaved preferentially H2O2 with K m values of 24 and 49 μM. Iodoacetamide competitively inhibited TEGPx1 with K i value of 0.45 mM and 1.10; phenanthroline competitively inhibited TEGPx2 with K i value of 0.12 mM. These results indicate the presence of two different forms of glutathione peroxidase in the developing camel tick embryos. This finding enhances our knowledge and understanding of the physiology of these ectoparasites and will encourage the development of new and untraditional control methods.  相似文献

Levels of Mo, As, Se, Fe, Cu, Zn, Ni, and Pb were determined in a vegetable commonly consumed in Pakistan. Samples were collected from three different sites (Ratokala, Phularwan, and Mailowal) supplied with three different water sources. Mo and Pb in water and Mo and As in C. sativum were higher than the suggested standards at the international level. Lower bioconcentration factor and pollution load index were seen at Site-I than at other sites. A positive association was found between the rhizosphere metals and those of the vegetable at all three sites. Enrichment factor at three sites ranged from 0.28 to 10.39. The highest value of daily intake of metals was found for Fe (0.245 mg/kg/day) and of the health risk index for As (70.41) at the wastewater inundated site. It is inferred that uptake of Mo, As, Cu, Ni, and Pb through C. sativum represents a high health danger to the individuals using this vegetable.  相似文献

Accumulation of different metals and metalloids was assessed in two vegetables radish (Raphanus sativus L.) and spinach (Spinacea oleracea L.) irrigated with domestic wastewater in the peri-urban areas of Khushab City, Pakistan. In general, the metal and metalloid concentrations in radish and spinach were higher at site-II treated with sewage water than those found at site-I treated with canal water. In case of radish at both sites the levels of metals (Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Se, Mo, Cd, and Pb) were below the permissible level except those of Mn, Ni, Mo, Cd, and Pb. At both sites, the transfer factor ranged from 0.047–228.3 mg kg?1 with Cr having the highest transfer factor. The metal pollution index in soil was in the following order: As > Fe > Ni > Zn > Cd > Mo > Se > Co > Pb > Mn > Cr > Cu, respectively. While in case of spinach at both sites, the concentrations of metals and metalloids in vegetable samples irrigated with canal and sewage water were observed below the permissible level except Mn, Ni, Zn, Mo, and Pb. At both sites, the transfer factor ranged from 0.038–245.4 mg kg?1 with Cr having the highest transfer factor. The metal pollution index in soil was in the following order: Cd > Ni > Co > Se > Mn > Zn > Mo > Pb > Fe > Cr > As > Cu, respectively.  相似文献

Recent climate change has resulted in changes to the phenology and distribution of invertebrates worldwide. Where invertebrates are associated with disease, climate variability and changes in climate may also affect the spatio‐temporal dynamics of disease. Due to its significant impact on sheep production and welfare, the recent increase in diagnoses of ovine haemonchosis caused by the nematode Haemonchus contortus in some temperate regions is particularly concerning. This study is the first to evaluate the impact of climate change on H. contortus at a continental scale. A model of the basic reproductive quotient of macroparasites, Q0, adapted to H. contortus and extended to incorporate environmental stochasticity and parasite behaviour, was used to simulate Pan‐European spatio‐temporal changes in H. contortus infection pressure under scenarios of climate change. Baseline Q0 simulations, using historic climate observations, reflected the current distribution of H. contortus in Europe. In northern Europe, the distribution of H. contortus is currently limited by temperatures falling below the development threshold during the winter months and within‐host arrested development is necessary for population persistence over winter. In southern Europe, H. contortus infection pressure is limited during the summer months by increased temperature and decreased moisture. Compared with this baseline, Q0 simulations driven by a climate model ensemble predicted an increase in H. contortus infection pressure by the 2080s. In northern Europe, a temporal range expansion was predicted as the mean period of transmission increased by 2–3 months. A bimodal seasonal pattern of infection pressure, similar to that currently observed in southern Europe, emerges in northern Europe due to increasing summer temperatures and decreasing moisture. The predicted patterns of change could alter the epidemiology of H. contortus in Europe, affect the future sustainability of contemporary control strategies, and potentially drive local adaptation to climate change in parasite populations.  相似文献
