Electron micrographs are presented of synaptic regions encountered in sections of frog sympathetic ganglia and earthworm nerve cord neuropile. Pre- and postsynaptic neuronal elements each appear to have a membrane 70 to 100 A thick, separated from each other over the synaptic area by an intermembranal space 100 to 150 A across. A granular or vesicular component, here designated the synaptic vesicles, is encountered on the presynaptic side of the synapse and consists of numerous oval or spherical bodies 200 to 500 A in diameter, with dense circumferences and lighter centers. Synaptic vesicles are encountered in close relationship to the synaptic membrane. In the earthworm neuropile elongated vesicles are found extending through perforations or gaps in the presynaptic membrane, with portions of vesicles appearing in the intermembranal space. Mitochondria are encountered in the vicinity of the synapse, and in the frog, a submicroscopic filamentary component can be seen in the presynaptic member extending up to the region where the vesicles are found, but terminating short of the synapse itself.  相似文献

Whereas polyploidy has been found in more than half the Gramineae,the occurrence of 2n gametes, a potentially major mechanismof polyploid production, has been rarely studied. In the presentwork, the frequency of 2n egg production in nine diploid (n= 2x = 14) subspecies of the polyploid complex Dactylis glomeratahas been estimated by determining the ploidy level of the progenyin 98 2X-4X crosses. The 2n egg origin of the 4X offspring wasverified using enzyme markers. The frequency of 2n egg productionin each subspecies was estimated from the number of 4X offspring,the number of florets in the seed plant and the fertility ofthe subspecies. The ploidy level of the progeny was determinedby chromosome counts or nuclear DNA quantification using flowcytometry. Plants producing 2n eggs were detected in 47% ofthe fertile crosses, involving eight subspecies. Variabilitywas related to individuals and not to subspecies. Average 2negg frequency was 0·49% for all crosses, and 1·53%for crosses where 2n egg production was detected. Individualfrequencies did not exceed 3·5%, with the exception of26% in one plant. Triploid block was higher and fertility, frequencyof high 2n gamete producers and average 2n gamete frequencywere lower in the 2X-4X crosses than in the opposite 4X-2X crossesinvolving the same plants, previously studied. Diplogynous anddiplandrous gamete production were not correlated with eachother. However, the widespread occurrence of plants producingboth kinds of 2n gametes in almost all the studied subspecies,as well as the presence of several individuals able to producehigh rates of 2n eggs or 2n pollen, support the hypothesis thattetraploids can be formed anywhere and anytime by sexual polyploidizationin diploid populations of Dactylis glomerata L. 2n eggs, polyploidy, Dactylis glomerata L., flow cytometry, sexual polyploidization  相似文献

The effects of irradiations with different proportions of red/farred light and of gibberellic acid on the phytochrome-mediated seed germination of Kalanchoë blossfeldiana cv. Feuerblüte, were studied. The seed coat transmits much more red than far-red light, and therefore the energy ratio between 660 nm and 730 nm is given only for the transmitted light. Decreasing this ratio from 65 to 1.0 caused only a very slight inhibition. If this ratio is further lowered to 0.64, a 10 min terminal irradiation after a 3-h white light photoperiod is inhibitory, but a 12-h photoperiod or continuous irradiation is not. If the ratio is decreased to 0.44 or 0.31, a 12-h photoperiod is now also inhibitory, although continuous irradiation and 10 min terminal irradiation are still more inhibitory. These results are discussed in terms of phytochrome phototransformations. Although gibberellic acid is unable to cause any germination in complete darkness, it can result in a very high germination percentage, if combined with treatments which by themselves do not induce any germination such as continuous far-red, terminal far-red after short photoperiods, or very short photoperiods at 25°C. These results point to a strong synergism between gibberellic acid and the so-called stabilized form of phytochrome, P*FR.  相似文献

SYNOPSIS. In vitro development of Eimeria canadensis from cattle was studied in monolayer cultures of various bovine cell lines grown on coverslips in Leighton tubes. Excysted sporozoites were used for inoculation of the cell cultures. Sporozoites entered the host cells within a few minutes, but apart from a reduction in the number of refractile bodies, changed little in appearance during the first 9 days. Beginning at 91/2 days postinoculation, sporozoites developed into sporozoite-shaped schizonts or, less frequently, transformed into trophozoites. Sporozoite-shaped schizonts with as many as 8 nuclei were observed transforming into spheroid schizonts. At 111/2 days, intermediate schizonts had a characteristic single mass of refractile granules and 60–80 nuclei. Deep invaginations, which resulted in the formation of several blastophores, usually occurred when schizonts had about 100 nuclei. Merozoites were formed as a result of radial outgrowth from the surface of spheroid schizonts as well as of blastophores. Mature merozoites were seen 1st after 13 days.  相似文献

MOST investigators concerned with interferon synthesis in vivo have used the experimental procedure described by Baron and Buckler1, in which circulating interferon is induced by intravenous administration of viruses. When interpreting results, however, it is difficult to know which cells are responsible for circulating interferon synthesis in the animal. Using a radiobiological approach, we have shown that after an intravenous injection of virus, interferon released into the blood stream of mice originates in cell populations of varying radiosensitivities, depending on the virus inoculated2. Myxo-virus-induced circulating interferon production is characterized by high radiosensitivity, for serum interferon titres are decreased by more than 90% in C3H/He mice after one total body X-irradiation of 250 r. Moreover, the species specificity of interferon has enabled us to show that circulating interferon induced by Newcastle disease virus (NDV) is of donor type in xenogeneic radiochimaeras, from which we concluded that cells responsible for interferon synthesis with this virus originate from haemopoietic stem cells3,4. Both granulocytes and lymphocytes fulfil the criteria of very radiosensitive elements derived from haemopoietic stem cells5,6. We wish to report that myxovirus-induced circulating interferon production is selectively depressed after administration of antilymphocyte serum (ALS).  相似文献

In 40 patients percutaneous transhepatic cholangiography was performed by means of the "skinny" Chiba needle. The intrahepatic bile ducts were visualized in 100% of the patients with dilated ducts and in 67% of those with ducts of normal calibre. The results compare to those in the literature and demonstrate the usefulness as well as the reduced morbidity of this new procedure. Therefore nonvisualization of the intrahepatic bile ducts by this method signifies that the ducts are not dilated.  相似文献
