The restoration of endangered relict populations is challenging in conservation biology because they require specific environmental conditions within an inhospitable regional climate. Urothemis edwardsii Selys is the most endangered dragonfly in the Mediterranean with only one known relict small population (Lac Bleu) left in Northeast Algeria. With the absence of successful (re-)colonization over the last two decades, the restoration of the species became a top priority. To improve the status of the species in Northeast Algeria, we carried out a reintroduction and translocation scheme during 2011–2015 and assessed the changes in distribution and population size. Our restoration plan led to the emergence of three populations of which one was restored (Lac Noir), one resulted from successful translocation (Lac Tonga Northeast), and one established after successful colonization (Lac Tonga Southwest). In three localities (Lac Noir, Lac Tonga Northeast, and Lac Tonga Southwest), signs of population growth were observed, whereas no significant trend in the source population (Lac Bleu) was detected. A new population (El Graeate) was also recorded in 2015, but its origin is uncertain. Capture-mark-recapture on adults conducted in 2015 in two sites (Lac Bleu and Lac Noir) showed low recapture rates and no sign of dispersal between the two sites. Dispersal capacity of the species and conservation implications of adult distribution are discussed. This study highlights the importance of using biological indicators in selecting host habitats for the restoration of critically threatened populations.  相似文献

The novel neuropsychotropic agent milacemide hydrochloride (2-n-pentylaminoacetamide HCl) is a highly selective substrate of the B form of monoamine oxidase (EC 1.4.3.4; MAO). Under the in vitro conditions used in the present study, milacemide acts as an enzyme-activated, partially reversible inhibitor of MAO-B. A reversible inhibition of MAO-A activity is also observed at high concentrations. The inhibitory activity of milacemide is significantly greater for MAO-B. In vivo, after single or repeated oral administration, a specific inhibition of MAO-B is apparent in brain and liver, with a lack of inhibition of the MAO-A activity. In contrast to the irreversible inhibitory action of L-deprenyl, the recovery of MAO-B activity in vivo after milacemide administration is significantly faster, a result suggesting that it is a partially reversible inhibitor. The selective inhibitory effect of milacemide for MAO-B in vivo is confirmed by its potentiation of phenylethylamine-induced stereotyped behavior, whereas vasopressor responses to tyramine were not affected. These observations suggest that milacemide could enhance dopaminergic activity in the brain and could be used as therapy for Parkinson's disease in association with L-3,4-dihydroxyphenylalanine.  相似文献

The polymerase-chain reaction was applied for detection of provirus DNA of the bovine leukaemia virus (BLV). A short fragment of 292 bp including region R and U5 LTR 5' of BLV was amplified, and the optimum parameters of amplification of this fragment were established. Electrophoresis revealed the presence of the 292 bp fragment from the leucocytes of four out of six cows showing a positive serological response to BLV antigens. Application of the polymerase-chain reaction in diagnosis of bovine leukaemia is suggested.  相似文献

A noniterative procedure based upon the minimum modified X2 approach is employed to test the model of homogeneity of one-dimensional margins in square tables. Such tables may arise from matched pairs with k outcomes. The special case of double dichotomy (i.e. matched pairs with two outcomes) reduces to the McNemar test statistic. The case of multiple matched controls is also dealt with. The Cochran's Q test is used to test the marginal homogeneity in cases comparing m distinct matched samples in addition to testing trends in proportions. Reference is made to the equivalence between these tests and the approach of hierarchical log-linear models for testing marginal homogeneity of square tables.  相似文献

The quality of the attachment of meat to bone is often reported to be insufficient by more and more poultry's consumers. This is particularly true for thigh meat in broilers. The aim of this study was to compare muscle to bone attachment (namely, tendons) from a biomechanical and a biochemical point of view in 50 standard (S) and 50 Label Rouge (LR) chickens. Carcasses weighted around 1.7 kg in the two groups. Two tendons were harvested and proceeded for passive stretch tests, prior to cooking or not, to determine main mechanical characteristics (maximum load, stiffness and longitudinal strain). Biochemical parameters such as dry matter percentage, total collagen content, collagen solubility and sulphated glycosaminoglycans (sGAGs) content were also determined. Results showed that biomechanical values differ largely between the two studied tendons. For a given tendon, the values were also different between the two groups of chickens mainly after cooking. The results clearly showed that, mainly after cooking, the mechanical resistance of tendon to stretch was better in LR than in S chickens. LR chickens were reported to have tendons with higher collagen and sGAGs contents associated with a lower collagen solubility. These differences may explain biomechanical differences observed for the two types of tendons and could be due to increased age and/or higher physical activity of LR chickens.  相似文献

Cell–cell adhesion couples the contractile cortices of epithelial cells together, generating tension to support a range of morphogenetic processes. E-cadherin adhesion plays an active role in generating junctional tension by promoting actin assembly and cortical signaling pathways that regulate myosin II. Multiple myosin II paralogues accumulate at mammalian epithelial cell–cell junctions. Earlier, we found that myosin IIA responds to Rho-ROCK signaling to support junctional tension in MCF-7 cells. Although myosin IIB is also found at the zonula adherens (ZA) in these cells, its role in junctional contractility and its mode of regulation are less well understood. We now demonstrate that myosin IIB contributes to tension at the epithelial ZA. Further, we identify a receptor type-protein tyrosine phosphatase alpha–Src family kinase–Rap1 pathway as responsible for recruiting myosin IIB to the ZA and supporting contractile tension. Overall these findings reinforce the concept that orthogonal E-cadherin–based signaling pathways recruit distinct myosin II paralogues to generate the contractile apparatus at apical epithelial junctions.  相似文献

Inclusion body myositis, the most common muscle disorder in the elderly, is partly characterized by abnormal expression of amyloid precursor protein (APP) and intracellular accumulation of its proteolytic fragments collectively known as β-amyloid. The present study examined the effects of β-amyloid accumulation on mitochondrial structure and function of skeletal muscle from transgenic mice (MCK-βAPP) engineered to accumulate intramyofiber β-amyloid. Electron microscopic analysis revealed that a large fraction of myofibers from 2-3-month-old MCK-βAPP mice contained numerous, heterogeneous alterations in mitochondria, and other cellular organelles. [(1)H-decoupled](13)C NMR spectroscopy showed a substantial reduction in TCA cycle activity and indicated a switch from aerobic to anaerobic glucose metabolism in the MCK-βAPP muscle. Isolated muscle fibers from the MCK-βAPP mice also exhibited a reduction in cytoplasmic pH, an increased rate of ROS production, and a partially depolarized plasmalemma. Treatment of MCK-βAPP muscle cells with Ru360, a mitochondrial Ca(2+) uniporter antagonist, reversed alterations in the plasmalemmal membrane potential (V(m)) and pH. Consistent with altered redox state of the cells, treatment of MCK-βAPP muscle cells with glutathione reversed the effects of β-amyloid accumulation on Ca(2+) transient amplitudes. We conclude that structural and functional alterations in mitochondria precede the reported appearance of histopathological and clinical features in the MCK-βAPP mice and may represent key early events in the pathogenesis of inclusion body myositis.  相似文献
