With a view toward understanding better the mechanism of action of follitropin, an attempt was made using granulosa cells obtained from intact immature estrogenized rats to study in short-term incubations the effect of highly purified ovine follitropin on the binding of the hormone to the cells and the associated aromatase response. A modified radioimmunoassay procedure has been used to monitor unlabeled physiologically fully active follitropin bound to the cell. A linear relationship between the actual amount of hormone bound to the cells and the estradiol produced in vitro has been established. The amount of ovine follitropin bound that can elicit a half-maximal response in estrogen production was calculated to be 400 pg. The number of follitropin binding sites per cell was 375 and the Kd of binding was 3.03 × 10?10m. By the addition of ovine follitropin antiserum at different time points of a 4-h incubation period, a continual need for follitropin support for estradiol production has been established. 相似文献

The adult male Japanese quail produces white foam from the cloacal gland, which is transferred to the female proctodeum during natural mating. The physiological role of foam on quail spermatozoa is still unclear. Therefore, attempts have been made to understand the effect of cloacal foam on motility and metabolism of quail spermatozoa. The profile of various biochemical constitutes in the foam extract was investigated. The addition of foam extract to neat semen completely disaggregated the clumps of spermatozoa leading to vigorous motility. The metabolic rate (MBRT) of the spermatozoa was significantly increased with the addition of foam extract. The foam extract was sub fractionated into seven different fractions by using the molecular cut off devices. Among all the seven sub-fractions from the foam extract, the addition of < 1 KDa sub-fraction contained lactate and has enhanced sperm motility and metabolism. Another fraction (3-10 KDa) has non-protein and non-heparin components which completely disaggregated the clumped quail spermatozoa. However, the remaining fractions did not show any effect on quail spermatozoa. It can be concluded from the present investigation that the lactate present in foam might be a fuel for sperm metabolism and motility. Cyclic monkeys were deprived of FSH for specific periods on different days of the follicular phase by injecting them with minimal doses of an FSH antiserum characterized for specificity and bioneutralizing ability. The effect of the antiserum on follicular maturation was assessed by determining (a) serum oestrogen concentrations through the midcycle period, (b) serum progesterone concentrations as an index of ovulation and luteal function, (c) laparoscopic examination of the surface of the ovary when necessary, and (d) overall cycle length. While antiserum injection on Day 5 of the cycle caused delay in the oestrogen surge from Days 9 to 11, injection on Day 6 led to the occurrence of two oestrogen surges, on Days 9 and 14. Laparoscopic examination showed that the earlier follicle had disappeared and a new follicle had appeared by Day 14. Antiserum injection on Day 7 of the cycle arrested further growth of the maturing follicle, but a new follicle appeared 9 days later, as indicated by a surge of oestrogen on Day 16. Injection of antiserum beyond Day 7 had no effect on follicular development, ovulation and luteal function. These observations suggest that the mature follicle becomes relatively independent of FSH support about 48 h before ovulation and this event could be a marker for follicular dominance. 相似文献

The present paper summarizes some of the important biological and physiological data recorded over a 30-year period on the
biology of bonnet monkeys in captivity. Data on sexual maturity, menstrual cyclicity, general behaviour, endocrine profile,
reproductive physiology, gestation, parturition, postpartum amenorrhoea in the female, and sexual maturity, hormone profile,
and seasonal variation in sperm count of the male monkeys are presented. In addition to the biological values, weights of
selected organs, vertebral and dental pattern are also presented. Menarche occurred at an age of 36±4 months and the first
conception in the colony occurred at an age of 54±4 months. The average menstrual cycle length was 28±4.3 days. Majority of
monkeys did not cycle regularly during March–June during which the temperature reached a peak. The pregnancy index of the
colony was 80% with controlled breeding. The gestation period was 166±5 days with 6–7 months postpartum amenorrhoea. Males
attained sexual maturity by the age of 6–7 years and exhibited the characteristic nocturnal surge of serum testosterone at

The ability of prolactin to influence the responsiveness of the lactating rat pituitary to luteinising hormone releasing hormone has been examinedin vitro. The pituitary responsivenessin vivo to luteinising hormone releasing hormone decreased as a function of increase in the lactational stimulus. Prolactin inhibited the spontaneousin vitro release of luteinising hormone and follicle stimulating hormone to a small extent, from the pituitary of lactating rats with the suckling stimulus. However, it significantly inhibited the release of these two hormones from luteinising hormone releasing hormone-stimulated pituitaries. The responsiveness of pituitaries of rats deprived of their litter 24 h earlier, to luteinising hormone releasing hormone was also inhibited by prolactin, although minimal. It was concluded that prolactin could be influencing the functioning of the pituitary of the lactating rat by (a) partially suppressing the spontaneous release of gonadotropin and (b) inhibiting the responsiveness of the pituitary to luteinising hormone releasing hormone. 相似文献

By using radioimmunoassay, the interaction of sheep lutropin (luteinizing hormone, LH) beta-subunit with rat ovarian receptors was investigated. The binding of beta-subunit was specific, although of much lower order than that of lutropin. Sheep lutropin beta-subunit effectively inhibited the binding of human choriogonadotropin (chorionic gonadotropin, gCG) to the ovary, showing that both occupy the same sites. The binding of sheep lutropin beta-subunit to ovary was not followed by any detectable increase in cyclic AMP. The ovarian response to lutropin in terms of cyclic AMP production was inhibited in the presence of free beta-subunit. The alpha-subunit of lutropin, when used at concentrations where contamination with whole lutropin was negligible, enhanced the degree of binding of beta-subunit; this did not lead to increased cyclic AMP in the tissue. Surprisingly, the binding of beta-subunit in vitro was drastically decreased by the prior removal of all endogenous rat lutropin bound to receptors. The implications of these data are discussed in the light of the reported biological activity of the beta-subunit. 相似文献
