This protocol describes a method by which a large collection of the leafy green vegetable lettuce (Lactuca sativa L.) germplasm was screened for likely drought-tolerance traits. Fresh water availability for agricultural use is a growing concern across the United States as well as many regions of the world. Short-term drought events along with regulatory intervention in the regulation of water availability coupled with the looming threat of long-term climate shifts that may lead to reduced precipitation in many important agricultural regions has increased the need to hasten the development of crops adapted for improved water use efficiency in order to maintain or expand production in the coming years. This protocol is not meant as a step-by-step guide to identifying at either the physiological or molecular level drought-tolerance traits in lettuce, but rather is a method developed and refined through the screening of thousands of different lettuce varieties. The nature of this screen is based in part on the streamlined measurements focusing on only three water-stress indicators: leaf relative water content, wilt, and differential plant growth following drought-stress. The purpose of rapidly screening a large germplasm collection is to narrow the candidate pool to a point in which more intensive physiological, molecular, and genetic methods can be applied to identify specific drought-tolerant traits in either the lab or field. Candidates can also be directly incorporated into breeding programs as a source of drought-tolerance traits.  相似文献

One of the problems that hamper the use of antisense DNAs as effective drugs is the non-specific binding of chemically-modified oligonucleotides to cellular proteins. We previously showed that the affinity of a model ssDNA-binding protein, the Ff gene 5 protein (g5p), was >300-fold higher for phosphorothioate-modified DNA (S-DNA) than for unmodified dA36, consistent with the propensity of S-DNA to bind indiscriminately to proteins. The current work shows that g5p binding is also sensitive to sugar and pyrimidine modifications used in antisense oligomers. Binding affinities of g5p for 10 36mer oligomers were quantitated using solution circular dichroism measurements. The oligomers contained C-5-propyne (prC), 2′-O-methyl (2′-O-Me) or 2′-OH (RNA) groups, alone or combined with the phosphorothioate modification. In agreement with reported increases in antisense activity, the addition of prC or 2′-O-Me modifications substantially reduced the affinity of oligomers for g5p by ~2-fold compared with the same DNA oligomer sequences containing only phosphorothioate linkages. That is, such modifications moderated the propensity of the phosphorothioate group to bind tightly to the g5p. The Ff g5p could be a useful model protein for assessing non-specific binding effects of antisense oligomer modifications.  相似文献

Two halophilic archaeal strains, R30T and tADLT, were isolated from an aquaculture farm in Dailing, China, and from Deep Lake, Antarctica, respectively. Both have rod-shaped cells that lyse in distilled water, stain Gram-negative and form red-pigmented colonies. They are neutrophilic, require >120?g/l NaCl and 48–67?g/l MgCl2 for growth but differ in their optimum growth temperatures (30?°C, tADLT vs. 40?°C, R30T). The major polar lipids were typical for members of the Archaea but also included a major glycolipid chromatographically identical to sulfated mannosyl glucosyl diether (S-DGD-1). The 16S rRNA gene sequences of the two strains are 97.4?% identical, show most similarity to genes of the family Halobacteriaceae, and cluster together as a distinct clade in phylogenetic tree reconstructions. The rpoB′ gene similarity between strains R30T and tADLT is 92.9?% and less to other halobacteria. Their DNA G?+?C contents are 62.4–62.9?mol?% but DNA–DNA hybridization gives a relatedness of only 44?%. Based on phenotypic, chemotaxonomic and phylogenetic properties, we describe two new species of a novel genus, represented by strain R30T (=?CGMCC 1.10593T?=?JCM 17270T) and strain tADLT (=?JCM 15066T?=?DSMZ 22187T) for which we propose the names Halohasta litorea gen. nov., sp. nov. and Halohasta litchfieldiae sp. nov., respectively.  相似文献

The chromosomes of ciliates are fragmented at reproducible sites during the development of the polyploid somatic macronucleus, but the mechanisms involved appear to be quite diverse in different species. In Paramecium aurelia, the process is imprecise and results in de novo telomere addition at locally heterogeneous positions. To search for possible determinants of chromosome fragmentation, we have studied an ~21-kb fragmentation region from the germ line genome of P. primaurelia. The mapping and sequencing of alternative macronuclear versions of the region show that two distinct multicopy elements, a minisatellite and a degenerate transposon copy, are eliminated by an imprecise mechanism leading either to chromosome fragmentation and the formation of new telomeres or to the rejoining of flanking sequences. Heterogeneous internal deletions occur between short direct repeats containing TA dinucleotides. The complex rearrangement patterns produced vary slightly among genetically identical cell lines, show non-Mendelian inheritance during sexual reproduction, and can be experimentally modified by transformation of the maternal macronucleus with homologous sequences. These results suggest that chromosome fragmentation in Paramecium is the consequence of imprecise DNA elimination events that are distinct from the precise excision of single-copy internal eliminated sequences and that target multicopy germ line sequences by homology-dependent epigenetic mechanisms.  相似文献

In order to illustrate the physiological variation of different generations and different thallus parts of Saccharina japonica, physiological parameters such as maximum and effective PSII photochemical efficiency, nutrient uptake, and elemental composition were determined in the laboratory. Photosynthetic analysis in different generations indicated that, although gametophytes had higher pigment contents than the sporophyte, they had lower values of F v/F m and ΔF/Fm. The highest Chl a/Chl c ratio was found in sporophyte generation (3.98?±?0.01) and in the basal part of fresh thallus (2.66?±?0.02). The sporophyte had significantly higher values of nitrate uptake but lower values of phosphorus uptake than the gametophytes. The contents of nitrogen and carbon as well as C/N in gametophytes were significantly higher than those in sporophytes. In addition, the basal part of the S. japonica thallus had the highest C content (22.31?±?1.50 %) but the lowest N content (2.02?±?0.16 %), as well as the highest value of C/N (11.02?±?0.34).  相似文献

Coastal ocean bacterioplankton control the flow of dissolved organic carbon (DOC) from terrestrial and oceanic sources into the marine food web, and regulate the release of inorganic carbon to atmospheric and offshore reservoirs. While the fate of the chemically complex coastal DOC reservoir has long been recognized as a critical feature of the global carbon budget, it has been problematic to identify both the compounds that serve as major conduits for carbon flux and the roles of individual bacterioplankton taxa in mediating that flux. Here we analyse random libraries of expressed genes from a coastal bacterial community to identify sequences representing DOC‐transporting proteins. Predicted substrates of expressed transporter genes indicated that carboxylic acids, compatible solutes, polyamines and lipids may be key components of the biologically labile DOC pool in coastal waters, in addition to canonical bacterial substrates such as amino acids, oligopeptides and carbohydrates. Half of the expressed DOC transporter sequences in this coastal ocean appeared to originate from just eight taxa: Roseobacter, SAR11, Flavobacteriales and five orders of γ‐Proteobacteria. While all major taxa expressed transporter genes for some DOC components (e.g. amino acids), there were indications of specialization within the bacterioplankton community for others (e.g. carbohydrates, carboxylic acids and polyamines). Experimental manipulations of the natural DOC pool that increased the concentration of phytoplankton‐ or vascular plant‐derived compounds invoked a readily measured response in bacterial transporter gene expression. This highly resolved view of the potential for carbon flux into heterotrophic bacterioplankton cells identifies possible bioreactive components of the coastal DOC pool and highlights differing ecological roles in carbon turnover for the resident bacterial taxa.  相似文献

A Gram-stain negative, rod-shaped, non-motile and pink-pigmented bacterial strain, designated strain HYL7-26T, was isolated from a soil in the Desert Park of Huyang forest located in Xinjiang, China. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain HYL7-26T belongs to the genus Pontibacter in the family Cytophagaceae. The 16S rRNA gene sequence similarity between strain HYL7-26T and type strains of Pontibacter species ranged from 93.2 to 96.0 %. Strain HYL7-26T was found to contain iso-C15:0 (15.9 %), iso-C17:0 3-OH (9.5 %) and summed feature 4 (comprising anteiso-C17:1 B and/or iso-C17:1 I, 21.0 %, as defined by the MIDI system) as the major cellular fatty acids. The major respiratory quinone was identified as MK-7 and the DNA G+C content was determined to be 43.8 mol%. sym-Homospermidine was the major polyamine observed in the cells. On the basis of phenotypic, chemotaxonomic and phylogenetic data, strain HYL7-26T is considered to represent a novel species of the genus Pontibacter, for which the name Pontibacter soli sp. nov. is proposed. The type strain is HYL7-26T (=CCTCC AB 206240T = NRRL B-59490T).  相似文献
