Formation and cell lineage patterns of the shoot apex of maize   总被引:1,自引:1,他引:0  
In maize, glossy (gl) mutants lack the wax layer normally present on the epidermis of young leaves. By insertion mutagenesis, unstable alleles (gl1-m) have been induced at the Gl1 locus. In the gl1-m8 strain, somatic reversions to wild-type frequently result in the formation of large sectors occupying predictable positions in all seedlings' leaves. In studies of 230 gl1-m8 seedlings with large reverted sectors covering around 50% of the first leaf, four patterns of sectoring were recognized: one large sector ending in all leaves at the main midrib (32.6% of cases); one central sector on leaves 1, 3 and 5 (or 2 and 4), corresponding to lateral stripes of Gl1 tissue on the other leaves (7.9%); a sector decreasing or increasing in successive leaves (9.1%); other types with one sector covering a leaf surface between 33 and 50% (19.1%) or with complex variegations (31.3%). Based on leaf sectoring, the pattern and stability of cell lineages during shoot apex establishment and embryonic activity leading to leaf primordia, are inferred from the genetic state (Gl or gl1-m) of leaf founder cells present in the apex at the ring of primordia insertion. A genetic experiment indicates that the large somatic reversions considered derived from both the LI and the LII layers of the apex. A large majority of the observed patterns of reversion can be interpreted as due to a single event of transposition. The data are discussed and relevant conclusions proposed in relation to the age of the proembryo at the time of apex formation, the permanent or impermanent state of initial cells of the apex, the polarization of cell divisions and the plane of early apex cell division as a mechanism leading to the bilateral symmetry of the maize seedling.  相似文献

R1 is a non-long terminal repeat (non-LTR) retrotransposable element that inserts into a specific sequence of insect 28S ribosomal RNA genes. We have previously shown that this element has been maintained through vertical transmission in the melanogaster species subgroup of Drosophila. To address whether R1 elements have been vertically transmitted for longer periods of evolutionary time, the analysis has been extended to 11 other species from four species groups of the genus Drosophila (melanogaster, obscura, testecea, and repleta). All sequenced elements appeared functional on the basis of the preservation of their open-reading frames and consistently higher rate of substitution at synonymous sites relative to replacement sites. The phylogenetic relationships of the R1 elements from all species analyzed were congruent with the species phylogenies, suggesting that the R1 elements have been vertically transmitted since the inception of the Drosophila genus, an estimated 50-70 Mya. The stable maintenance of R1 through the germ line appears to be the major mechanism for the widespread distribution of these elements in Drosophila. In two species, D. neotestecea of the testecea group and D. takahashii of the melanogaster group, a second family of R1 elements was also present that differed in sequence by 46% and 31%, respectively, from the family that was congruent with the species phylogeny. These second families may represent occasional horizontal transfers or, alternatively, they could reflect the ability of R1 elements to diverge into new families within a species and evolve independently.   相似文献

Attitudes of the parents may seriously interfere with the handicapped child''s ability to develop his maximum level of functioning and adjustment. Primate--and, specifically, monkey--malaria infections are commonly used for understanding the pathology of and immune response to the human disease because they are thought to resemble most closely the host-parasite relationship found in humans. Plasmodium cynomolgi is used extensively as a model for the human parasite, P. vivax, and P. knowlesi is used primarily as a model for the development of erythrocytic-stage vaccines. Both of these simian parasites can naturally infect man, resulting in mildly symptomatic episodes of the disease. The phylogenetic relationship between these two simian parasites and previously characterized Plasmodium species, including P. vivax, was examined by comparison of the asexually expressed small- subunit ribosomal RNA genes. Our analysis confirmed that P. vivax is most closely related to P. cynomolgi and that it remains an appropriate model of the human pathogen. Furthermore, with P. knowlesi and P. fragile, these two species form a group of closely related species, distant from other Plasmodium species. What is considered to be the most ancient of the human malaria pathogens, P. malariae, was also included in the analysis and does not group at all with other simian or human parasites.   相似文献

Kenaf (Hibiscus cannabinus L.) forage potential can be enhanced through its regrowth capacity and higher production in narrow rows. A field experiment was conducted in Matamoros, Coahuila, Mexico, during 2 growing seasons (2004 and 2005) to study the effects of plant height and row spacing on kenaf forage potential with multiple harvests. This study evaluated the effects of (1) 2 plant heights at cutting (1.0-1.2 m and 1.8-2.0 m) and (2) 4 inter row spacings (0.19, 0.38, 0.57 and 0.76 m) using a 2 x 4 factorial arrangement of treatments in a completely randomized block design with 4 replications. Dry matter (DM) and crude protein (CP) yields, DM partitioning, neutral detergent fiber (NDF) and CP concentrations were determined. Heights at cutting × row spacing interactions were not significant for the monitored variables (p>0.05). Kenaf response to treatments was only relevant for main effects (p≤0.05). Row spacing and plant height affected DM and CP yields (p≤0.05), whereas only plant height affected chemical composition and DM partitioning (p≤0.05). Dry matter (17.0%-26.0%), and CP (12.4%-15.6%) yields were higher (p≤0.05) when plant heights had reached 1.8 to 2.0 m. Row spacing reduction from 0.76 m to 0.38 and 0.19 m increased DM yield (20.4-33.4%) and CP yield (24.2-38.5%) (p≤0.05). Kenaf forage potential increases when planted in narrow rows and harvested 2 or 3 times during the growing season.  相似文献

We exploited the AFLP®1(AFLP® is a registered trademark of Keygene, N.V.) technique to map and characterise quantitative trait loci (QTLs) for grain yield and two grain-related traits of a maize segregating population. Two maize elite inbred lines were crossed to produce 229 F2 individuals which were genotyped with 66 RFLP and 246 AFLP marker loci. By selfing the F2 plants 229 F3 lines were produced and subsequently crossed to two inbred testers (T1 and T2). Each series of testcrosses was evaluated in field trials for grain yield, dry matter concentration, and test weight. The efficiency of generating AFLP markers was substantially higher relative to RFLP markers in the same population, and the speed at which they were generated showed a great potential for application in marker-assisted selection. AFLP markers covered linkage group regions left uncovered by RFLPs; in particular at telomeric regions, previously almost devoided of markers. This increase of genome coverage afforded by the inclusion of the AFLPs revealed new QTL locations for all the traits investigated and allowed us to map telomeric QTLs with higher precision. The present study has also provided an opportunity to compare simple (SIM) and composite interval mapping (CIM) for QTL analysis. Our results indicated that the method of CIM employed in this study has greater power in the detection of QTLs, and provided more precise and accurate estimates of QTL positions and effects than SIM. For all traits and both testers we detected a total of 36 QTLs, of which only two were in common between testers. This suggested that the choice of a tester for identifying QTL alleles for use in improving an inbred is critical and that the expression of QTL alleles identified may be tester-specific.  相似文献

Novel proapoptotic Smac mimics/IAPs inhibitors have been designed, synthesized and characterized. Computational models and structural studies (crystallography, NMR) have elucidated the SAR of this class of inhibitors, and have permitted further optimization of their properties. In vitro characterization (XIAP BIR3 and linker-BIR2–BIR3 binding, cytotox assays, early ADMET profiling) of the compounds has been performed, identifying one lead for further in vitro and in vivo evaluation.  相似文献

Background  

The physiological regulation of ciliary beat frequency (CBF) within the fallopian tube is important for controlling the transport of gametes and the fertilized ovum. Progesterone influences gamete transport in the fallopian tube of several mammalian species. In fallopian tubes isolated from cows, treatment with 20 micromolar progesterone caused a rapid reduction of the tubal CBF. The aims of this study were to establish methodology for studying fallopian tube CBF in the mouse, as it is an important model species, and to investigate if progesterone rapidly affects the CBF of mice at nM concentrations.  相似文献
