Yams (Dioscorea spp.) consist of approximately 600 species. Presently, these species are threatened by genetic erosion due to many factors such as pest attacks and farming practices. In parallel, complex taxonomic boundaries in this genus makes it more challenging to properly address the genetic diversity of yam and manage its germplasm. As a first step toward evaluating and preserving the genetic diversity yam species, we use a phylogenetic diversity (PD) approach that has the advantage to investigate phylogenetic relationships and test hypotheses of species monophyly while alleviating to the problem of ploidy variation within and among species. The Bayesian phylogenetic analysis of 62 accessions from 7 species from three regions of Cameroon showed that most Dioscorea sections were monophyletic, but species within sections were generally non-monophyletic. The wild species D. praehensilis and cultivated D. cayenensis were the species with the highest PD. At the opposite, D. esculenta has a low PD and future studies should focus on this species to properly address its conservation status. We also show that wild species show a stronger genetic structure than cultivated species, which potentially reflects the management of the yam germplasm by farmers. These findings show that phylogenetic diversity is a promising approach for an initial investigation of genetic diversity in a crop consisting of closely related species.  相似文献

In an attempt to minimize observer bias, numerical taxonomy methods were used to describe and classify humpback whale sounds. The spectrograms (N = 1255) were digitized into a 16 × 21 binary matrix. The rows were 16 frequencies selected on a logarithmic scale (0.12–8 kHz). The columns were 21 time samples taken every 0.1 s. Each point of the matrix was coded 1 if it lay over part of the sound. Other binary variables were added to code for relative intensity within a sound, frequency modulation and amplitude modulation. The sounds were then compared using the Jaccard similarity coefficient for binary data, and classified with average linkage cluster analysis. This technique produced 115 clusters, which were compared with my aural and visual impressions of the sounds. I agreed with most major categories identified by cluster analysis, but many small clusters had to be fused to other categories. This was partially due to the technique used, and to the complexity of the repertoire under study. Improvements are proposed to further reduce observer bias in classification of sounds, and thus make studies of animal communication performed by different researchers or on different species more easily comparable.  相似文献

Onion maggot (Delia antiqua) is an economically important pest of Allium crops in temperate regions throughout the world. Management of this pest is necessary to achieve economic returns and depends on insecticide regimes and cultural management. Current cultural management especially altering planting date, field location and crop rotation depend on monitoring. We evaluated the effect of shape, size, colour and chemical attractants on trap catch of field populations of adult D. antiqua flies in upstate New York. White, large diameter, spherical traps in conjunction with Delia Lure attractant performed the best in attracting and catching D. antiqua adults. These results suggest an improved means of attracting and capturing D. antiqua populations which could be useful in monitoring efforts and development of attract and kill strategies for pest control.  相似文献

Antimicrobial peptides possess cationic and amphipathic properties that allow for interactions with the membrane of living cells. Bacteriocins from lactic acid bacteria, in particular, are currently being studied for their potential use as food preservatives and for applications in health care. However, bacteriocin exploitation is often limited owing to low production yields. Gene cloning and heterologous protein or peptide production is one way to possibly achieve overexpression of bacteriocins to support biochemical studies. In this work, production of recombinant active pediocin PA-1 (PedA) was accomplished in Escherichia coli using a thioredoxin (trx) gene fusion (trx-pedA) expression approach. Trx-PedA itself did not show any biological activity, but upon cleavage by an enterokinase, biologically active pediocin PA-1 was obtained. Recombinant pediocin PA-1 characteristics (molecular mass, biological activity, physicochemical properties) were very similar to those of native pediocin PA-1. In addition, a 4- to 5-fold increase in production yield was obtained, by comparison with the PA-1 produced naturally by Pediococcus acidilactici PAC 1.0. The new production method, although not optimized, offers great potential for supporting further investigations on pediocin PA-1 and as a first-generation process for the production of pediocin PA-1 for high-value applications.  相似文献

In the humid forest zone of Southern Cameroon, farmers generally associate cocoa with native and exotic trees in complex agroforestry systems. Despite the socio-economic and ecological importance of these systems, few studies have investigated their plant composition. We investigated tree composition of cocoa agroforests along a gradient of market access, population density and resource use intensity in the humid forest zone of southern Cameroon, comprising (i) the sub-region of Yaoundé, (ii) the sub-region of Mbalmayo, and (iii) the sub-region of Ebolowa. Market access, population density and resource use intensity all decreased from the first to the third sub-region. We quantified the diversity of tree species associated with cocoa within individual agroforests, among agroforests in the same region, and among the three sub-regions, and classified the tree species according to their main uses. A total of 9.1 ha belonging to 60 cocoa agroforests were inventoried in 12 villages. We encountered a total of 206 tree species with an average of 21 tree species per agroforest. In the more urbanized area around Yaoundé, agroforests were less diverse than in the other sub-regions. In all of the agroforests, food producing tree species tended to be more frequent than other species. Two thirds of the food trees were native forest species and one third was introduced. From Ebolowa to Yaoundé, the density of food producing trees doubled and the density of exotic food-producing species increased relative to native species. Some local species producing high-value non-timber forest products were found in the agroforests, but their density was far lower than that of exotic tree species. The agroforests also provide medicine, charcoal and other products for household consumption and sale. We conclude that unless there are specific efforts to promote local forest tree species in cocoa agroforests, these will progressively lose importance with increasing market access, population pressure and land use intensity.  相似文献

Desulfovibrio species are representatives of microorganisms at the boundary between anaerobic and aerobic lifestyles, since they contain the enzymatic systems required for both sulfate and oxygen reduction. However, the latter has been shown to be solely a protective mechanism. By implementing the oxygen-driven experimental evolution of Desulfovibrio vulgaris Hildenborough, we have obtained strains that have evolved to grow with energy derived from oxidative phosphorylation linked to oxygen reduction. We show that a few mutations are sufficient for the emergence of this phenotype and reveal two routes of evolution primarily involving either inactivation or overexpression of the gene encoding heterodisulfide reductase. We propose that the oxygen respiration for energy conservation that sustains the growth of the O2-evolved strains is associated with a rearrangement of metabolite fluxes, especially NAD+/NADH, leading to an optimized O2 reduction. These evolved strains are the first sulfate-reducing bacteria that exhibit a demonstrated oxygen respiratory process that enables growth.  相似文献

Insects provide examples of many cunning stratagems to cope with the challenges of living in a world dominated by surface forces. Despite being the current masters of the land environment, they are at constant risk of being entrapped in liquids, which they prevent by having waxy and hairy surfaces. The problem is particularly acute in an enclosed space, such as a plant gall. Using secreted wax to efficiently parcel and transport their own excrement, aphids were able to solve this problem 200 Myr ago. Here, we report on the physical and physiological significance of this ingenious solution. The secreted powdery wax has three distinct roles: (i) it is hydrophobic, (ii) it creates a microscopically rough inner gall surface made of weakly compacted wax needles making the gall ultra-hydrophobic, and (iii) it coats the honeydew droplets converting them into liquid marbles, that can be rapidly and efficiently moved.  相似文献
