Transient time courses ("induction") and the intensity of thedelayed fluorescence of chlorophyll a (measured between 0.1and 3.9 msec after a 0.9 msec excitation period) were studiedwith a phosphoroscope at temperatures between 40 and –170°Cin Tris-washed chloroplasts. Tris-washing of chloroplasts changed the temperature dependenciesof the induction and the intensity of the delayed fluorescence.From the analysis of the induction each photosystem II reactioncenter appears to be linked to a donor pool which can supplyone electron to the acceptor pool in Tris-washed chloroplasts. An artificial electron donor, diphenylcarbazide affected thedelayed fluorescence above –100°C evidence that electronsare donated to photosystem II in at least two different ways. An electron transport inhibitor, 3-(3',4'-dichlorophenyl)-l,l-dimethylurea,changed the induction of the delayed fluorescence at temperaturesabove –60°C. The temperature dependence of the electron transport in thevicinity of photosystem II was characterized from these results. (Received May 27, 1980; )  相似文献

Regulation of cytosol 5'-nucleotidase by adenylate energy charge   总被引:5,自引:0,他引:5  
In the physiological range of the adenylate energy charge in liver (0.7-0.9), th rate of AMP-hydrolysis catalysed by rat liver cytosol 5'-nucleotidase (5'-ribonucleotide phosphohydrolase, EC 3.1.3.5) increased sharply with decreasing energy charge. In addition, a decrease in the concentration of Pi caused marked acceleration of the AMP-hydrolysing activity over the physiological range of adenylate energy charge. These responses seem to serve to protect the cells against a metabolic stress which could result from sudden utilization of ATP by removal of AMP. The AMP-hydrolysing activity of this enzyme decreased sharply as the size of the adenine nucleotide pool decreased in the physiological range. This effect may be a self-limiting response to prevent excess depletion of the pool. IMP-hydrolysing activity of this enzyme increased with increasing adenylate energy charge. But no marked response to its variation within the physiological range was observed. On the basis of the data obtained in this study, the IMP-hydrolysing activity of the cytosol 5'-nucleotidase in rat liver cells seems to be comparable to that of AMP deaminase reaction, but the AMP-hydrolysing activity was estimated to be less than 10% of AMP deaminase reaction at energy charge value of about 0.7. This strongly suggests that the AMP leads to IMP leads to inosine pathway is more significant that the AMP leads to adenosine leads to inosine pathway in rat liver.  相似文献

The concentration of ACTH in extracts of rat anterior pituitary was measured by both radioimmunoassay and bioassay at different stages following adrenalectomy. Both types of ACTH activity decreased the day immediately following adrenalectomy but increased gradually afterwards. Immunological ACTH activity increased to 250% of the control value and biological ACTH activity increased to 490% of control value 3 weeks after adrenalectomy. The increase in biological ACTH activity occurred earlier, and the rate of increase was greater, than that of the immunological ACTH activity. The distributions of molecular weight forms of ACTH in extracts of anterior pituitary lobes was determined by gel filtration. Three molecular weight forms of immunoassayable ACTH were detected. Biological ACTH activity appeared in the 2nd and the 3rd peaks. A striking change was observed after adrenalectomy in the distribution of biologically active forms of ACTH. The ratio of biological ACTH activity to immunological ACTH activity in each peak changed at various stages after adrenalectomy. This indicated the heterogenous nature of the ACTH included in each peak. At 2 and again at 3 weeks, biological activity markedly increased until it exceeded the immunological ACTH activity in the 2nd peak. Dexamethasone had little influence on the elution profile of either immunoassayable and biologically active ACTH in gel filtration. Adrenalectomy may possibly have an effect on the intracellular posttranslational processing of ACTH precursors which leads to the development of biological ACTH activity.  相似文献

Using B-mode ultrasonography, an attempt was made to measure the volume of extraocular muscles and retrobulbar fat in 31 patients (62 orbits) with Graves' disease. None of the patients had exophthalmometric measurements greater than 21 mm or had eye symptoms. The mean value of muscle volume of Graves' patients was significantly larger than that of normal controls (6.48 +/- 2.70 cm3 and 3.25 +/- 1.30 cm3, respectively, p less than 0.001). All of the patients had extraocular muscle swelling, although 2 of them had no extraocular muscle change for their unilateral eye. The extraocular muscle volume increased as the degree of the proptosis increased. The fat volume tended to increase in parallel with the degree of the proptosis. In the Graves' group with obvious proptosis (Hertel reading: 19--21 mm), the fat volume increased more significantly than in any other group. The ratio of extraocular muscle volume to retrobulbar fat volume was significantly higher in Graves' disease, but it did not increase as the degree of the proptosis increased. A significant correlation between proptosis and muscle volume plus fat volume was observed. No significant difference of the extraocular muscle volume was observed between the patients untreated and treated with antithyroid drugs. The data show a uniform enlargement of the extraocular muscles in Graves' disease and also suggest an involvement of increased retrobulbar fat volume in a group of obvious exophthalmos. The degree of the proptosis is in aclose proportion ot the quantitative change of the orbital soft tissue.  相似文献

β-Helical poly(L -glutamic acid) in a gel state was found to be easily converted to the antiparallel β form by heating. Two β forms were obtained, depending on the temperature of heating. Temperatures between 40° and 85°C produced a β form with a spacing between pleated sheets (d001) of 9.03 Å, termed β1. If the heating was carried out at temperatures higher than 85°C, the β1 form underwent another conformational transition reducing the d001 value from 9.03 to 7.83 Å (termed β2) without any prominent change in the fiber repeat distance (i.e., the polypeptide backbone conformation). The time course of these two transitions was followed by measuring the infrared spectra of the samples, and it was concluded that the α → β1 transition in its initial stage obeys a pseudo-first order rate process with activation enthalpy and entropy of 54 kcal/mol and 92 eu, respectively. On the other hand, the typical sigmoidal conversion curves observed for the transition between the two types of β forms (β1 → β2) indicate that this transition proceeds via a socalled “nucleation and growth” process. The kinetic theory of phase transitions developed by Avrami can be applied with success to explain this transition. The infrared spectra, in the region from 1800 to 200 cm?1, were measured for these two β forms and the results showed that the conformation of the side chains and the mode of the hydrogen bonding between the side-chain carboxyl groups undergo appreciable change during the transition. The heat-induced conformational transition of poly(L -Glu78 L -Val22) was also studied. The copolymer was transformed from the α-helical conformation directly to the β2 form. The reason for this was thought to be due to the fact that the L -valine residues and the L -glutamyl residues near the L -valine residues have a strong tendency to take the more compact β2 form.  相似文献

Taka amylase A glycopeptide (TA-GP) strongly inhibited the interaction of antimannan (antibodies directed towards mannan from Saccharomyces cerevisiae) with yeast mannan, whereas ovalbumin glycopeptide (OA-GP) did so only poorly. We inferred that this is due to the strong reactivity of antimannan with terminal trimannosides composed of Manα1→2Man or Manα1→3Man linkages which occur in mannan and TA-GP. In contrast, TA-GP and OA-GP were nearly equally reactive with concanavalin A having the ability to interact with terminal mannose and 2-0-mannose residues which occur abundantly in these glycopeptides. Thus, antimannan should be useful as a probe for characterizing glycoproteins from extracellular fluids or cellular membranes.  相似文献

A versatile, two-step chromatographic method using DEAE-Toyopearl(Toyo Soda, Japan) is described for purifying photosystem IIreaction center complex from digitonin extracts of spinach thylakoidmembranes. The method is very simple and brings about an approximatefour-fold increase in the specific activity, on a chlorophyllbasis, of 2,4-dichlorophenol-indophenol photoreduction with1,5-diphenylcarbazide (to about 2,000 µ electron equivalentsper mg chlorophyll per h), with an approximate 40 percent recoveryin chlorophyll. The SDS-polyacrylamide gel electrophoresis performedin the presence of 4 M urea in the analyzing gel shows fourpolypeptide bands of the photosystem II reaction center of about47, 43, 30 and 9 kilodaltons. The absorption and fluorescence properties, as well as the pigmentand chemical compositions and the above mentioned polypeptideprofile of the purified complex are essentially identical withthose of the preparations isolated by the previously describedmethod (Satoh 1982). The digitonin solubilization of thylakoid membranes destroysthe water splitting machinery, so that the purified complexshows no oxygen evolving activity, even although 0.6–0.7atoms of manganese per 50 chlorophyll molecules still remain. Transcutaneous pressure with pressure probes of arbitrary diameters have been commonly used for measuring the threshold and magnitude of muscle pain, yet this procedure lacks scientific validation. To examine the valid probe dimensions, we conducted physiological experiments using 34 human subjects. Pin-prick pain, pressure pain threshold (PPT) to pressure probes of various diameters, heat pain threshold, and electrical pain threshold of deep tissues were measured before and after application of surface lidocaine anesthesia to the skin surface over the brachioradial muscle in a double-blinded manner. The anesthesia neither affected PPT with larger probes (diameters: 1.6 and 15?mm) nor increased electric pain threshold of deep structures, whereas it diminished pain count in pin-prick test and PPT with a 1.0?mm diameter probe, suggesting that mechanical pain thresholds measured with 1.6 and 15?mm probes reflect the pain threshold of deep tissues, possibly muscle. Pain thresholds to heat did not change after application of the anesthesia. These results suggest that larger pressure probes can give a better estimation of muscular pain threshold.  相似文献

Many decapod crustaceans in marine intertidal habitats release larvae toward coastal oceans, from which postlarvae (decapodids: settling-stage larvae) return home. Decapodid settlement processes are poorly understood. Previous studies showed that in Kyushu, Japan, the callianassid shrimp population on an intertidal sandflat of an open bay joining the coastal ocean near a large estuary released eight batches of larvae basically in a semilunar cycle from June through October and that decapodids performed diel vertical migration, occurring in the water column nocturnally. We conducted (a) frequent sampling for population density and size-composition on the sandflat through one reproductive season, (b) planktonic and benthic sampling for decapodids around the bay mouth, and (c) current meter deployment at three points across the bay mouth for tidal harmonic analysis. On the sandflat, six batches of newly-settled decapodids (settlers) occurred in a semilunar periodicity until October, with peaks occurring 0–3 days before syzygy dates except for the first one. For larval Batches 1–4, buoyancy-driven shoreward subsurface currents during July to mid-October would transport some pre-decapodid-stage larvae (zoeae) toward the bay. The absence of expected settler Batches 7–8 would be due to the converse subsurface currents caused by water-column mixing and seasonal winds after mid-October, carrying zoeae offshore. Once in the bay, phasing of night and nighttime-averaged shoreward tidal current explained the settlement pattern for Batches 1–4. For Batches 5–6 occurring in mid-September to mid-October, water currents generated by seasonal wind and tidal forcings may have caused peak settlement after the time expected from tidally-driven decapodid transport.  相似文献
