The first step in developing regenerative medicine approaches to treat renal diseases using pluripotent stem cells must be the generation of intermediate mesoderm (IM), an embryonic germ layer that gives rise to kidneys. In order to achieve this goal, establishing an efficient, stable and low-cost method for differentiating IM cells using small molecules is required. In this study, we identified two retinoids, AM580 and TTNPB, as potent IM inducers by high-throughput chemical screening, and established rapid (five days) and efficient (80% induction rate) IM differentiation from human iPSCs using only two small molecules: a Wnt pathway activator, CHIR99021, combined with either AM580 or TTNPB. The resulting human IM cells showed the ability to differentiate into multiple cell types that constitute adult kidneys, and to form renal tubule-like structures. These small molecule differentiation methods can bypass the mesendoderm step, directly inducing IM cells by activating Wnt, retinoic acid (RA), and bone morphogenetic protein (BMP) pathways. Such methods are powerful tools for studying kidney development and may potentially provide cell sources to generate renal lineage cells for regenerative therapy. 相似文献

Summary Males of the melon flyDacus cucurbitae mate with females for 10 hours or more, usually starting at dawn and terminating at dusk. We tested the sperm-loading hypothesis
(Dickinson, 1986) that males remaining with females for long periods of time benefit by numerically overwhelming the sperm of their
competitors. The amount of sperm transferred to a female increased with time after mounting. The number of feamles which laid
eggs at least once during experimental periods was positively correlated with mating duration. Oviposition rate was positively
correlated with mating duration, as well. Egg hatchability was not influenced by mating duration. Mating duration was a major
determinant of paternity when females were doubly mated with one male for 6 hours and another male for 2 hours. Females whose
first matings were longer showed first male sperm predominance, while females whose second matings were longer showed last

The genome organizer special AT‐rich sequence binding protein 1 (SATB1) regulates specific functions through chromatin remodeling in T helper cells. It was recently reported by our team that T cells from SATB1 conditional knockout (SATB1cKO) mice, in which the Satb1 gene is deleted from hematopoietic cells, impair phosphorylation of signaling molecules in response to T cell receptor (TCR) crosslinking. However, in vivo T cell responses upon antigen presentation in the absence of SATB1 remain unclear. In the current study, it was shown that SATB1 modulates T cell antigen responses during the induction and effector phases. Expression of SATB1 was upregulated in response to TCR stimulation, suggesting that SATB1 is important for this antigen response. The role of SATB1 in TCR responses and induced experimental autoimmune encephalomyelitis (EAE) was therefore examined using the myelin oligodendrocyte glycoprotein peptide 35‐55 (MOG35‐55) and pertussis toxin. SATB1cKO mice were found to be resistant to EAE and had defects in IL‐17‐ and IFN‐γ‐producing pathogenic T cells. Thus, SATB1 expression appears necessary for T cell function in the induction phase. To examine SATB1 function during the effector phase, a tamoxifen‐inducible SATB1 deletion system, SATB1cKO‐ER‐Cre mice, was used. Encephalitogenic T cells from MOG35‐55‐immunized SATB1cKO‐ER‐Cre mice were transferred into healthy mice. Mice that received tamoxifen before the onset of paralysis were resistant to EAE. Furthermore, no disease progression occurred in recipient mice treated with tamoxifen after the onset of EAE. Thus, SATB1 is essential for maintaining TCR responsiveness during the induction and effector phases and may provide a novel therapeutic target for T cell‐mediated autoimmune diseases. 相似文献

Background
The neural mechanisms underlying body dissatisfaction and emotional problems evoked by social comparisons in patients with anorexia nervosa (AN) are currently unclear. Here, we elucidate patterns of brain activation among recovered patients with AN (recAN) during body comparison and weight estimation with functional magnetic resonance imaging (fMRI).Methods
We used fMRI to examine 12 patients with recAN and 13 healthy controls while they performed body comparison and weight estimation tasks with images of underweight, healthy weight, and overweight female bodies. In the body comparison task, participants rated their anxiety levels while comparing their own body with the presented image. In the weight estimation task, participants estimated the weight of the body in the presented image. We used between-group region of interest (ROI) analyses of the blood oxygen level dependent (BOLD) signal to analyze differences in brain activation patterns between the groups. In addition, to investigate activation outside predetermined ROIs, we performed an exploratory whole-brain analysis to identify group differences.Results
We found that, compared to healthy controls, patients with recAN exhibited significantly greater activation in the pregenual anterior cingulate cortex (pgACC) when comparing their own bodies with images of underweight female bodies. In addition, we found that, compared with healthy controls, patients with recAN exhibited significantly smaller activation in the middle temporal gyrus corresponding to the extrastriate body area (EBA) when comparing their own bodies, irrespective of weight, during self-other comparisons of body shape.Conclusions
Our findings from a group of patients with recAN suggest that the pathology of AN may lie in an inability to regulate negative affect in response to body images via pgACC activation during body comparisons. The findings also suggest that altered body image processing in the brain persists even after recovery from AN.37.
A field observation of the dragonflyNannophya pygmaea revealed that males prefer some territorial sites to others, and that these same sites attract more females than others (Tsubaki
& Ono, 1986, 1987). In this paper, we asked if males choose territorial sites in response to female dispersion or distribution
of resources. We conducted 3 types of removal experiments to test the following 2 hypotheses; (1) a male may assess the territory
quality by the female encounter rate at his site (learning), (2) a male may assess the resource quality (or quantity) in the
territory. The results of our experiments show that males discriminate attractive and less attractive territorial sites without
any mating experience within the study area. Moreover, the territorial site preference of males was not affected by the mating
experience. Therefore, males probably choose territorial sites by resource quality rather than by female dispersion. 相似文献

Central place foraging by larvae of the charaxine butterfly,Polyura pyrrhus, was studied. Larvae made foraging trips from the silken pads they constructed on leaflets of their foodplant,Acacia sp. A foraging trip sometimes involved complete depletion of a single patch of foodplant pinnules. Larvae which did not deplete
a patch appeared to eat until they were satiated, whereas larvae which depleted a patch either visited another patch (multiple-patch
foraging) or returned directly to the pad (single-patch foraging). If the food intake at the first patch was small a larva
tended to make a “multiple-patch” decision, especially when the pinnule-patch was distant from the resting pad. The duration
between successive foraging trips (resting time on the pad) was much longer than the round trip duration: on average about
3 h and 15 min, respectively. The resting time is suggested to be a handling time (i.e., digesting food in the gut) and was
disproportional to the amount of food consumed, i.e., the handling efficiency was higher when the larva consumed a larger
amount of food. This may be the reason why larvae usually ate until they were satiated.
A food-intake-rate maximizing model was constructed to describe the decision rule as to whether a larva should make a single-patch
or a multiple-patch foraging trip. One of the model's predictions (i.e., larvae should engage in multiple-patch foraging when
the food intake at the first patch is small) qualitatively corresponds with data, however, the model does not explain the
effect of travelling time on decision making in larvae. Several other factors which may influence the decision making of larvae

Male rat liver microsomes oxidized androsta-5,16-dien-3 beta-ol (delta 16-ANDO) to delta 16-ANDO-5,6 alpha-, -5,6 beta-, -16,17 alpha-, and -16,17 beta-epoxides and delta 16-ANDO-5 alpha,6 beta-, -16 alpha,17 beta-, and -16 beta,17 alpha-glycols in the presence of an NADPH-generating system and the microsomal lipid peroxidation accelerator, Fe2+-ADP. The hepatic microsomes hydrolyzed all the delta 16-ANDO epoxides to the glycols. delta 16-ANDO-5 alpha,6 beta-glycol was the sole metabolite from both 5,6 alpha- and 5,6 beta-epoxides. Microsomal epoxide hydrolase also hydrolyzed delta 16-ANDO-16,17 alpha-epoxide specifically to the 16 beta,17 alpha-glycol and the isomeric 16,17 beta-epoxide to the 16 alpha,17 beta- and 16 beta,17 alpha-glycols approximately in the equal ratio. The delta 5-epoxidation of delta 16-ANDO by microsomes occurred only under the conditions that lipid peroxidation took place. Direct evidence was obtained for the participation of microsomal lipid hydroperoxides in the epoxidation of delta 16-ANDO by using photochemically prepared hydroperoxides of phospholipids separated from the hepatic microsomes. The hydroperoxides generated active oxygens, tentatively assigned as alk(ylper)oxy radicals, by the action of ferrous ion and epoxidized delta 16-ANDO to afford the 5,6- and 16,17-epoxides. The Fe2+-ADP-mediated epoxidation of delta 16-ANDO by the phospholipid hydroperoxides occurred preferentially at delta 5 to delta 16 and afforded the 5,6 beta-epoxide in a higher ratio than the 5,6 alpha-epoxide, similar to the Fe2+-ADP-mediated microsomal epoxidation, while the alpha-epoxide was preferentially formed to the beta-epoxide for delta 16 in the epoxidation by both systems. 相似文献

Individual differences in several reproductive parameters of female Pieris rapae were investigated in a controlled laboratory condition. Lifetime and age-specific fecundity showed considerable variability between individuals. Larger females began oviposition at an earlier age than smaller ones, and larger females were more fecund than smaller ones. Larger females laid a larger proportion of their eggs in the early stages of their reproductive lifetime, whilst smaller females laid the larger proportion of their eggs later in their reproductive lifetime. The significance of the variance in age-specific fecundity associated with female size is discussed with respect to the seasonal change in size and habitat utilization of this species. 相似文献
