Background
We have previously shown that lung collectins, surfactant protein A (SP-A) and surfactant protein D, interact with Toll-like receptor (TLR) 2, TLR4, or MD-2. Bindings of lung collectins to TLR2 and TLR4/MD-2 result in the alterations of signaling through these receptors, suggesting the immunomodulatory functions of lung collectins. Mannose binding lectin (MBL) is another collectin molecule which has structural homology to SP-A. The interaction between MBL and TLRs has not yet been determined.Methods
We prepared recombinant MBL, and analyzed its bindings to recombinant soluble forms of TLR4 (sTLR4) and MD-2.Results
MBL bound to sTLR4 and MD-2. The interactions were Ca2+-dependent and inhibited by mannose or monoclonal antibody against the carbohydrate-recognition domain of MBL. Treatment of sTLR4 or MD-2 by peptide N-glycosidase F significantly decreased the binding of MBL. SP-A bound to deglycosylated sTLR4, and this property did not change in chimeric molecules of SP-A/MBL in which Glu195–Phe228 or Thr174–Gly194 of SP-A were replaced with the corresponding MBL sequences.General Significance
These results suggested that MBL binds to TLR4 and MD-2 through the carbohydrate-recognition domain, and that oligosaccharide moieties of TLR4 and MD-2 are important for recognition by MBL. Since our previous studies indicated that lung collectins bind to the peptide portions of TLRs, MBL and lung collectins interact with TLRs by different mechanisms. These direct interactions between MBL and TLR4 or MD-2 suggest that MBL may modulate cellular responses by altering signals through TLRs. 相似文献72.

A terrestrial ecosystem model, called the Vegetation Integrative Simulator for Trace gases model (VISIT), which fully integrates
biogeochemical carbon and nitrogen cycles, was developed to simulate atmosphere–ecosystem exchanges of greenhouse gases (CO2, CH4, and N2O), and to determine the global warming potential (GWP) taking into account the radiative forcing effect of each gas. The
model was then applied to a cool-temperate deciduous broad-leaved forest in Takayama, central Japan (36°08′N, 137°25′E, 1420 m
above sea level). Simulations were conducted at a daily time step from 1948 to 2008, using time-series meteorological and
nitrogen deposition data. VISIT accurately captured the carbon and nitrogen cycles of this typical Japanese forest, as validated
by tower and chamber flux measurements. During the last 10 years of the simulation, the model estimated that the forest was
a net greenhouse gas sink, having a GWP equivalent of 1025.7 g CO2 m−2 y−1, most of which (1016.9 g CO2 m−2 y−1) was accounted for by net CO2 sequestration into forest biomass regrowth. CH4 oxidation by the forest soil made a small contribution to the net sink (11.9 g CO2-eq. m−2 y−1), whereas N2O emissions were a very small source (3.2 g CO2-eq. m−2 y−1), as expected for a volcanic soil in a humid climate. Analysis of the sensitivity of GWP to changes in temperature, precipitation,
and nitrogen deposition indicated that warming temperatures would decrease the size of the sink, mainly as a result of increased

Novel splice variants of ING4 and their possible roles in the regulation of cell growth and motility
The ING4 gene is a candidate tumor suppressor gene that functions in cell proliferation, contact inhibition, and angiogenesis. We identified three novel splice variants of ING4 with differing activities in controlling cell proliferation, cell spreading, and cell migration. ING4_v1 (the longest splice variant), originally identified as ING4, encodes an intact nuclear localization signal (NLS), whereas the other three splice variants (ING4_v2, ING4_v3, and ING4_v4) lack the full NLS, resulting in increased cytoplasmic localization of these proteins. We found that one of the three ING4 variants, ING4_v2, is expressed at the same level as the original ING4 (ING4_v1), suggesting that ING4 variants may have significant biological functions. Growth suppressive effects of the variants that have a partial NLS (ING4_v2 and ING4_v4) were attenuated by a weaker effect of the variants on p21(WAF1) promoter activation. ING4_v4 lost cell spreading and migration suppressive effects; on the other hand, ING4_v2 retained a cell migration suppressive effect but lost a cell spreading suppressive effect. Therefore, ING4_v2, which localized primarily into cytoplasm, might have an important role in the regulation of cell migration. We also found that ING4_v4 played dominant-negative roles in the induction of p21(WAF1) promoter activation and in the suppression of cell motility by ING4_v1. In addition, ING4 variants had different binding affinities to two cytoplasmic proteins, protein-tyrosine phosphatase, receptor type, f polypeptide (PTPRF), interacting protein (liprin), alpha1, and G3BP2a. Tacrolimus is widely used as an immunosuppressant in liver transplantation, and tacrolimus-induced acute kidney injury (AKI) is a serious complication of liver transplantation. For early detection of AKI, various urinary biomarkers such as monocyte chemotactic protein-1, liver-type fatty acid-binding protein, interleukin-18, osteopontin, cystatin C, clusterin and neutrophil gelatinase-associated lipocalin (NGAL) have been identified. Here, we attempt to identify urinary biomarkers for the early detection of tacrolimus-induced AKI in liver transplant patients. Urine samples were collected from 31 patients after living-donor liver transplantation (LDLT). Twenty recipients developed tacrolimus-induced AKI. After the initiation of tacrolimus therapy, urine samples were collected on postoperative days 7, 14, and 21. In patients who experienced AKI during postoperative day 21, additional spot urine samples were collected on postoperative days 28, 35, 42, 49, and 58. The 8 healthy volunteers, whose renal and liver functions were normal, were asked to collect their blood and spot urine samples. The urinary levels of NGAL, monocyte chemotactic protein-1 and liver-type fatty acid-binding protein were significantly higher in patients with AKI than in those without, while those of interleukin-18, osteopontin, cystatin C and clusterin did not differ between the 2 groups. The area under the receiver operating characteristics curve of urinary NGAL was 0.876 (95% confidence interval, 0.800–0.951; P<0.0001), which was better than those of the other six urinary biomarkers. In addition, the urinary levels of NGAL at postoperative day 1 (p = 0.0446) and day 7 (p = 0.0006) can be a good predictive marker for tacrolimus-induced AKI within next 6 days, respectively. In conclusion, urinary NGAL is a sensitive biomarker for tacrolimus-induced AKI, and may help predict renal event caused by tacrolimus therapy in liver transplant patients. 相似文献

The effect of dietary polyunsaturated fatty acids and α-tocopherol supplementation on erythrocyte lipid peroxidation and immunocompetent cells in mice was studied comparatively using seven dietary oils (15% oil/diet, w/w) including fish oil rich in eicosapentaenoic acid (EPA, 20:5, n–3) and docosahexaenoic acid (DHA, 22:6, n–3). A 43% increase in spleen weight, about twice as many spleen cells and no change in the subpopulations of spleen cells, as well as a significant depression of mitogen-induced blastogenesis of both T and B cells in the spleen were observed in mice fed fish oil for 30 days in comparison with soybean oil diet-fed mice. In the fish oil diet-fed mice, membranous lipid hydroperoxide (hydroperoxides of phosphatidylcholine and phosphatidylethanolamine) accumulation as a marker of oxidative senescence in red blood cells (RBC) was 2.7–3.5 times higher than that in mice fed soybean oil, although there was no difference in the plasma phosphatidylcholine hydroperoxide concentration. In spite of the supplementation of α-tocopherol to up to 10 times the level in the basal diet, the degeneration of spleen cells and the stimulated oxidative senescence of RBC found by the fish oil feeding could not be prevented. The results suggest that oral intake of excess polyunsaturated fatty acids, i.e. EPA and DHA, in a fish oil diet can lead to acceleration of membrane lipid peroxidation resulting in RBC senescence linked to the lowering of immune response of spleen cells, and that supplementation of α-tocopherol as antioxidant does not always effectively prevent such oxidative degeneration as observed in spleen cells and RBC in vivo. 相似文献

利用斜纹夜蛾Spodoptera litura培养细胞,对近年来在日本本州、九州和四国等地发现并筛选出的对斜纹夜蛾幼虫具有强烈杀虫活性的3株斜纹夜蛾核型多角体病毒(SpltMNPV)(K-3、G1-2和G10-3)进行了生物学活性和分子生物学的初步研究,克隆了多角体蛋白基因,并进行了序列分析和比较。结果表明:(1)SpltMNPV日本分离株K-3、G1-2和G10-3分别具有不同的特征性酶切图谱,分别属于3种基因型(A型、B型和C型); (2)3个分离株的芽生型病毒(budded virus)产生能力和多角体产生能力有差异,免疫印迹分析表明,多角体蛋白的分子量也不同;(3)日本株SpltMNPV核型多角体蛋白结构基因由747个核苷酸编码序列(编码249个氨基酸)组成,其序列与中国株SpltMNPV的同源性为98.9%,与其他6种核型多角体病毒有较高的同源性(61.7%~74.2%),但其5′端侧翼序列(nt-1~-100)与AcMNPV和BmNPV相比差异显著,在对该基因表达调控起决定性作用的8个高度保守核苷酸序列中(nt-44~-51)有2处发生自然突变。 相似文献

Weak acid hypochlorous solution (WAHS) is known to have efficacy for inactivating
pathogens and to be relatively safe with respect to the live body. Based on these
advantages, many animal facilities have recently been introducing WAHS for daily cleaning
of animal houses. In this study, we determined the effect of WAHS in inactivating specific
pathogens of laboratory rodents and pathogens of opportunistic infection. WAHS with an
actual chloride concentration of 60 ppm and a pH value of 6.0 was generated using
purpose-built equipment. One volume of mouse hepatitis virus (MHV), Sendai virus,
lymphocytic choriomeningitis virus, Bordetella bronchiseptica,
Pasteurella pneumotropica, Corynebacterium kutscheri,
Staphylococcus aureus, and Pseudomonas aeruginosa was
mixed with 9 or 99 volumes of WAHS (×10 and ×100 reaction) for various periods (0.5, 1,
and 5 min) at 25°C. After incubation, the remaining infectious viruses and live bacteria
were determined by plaque assay or culture. In the ×100 reaction mixture, infectious
viruses and live bacteria could not be detected for any of the pathogens examined even
with the 0.5-min incubation. However, the effects for MHV, B.
bronchiseptica, and P. aeruginosa were variable in the ×10
reaction mixture with the 0.5- and 1-min incubations. Sufficient effects were obtained by
elongation of the reaction time to 5 min. In the case of MHV, reducing organic substances
in the virus stock resulted in the WAHS being completely effective. WAHS is recommended
for daily cleaning in animal facilities but should be used properly in order to obtain a
sufficient effect, which includes such things as using a large enough volume to reduce

In an early stage of development in murine embryos, axial rotation occurs and the body axis changes from a ventrally concave U-shape to a ventrally convex C-shape. In this study, axial rotation in Sprague-Dawley rat embryos occurred in about 5 h in vitro (from 27 h to 32 h in cultures of head-fold stage embryos). In sagittal sections, the somites in the mid-region of the body changed from a trapezoidal shape with a short dorsal side and long ventral side to the reverse trapezoidal shape with a long dorsal side and a short ventral side. The dorsal part of these somites acquired the ability to react with actin-specific antibody and developed into dermatome. On treatment with 0.1 μg/ml cytochalasin D during this 5 h period, embryos became ventrally concave with two lordosis bends. The somites in the bends had a short dorsal side, which did not show any evidence of dermatome or intense immunocytochemical staining. These results suggest that the increase in length of the dorsal side of the somites is a cause of the axial rotation and that the organization of actin filaments plays an important role in the conformational change of the somites. 相似文献

The ter (teratoma) mutation causes primordial germ cell (PGC) deficiency in ter/ter embryos at 9.5–12.5 days of post-coitum (dpc) in mouse strains 129/Sv- ter and LTXBJ- ter . To study the effects of the ter mutation on the PGC development more precisely, we examined the PGC number and distribution in 7.5–12.5 dpc embryo of ter congenic C57BL/6J- ter strain using their complete serial sections. The ter genotypes of embryos were identified by the polymerase chain reaction (PCR) polymorphisms of the microsatellite DNA of the Grl -1 locus mapped near the ter locus. Results showed that: (i) the PGC number in ter/ter embryos was similar to those of + / ter and + / + embryos at 7.5 dpc, and did not increase at 8.0–12.5 dpc, although those of normal littermates did usually; (ii) the PGC migration to genital ridges was never affected in all embryos; and (iii) the ter genotype difference in the PGC numbers was not recognized between + / ter and + / + embryos. We concluded that the ter mutation does not affect the PGC appearance around 7.5 dpc, but first causes PGC deficiency around 8.0 dpc at the beginning of their migration and proliferation, suggesting that the normal function of the ter gene may be essential for the proliferation or survival mechanisms of PGC. 相似文献

The spin traps substituted with some groups at the 4-position of dimethyl-1-pyrroline N-oxide(DMPO) were compared with DMPO itself regarding their abilities as spin traps and their physical properties. 4,5,5-Trimethyl-l-pyroHine N-oxide (4MDMPO) and 5,5-dimethyl- 4-phenyl-l-pyrolline N-oxide (4PDMPO) were synthesized by the Bonnett method, and 5,5-dimethyl-4-hydroxymethyl-l-pyrolline N-oxide (4HMDMPO) was made by a unique method from 2(5H)-furanone. The melting points of 4MDMPO, 4PDMPO and 4HMDMPO were higher than that of DMPO. The magnitude of hydrophilicity was in the order of 4HMDMPO, DMPO, 4MDMPO, and 4PDMPO based on the partition coefficient experiments in a 1-octanol-water system. Several radicals, O2, HO-, -CH3, -CH2OH, -CH(CH3)OH, (CH3)3 CO and H radicals, were trapped with these DMPO derivatives for comparison with the trapping by DMPO itself. Spin adducts of O J with the three DMPO derivatives showed ESR spectra similar to that of DMPO. In spite of the formation of diastereomers arising from spin trapping, the line-width enlargement was very small. The intensities and the decay rates of the spectra of 4MDMPO-O2-, 4PDMPO-O2- 4HMDMPO-O2- and DMPO-O2- were almost equal. In the trapping of the OH radical by 4MDMPO, 4PDMPO and 4HMDMPO, the eight-line ESR spectra observed were different from the well-known four-line spectrum of DMPO-OH. 相似文献
