Background:Staphylococcus aureus as a causative agent of hospital-acquired infections has been considered as the primary concern in biomaterial-related infections (BAIs).Methods:Following the purification of polysaccharide intercellular adhesion (PIA) as an efficient macromolecule in biofilm formation in the native condition, recombinant S. epidermidis surface-exposed rSesC protein, with the most homology to clumping factor A (ClfA) in S. aureus was cloned and expressed in a prokaryotic host as well. Fourier transform infrared spectrometry (FTIR) and Western blotting procedure analyzed purified PIA and protein, respectively. Then, the immune response was evaluated by measuring total IgG titers. Moreover, the capacity of Anti-biofilm forming activity of arisen antibodies to a biofilm-forming S. aureus strains was assessed by the semi-quantitative micro-plate procedure.Results:Data showed that the total IgGs were boosted in mice immunized sera. By performing an inhibition assay, the biofilm inhibitory effect of secreted antibodies to test strain was observed. Arisen antibodies against the mixture significantly were more potent than PIA and rSesC, when comparing individual antigens in a biofilm inhibition assay.Conclusion:immunization of mice with mentioned antigens especially a mixture of them, could eliminate the biofilm formation process in S. aureus. Hopefully, this study corresponds to the suggestion that the immunization of mice with PIA and rSesC candidate vaccines could protect against S. aureus infection.Key Words: PIA, Purification, Staphylococcus aureus, rSesC, Vaccine candidates  相似文献

A joint growth-carcass analysis was conducted to develop equations for predicting carcass quality traits associated with variation in growth path of crossbred cattle. During a four-year period (1994-1997) of the Australian "Southern Crossbreeding Project", mature Hereford cows (r = 581) were mated to 97 sires of Jersey, Wagyu, Angus, Hereford, South Devon, Limousin, and Belgian Blue breeds, resulting in 1141 calves. Data included body weight measurements of steers and heifers from birth until slaughter and four carcass quality traits: hot standard carcass weight, rump fat depth, rib eye muscle area, and intramuscular fat content. The model provides nine outputs: median and mean of carcass quality traits, predicted means, and lower and upper confidence intervals, as well as predicted intervals of carcass quality traits (95%) and economic values for domestic market and export markets. Input to the model consists of sex, sire breeds, age (in days)-weight (kg) pairs and slaughter age (500 days for heifer and 700 days for steers). The prediction model is able to accommodate different sexes across seven sire breeds and various management groups at any slaughter age. Its strength lies in its simplicity and flexibility, desirable to accommodate producers with different management schemes. In general, fat depth and intramuscular fat were found to be more affected by differences in growth rate than hot carcass weight and eye muscle area. Also, export market value was more sensitive to growth rate modifications than domestic market value. This model provides a tool by which the producer can estimate the impact of management decisions.  相似文献

Background

As a potent CD8+ T cell activator, peptide vaccine has found its way in vaccine development against intracellular infections and cancer, but not against leishmaniasis. The first step toward a peptide vaccine is epitope mapping of different proteins according to the most frequent HLA types in a population.

Methods and Findings

Six Leishmania (L.) major-related candidate antigens (CPB,CPC,LmsTI-1,TSA,LeIF and LPG-3) were screened for potential CD8+ T cell activating 9-mer epitopes presented by HLA-A*0201 (the most frequent HLA-A allele). Online software including SYFPEITHI, BIMAS, EpiJen, Rankpep, nHLApred, NetCTL and Multipred were used. Peptides were selected only if predicted by almost all programs, according to their predictive scores. Pan-A2 presentation of selected peptides was confirmed by NetMHCPan1.1. Selected peptides were pooled in four peptide groups and the immunogenicity was evaluated by in vitro stimulation and intracellular cytokine assay of PBMCs from HLA-A2+ individuals recovered from L. major. HLA-A2 individuals recovered from L. major and HLA-A2+ healthy donors were included as control groups. Individual response of HLA-A2+ recovered volunteers as percent of CD8+/IFN-γ+ T cells after in vitro stimulation against peptide pools II and IV was notably higher than that of HLA-A2 recovered individuals. Based on cutoff scores calculated from the response of HLA-A2 recovered individuals, 31.6% and 13.3% of HLA-A2+ recovered persons responded above cutoff in pools II and IV, respectively. ELISpot and ELISA results confirmed flow cytometry analysis. The response of HLA-A2 recovered individuals against peptide pools I and III was detected similar and even higher than HLA-A2+ recovered individuals.

Conclusion

Using in silico prediction we demonstrated specific response to LmsTI-1 (pool II) and LPG-3- (pool IV) related peptides specifically presented in HLA-A*0201 context. This is among the very few reports mapping L. major epitopes for human HLA types. Studies like this will speed up polytope vaccine idea towards leishmaniasis.  相似文献

This study investigated the potential of Persian shallot extract as an anticancer agent in HepG2 tumor cell line, an in vitro human hepatoma cancer model system. The inhibitory effect of Persian shallot on the growth of HepG2 cells was measured by MTT assay. To explore the underlying mechanism of cell growth inhibition of Persian shallot, the activity of Persian shallot in inducing apoptosis was investigated through the detection of annexin V signal by flow cytometry and expression of some apoptosis related genes such p21, p53, puma, caspase-8 family-Bcl-2 proteins like bid, bim, bcl-2 and bax were measured by real-time PCR in HepG2 cells. Persian shallot extract inhibited the growth of HepG2 cells in a dose-dependent manner. The IC50 value (inhibiting cell growth by 50%) was 149 μg/ml. The results of real-time PCR revealed a significant up-regulation of bid, bim, caspase-8, puma, p53, p21 and bax genes and a significant downregulation of bcl-2 gene in HepG2 cells treated with Persian shallot extract significantly. Therefore, this is the first report on an increased expression of bid, bim, caspase-8, puma, p53, p21 and bax genes and down regulation of bcl-2 gene indicating that the Persian shallot extract possibly induced the process of cell death through the intrinsic and extrinsic apoptosis pathways and triggers the programmed cell death in HepG2 tumor cell lines by modulating the expression of pro-/anti-apoptotic genes. Furthermore, we showed that Persian shallot extract increased annexin V signal and expression, resulting in apoptotic cell death of HepG2 cells after 24 h treatment. Therefore, according to the results of this study, the Persian shallot extract could be considered as a potential candidate for production of drug for the prevention or treatment of human hepatoma.  相似文献

Background

The goal of this work is to develop a non-invasive method in order to help detecting Alzheimer's disease in its early stages, by implementing voice analysis techniques based on machine learning algorithms.

Methods

We extract temporal and acoustical voice features (e.g. Jitter and Harmonics-to-Noise Ratio) from read speech of patients in Early Stage of Alzheimer's Disease (ES-AD), with Mild Cognitive Impairment (MCI), and from a Healthy Control (HC) group. Three classification methods are used to evaluate the efficiency of these features, namely kNN, SVM and decision Tree. To assess the effectiveness of this set of features, we compare them with two sets of feature parameters that are widely used in speech and speaker recognition applications. A two-stage feature selection process is conducted to optimize classification performance. For these experiments, the data samples of HC, ES-AD and MCI groups were collected at AP-HP Broca Hospital, in Paris.

Results

First, a wrapper feature selection method for each feature set is evaluated and the relevant features for each classifier are selected. By combining, for each classifier, the features selected from each initial set, we improve the classification accuracy by a relative gain of more than 30% for all classifiers. Then the same feature selection procedure is performed anew on the combination of selected feature sets, resulting in an additional significant improvement of classification accuracy.

Conclusion

The proposed method improved the classification accuracy for ES-AD, MCI and HC groups and promises the effectiveness of speech analysis and machine learning techniques to help detect pathological diseases.  相似文献

Theileria annulata, a protozoan parasite of cattle and domestic buffaloes, is transmitted by ticks of the genus Hyalomma, and causes a disease named Mediterranean or tropical theileriosis. In this research 50 cattle naturally infected with Theileria annulata were treated with the extract of the plant Peganum harmala. The treatment was continued for 5 days, the dose of the extract being 5 mg/kg per day. After the treatment, 39 cattle responded to the treatment and recovered, but 11 did not respond to the treatment and died. The recovery rate of animals treated with the extract of the plant Peganum harmala was 78%.  相似文献

Metabolic syndrome is known as a frequent precursor of type 2 diabetes mellitus (T2D). This disease could affect 8% of the people worldwide. Given that pancreatic β-cell dysfunction and loss have central roles in the initiation and progression of the disease, the understanding of cellular and molecular pathways associated with pancreatic β-cell dysfunction can provide more information about the underlying pathways involved in T2D. Multiple lines evidence indicated that oxidative stress, microRNA, and long noncoding RNA play significant roles in various steps of diseases. Oxidative stress is one of the important factors involved in T2D pathogenesis. This could affect the function and survival of the β cell via activation or inhibition of several processes and targets, such as receptor-signal transduction, enzyme activity, gene expression, ion channel transport, and apoptosis. Besides oxidative stress, microRNAs and noncoding RNAs have emerged as epigenetic regulators that could affect pancreatic β-cell dysfunction. These molecules exert their effects via targeting a variety of cellular and molecular pathways involved in T2D pathogenesis. Here, we summarized the molecular aspects of pancreatic β-cell dysfunction. Moreover, we highlighted the roles of oxidative stress, microRNAs, and noncoding RNAs in pancreatic β-cell dysfunction.  相似文献
