Malaria starts with the infection of the liver by Plasmodium sporozoites. This form of the parasite migrates through several host cells breaching their plasma membranes before infecting a final hepatocyte which they enter forming a parasitophorous vacuole. It is still controversial why Plasmodium sporozoites migrate through host cells. By reviewing the most recent literature, we hope to give an insight on the different steps of host invasion in which migration through cells is involved and on the possible role for this mechanism in infection. 相似文献

Upon entering their host, Plasmodium sporozoites travel directly to the liver. Once there, they migrate through several hepatocytes before they infect a final one. During migration, sporozoites breach the plasma membrane of traversed hepatocytes, but to infect they must form a parasitophorous vacuole, in which the intra-hepatic form of the parasite grows and multiplies. During this period there is a remarkable parasite multiplication, but little is known about the requirements and strategies that are developed to be successful. Hepatocyte growth factor and its receptor on hepatocytes might enhance early Plasmodium development within these cells. Malaria is transmitted through the bite of an infected mosquito, which introduces Plasmodium sporozoites into the mammalian host. Sporozoites rapidly reach the liver of the host where they are sequestered, a process probably mediated by circumsporozoite (CS) protein. Once in the liver, sporozoites migrate through several hepatocytes by breaching their plasma membranes before infecting a final hepatocyte with formation of a vacuole around the sporozoite, where development occurs into blood stage parasites. We propose that migration through several host cells activates sporozoites for ultimate productive invasion. This migration triggers sporozoite exocytosis, which is necessary for hepatocyte invasion, probably because it provides molecules, such as thrombospondin-related anonymous protein (TRAP), likely required for sporozoite invasion with the formation of a vacuole. How sporozoites migrate from the skin to the liver and invade hepatocytes remains unclear. Peptide-based vaccines aimed at the induction of effective T cell responses against established cancers have so far only met with limited clinical success and clearly need to be improved. In a preclinical model of human papillomavirus (HPV)16-induced cervical cancer we show that prime-boost vaccinations with the HPV16-derived 35 amino-acid long peptide E7(43-77), containing both a CTL epitope and a Th epitope, resulted in the induction of far more robust E7-specific CD8(+) T cell responses than vaccinations with the minimal CTL epitope only. We demonstrate that two distinct mechanisms are responsible for this effect. First, vaccinations with the long peptide lead to the generation of E7-specific CD4(+) Th cells. The level of the induced E7-specific CD8(+) T cell response proved to be dependent on the interactions of these Th cells with professional APC. Second, we demonstrate that vaccination with the long peptide and dendritic cell-activating agents resulted in a superior induction of E7-specific CD8(+) T cells, even when T cell help was excluded. This suggests that, due to its size, the long peptide was preferably endocytosed, processed, and presented by professional APCs. Moreover, the efficacy of this superior HPV-specific T cell induction was demonstrated in therapeutic prime-boost vaccinations in which the long peptide admixed with the dendritic cell-activating adjuvant oligodeoxynucleotide-CpG resulted in the eradication of large, established HPV16-expressing tumors. Because the vaccine types used in this study are easy to prepare under good manufacturing practice conditions and are safe to administer to humans, these data provide important information for future clinical trials. 相似文献

The conventional random pore model assumes a homogeneous cell distribution in the gel matrix used to immobilize cells. However, the validity of this model is restricted to values of the exponent alpha, between 1.8 and 2.25, of a model power function relating the diffusivity coefficient in the matrix with the overall cell volume fraction in the system. Based on the analysis of published data for diffusion in gels with immobilized cells and on the homogeneous approach for the random pore model developed in a previous work, a new, nonhomogeneous approach is proposed for alpha values outside the range 1.8-2.25. To explain these data, two main types of nonhomogeneous cell distribution were considered: (1) nonhomogeneous cell distribution in the gel for alpha > 2.25 (type 1) and (2) nonhomogeneity related with anisotropy of cell space orientation when alpha < 1.8 (type 2). In the case of nonhomogeneity of type 1, the cell volume fraction in the layers occupied by cells must be considered in place of the concept previously used for homogeneous distribution, viz., the average cell volume fraction. This model underlines that accumulation of cells in a thin layer close to the surface improves their nutrient intake. For nonhomogeneity of type 2, the tortuosity of such a system is smaller than should be expected if spherical cells were considered, thereby changing the effective diffusion. The purpose of this work was to evaluate the response of the antioxidant system of goldfish Carassius auratus during anoxia and reoxygenation. The exposure of goldfish to 8 h of anoxia induced a 14% decrease in total glutathione levels in the kidney, although the liver, brain, and muscle were unaffected. Anoxia also resulted in increases in the activities of liver catalase, brain glucose-6-phosphate dehydrogenase, and brain glutathione peroxidase (by 38, 26, and 79%, respectively) and a decrease in kidney catalase activity (by 17.5%). After 14 h of reoxygenation, liver catalase and brain glutathione peroxidase activities remained higher than controls and several other tissue-specific changes occurred in enzyme activities. Superoxide dismutase activity was unaffected by anoxia and reoxygenation. The levels of conjugated dienes, as indicators of lipid peroxidation, increased by 114% in liver after 1 h of reoxygenation and by 75% in brain after 14 h of reoxygenation. Lipid peroxidation was unaffected in kidney and depressed during anoxia and reoxygenation (by 44-61%) in muscle. Regulation of the goldfish antioxidant system during anoxia may constitute a biochemical mechanism that minimizes oxidative stress following reoxygenation. 相似文献

Variation in STR loci of the human myelin basic protein gene: north Portugal and São Tomé e Príncipe

Allele frequencies and a single-base substitution polymorphism for 3 short tandem repeat (STR) loci of the human myelin basic protein (MBP) gene were evaluated in North Portugal and S?o Tomé e Príncipe. Strong linkage disequilibrium between loci MBPB and MBPC was found. However, the patterns of nonrandom allelic associations were very different in the 2 populations: levels of haplotypic diversity and heterozygosity were higher in the S?o Tomé population. Similarly, a difference in the frequency of base substitution G-->A at position 124 was found: the frequency reached 4.1% in North Portugal and 0.5% in S?o Tomé. In both populations it was always found to be associated with haplotypes B10/C11 and B12/C9. 相似文献

Silica gel, aluminium oxide, diatomaceous earth, polyamide, cyano, diol and amino plates have been tested for their capacity to separate the color pigments of six chili powders of different origin by both adsorption and reversed-phase thin-layer chromatography. The plates were evaluated at 340 and 440 nm wavelengths. Best separation of color pigments was obtained on impregnated diatomaceous earth layer using acetone-water 17:3 v/v eluent. It was found that the pigment composition of chili powders showed marked differences. Principal component analysis employed for the classification of the chili powders according to their pigment composition indicated that these differences can be used for the determination of the similarity or dissimilarity of the chili powders. 相似文献

The first results are presented of an aerobiological analysis of the atmosphere of the town of Almería, carried out between
November 1995 and October 1996. A Lanzoni volumetric spore trap was used for sample collection. The composition and seasonal
evolution of the pollen spectrum were determined over a 1-year period in relation to the vegetation and climatic conditions
of the study area. Twenty-six pollen types were identified as accounting for >0.05% of the total pollen collected. The main
sources of airborne pollen were Palmae (17.76%),Olea (16.10%), Chenopodiaceae/Amaranthaceae (13.99%), Urticaceae (10.18%) and Poaceae (8.64%). The annual pollen variation presented
a period of maximum emission from March to June, with a subsequent, less intensive period from August to November. The minimum
pollen values were obtained from December to February. The highest concentrations occurred in May, which was also the month

The effects of monovalent and divalent cations on the rheological behavior of Halomonas eurihalina exopolysaccharide (EPS) were studied. Sodium, potassium, magnesium and calcium were added and the relative abilities to increase
viscosity were as follows: KCl > NaCl > MgCl2 > CaCl2. The highest viscosity value was measured in acidic 10−4 M KCl, in which a gel formed. A loss of sulfate content seemed to correlate with the increase of viscosity. H. eurihalina produced EPS in all growth media. Addition of hydrophobic substrates to culture media produced changes in chemical
composition and emulsifying activity of the EPS. Xylene was the most effectively emulsified substance and the EPS produced
on tetradecane and on corn oil the most active emulsifier.
