Natural selection should favor females that avoid ovipositing where risk of predation is high for their progeny. Despite the large consequences of such oviposition behavior for individual fitness, population dynamics, and community structure, relatively few studies have tested for this behavior. Moreover, these studies have rarely assessed the mode of detection of predators, compared responses in prey species that vary in vulnerability to predators, or tested for the behavior in natural habitats. In an outdoor artificial pool experiment, we tested the oviposition responses of two dipteran species, Culiseta longiareolata (mosquito) and Chironomus riparius (midge), to the hemipteran predator, Notonecta maculata. Both dipteran species have similar life history characteristics, but Culiseta longiareolata larvae are highly vulnerable to predation by Notonecta, while Chironomus riparius larvae are not. As their vulnerabilities would suggest, Culiseta longiareolata, but not Chironomus riparius, strongly avoided ovipositing in pools containing Notonecta. An experiment in natural rock pools assessing oviposition by Culiseta longiareolata in response to Notonecta maculata yielded an oviposition pattern highly consistent with that of the artificial pool experiment. We also demonstrated that the cue for oviposition avoidance by Culiseta longiareolata was a predator-released chemical: Notonecta water (without Notonecta replenishment) repelled oviposition for 8 days. Oviposition avoidance and mode of detection of the predator have important implications for how to assess the true impact of predators and for the use of commercially produced kairomones for mosquito control.  相似文献

During a period of four years, multidrug-resistant Escherichia coli of particular serotypes have been isolated from 255 elderly (>65 years) patients from four hospitals in central Israel. 83% of the isolates belonged to one of four predominant serotypes (O153:H31, O101:H-, O2:H42, and O102:H6). All isolates were producers of extended spectrum beta-lactamases and resistant to ciprofloxacin. To our knowledge, the involved serotypes have hitherto not been reported as etiological agents of extraintestinal human infections (MEDLINE). Pulsed-field gel electrophoresis of isolates from one of the most frequent serotypes indicated a clonal relationship between them. Further investigation of these strains and analysis of their virulence factors may help to confine their spread.  相似文献

Interleukin-6 (IL-6) is a pleiotropic inflammatory cytokine, also endowed with inflammation-inhibiting properties. The status of interleukin-10 (IL-10) as an anti-inflammatory cytokine is more solidly established. The roles of IL-6 and IL-10 in the context of organ physiology, and their possible modulation by the aging process, are not satisfactorily understood. The purpose of this work was to characterize organ IL-6 and IL-10 expression in different cellular compartments in mice, under steady-state and stress conditions. The former was evaluated by immunohistochemistry (IHC) and enzyme-linked immunosorbent assay (ELISA) analyses of organ lysates (LYS) (addressing the intracellular compartment), while the latter was assessed by ELISA analyses of organ-conditioned media (CM), obtained after 48 hrs of organ culturing (addressing the potential of cytokine secretion/diffusion). Under steady-state conditions, the overall level of IL-6 and IL-10 expression was relatively low in both age groups (exceptionally, IHC staining demonstrated an enhanced expression of these cytokines in the heart, skeletal muscle and brain of young mice). Much more elevated levels of IL-6 and IL-10 expression were demonstrated in organ CM, possibly emphasizing the role of these cytokines in the context of organ stress. This was most characteristically shown in the highly specialized organs (heart, skeletal muscle and kidney) and liver of old mice, as compared with the other lymphoreticular organs (lungs, spleen, small intestine) tested. Thus, IL-10 was markedly upregulated in the highly specialized organs, while IL-6 was considerably reduced in the lymphoreticular organs. In addition, aging appears to be associated with altered patterns of intracellular expression and secretion/diffusion potentials of IL-6 and IL-10 in the heart and skeletal muscle, as demonstrated by reduced IHC staining on one hand, and an increased detection in organ CM, on the other. These findings may contribute to a better understanding of the unique functions of organ IL-6 and IL-10 in various age groups, and suggest an important role in organ response to stress in old age.  相似文献

Carrot cells (Daucus carota L.) in suspension culture exposed to medium containing 150 mM NaCl plasmolyzed immediately and deplasmolyzed within 35 to 40 hr. Three days after exposure to NaCl the cells resumed proliferation. Accommodation to salinity and renewal of growth was accompanied by absorption of Na+ from the external medium. On completion of deplasmolysis, K+ concentration in the cytosol doubled and Na+ concentration approximated that of K+. The vacuolar K+ concentration was practically unchanged while Na+ accumulated to a concentration double that of K+. Cl−- accumulation started later and eventually exceeded that of Na+ plus K+. Malate was redistributed during accommodation to salinity and eventually returned to its initial level. Amino acid content in the cytosol increased fivefold, while in the vacuole it remained unchanged. These results show that: 1) recovery from osmotic shock requires absorption of easily penetrating solute, mainly Na+; 2) distribution of solutes, absorbed or synthesized in cells exposed to salinity, is a dynamic process; 3) cells could grow and proliferate in high NaCl content in the cytosol; 4) red beet root cells grown in the presence of NaCl contain higher cytoplasmic Na+ than K+; and 5) during adjustment to salinity small spherical carrot cells survive the osmotic shock and do not show any detectable damage.  相似文献

Plants of the genus Pistacia (Anacardiaceae) serve as obligate hosts for a group of specialized gall-forming aphids (Homoptera: Fordinae). The aphids regularly migrate between the Pistacia (primary) host plants and the roots of non-specific grasses and cereals (secondary hosts). Gall density varies considerably between trees and sites. The intimate relationships between the aphids and their primary host, the natural variation of host susceptibility, and the heterogeneous geographical environment may promote local adaptation and deme formation in the aphid populations. Indeed, previous analyses of the genetic structure of the aphid Baizongia pistaciae, which forms large galls on the deciduous P. palaestina trees, suggested deme formation (Martinez et al. 2005). In this study, we analyzed the genetic structure of the B. pistaciae population at eight sites and 78 trees throughout Israel and a single population in Turkey, using two molecular markers (AFLP fingerprints and COI sequencing). The genetic distance between the Israeli populations was found to be low (D = 0.01–0.02), and there was no genetic differentiation found between any population pairs. In five of the Israeli populations, we also compared the genetic identity between aphids forming galls on the same tree and between galls on neighboring trees. The analysis indicated that the genetic identity of different galls within a tree resembles the correspondence between trees within a population. Our results showed no indication of deme formation or any hierarchical genetic substructuring within B. pistaciae populations in Israel. The extensive gene flow between aphid colonies may be explained by their dispersal abilities and the potential bridging role of the secondary hosts.  相似文献

The involvement of genotype, family group and sex in the regulation of the response to young was studied in non-lactating wild versus laboratory house mice (Mus musculus). Regardless of the sex, naive wild mice typically killed young, in contrast to laboratory mice, which characteristically exhibited parental care. After cohabiting with parents attending their offspring, wild males exhibited paternal behaviour, while the majority of wild virgin females continued to display infanticide. Contrary to the prevailing view, the results indicate that (a) parental behaviour in wild mice does not occur spontaneously, and (b) the domesticated mouse may not always reliably represent the species Mus musculus in behavioural studies.  相似文献

We modified a series of (N)-methanocarba nucleoside 5′-uronamides to contain dialkyne groups on an extended adenine C2 substituent, as synthetic intermediates leading to potent and selective A3 adenosine receptor (AR) agonists. The proximal alkyne was intended to promote receptor recognition, and the distal alkyne reacted with azides to form triazole derivatives (click cycloaddition). Click chemistry was utilized to couple an octadiynyl A3AR agonist to azido-containing fluorescent, chemically reactive, biotinylated, and other moieties with retention of selective binding to the A3AR. A bifunctional thiol-reactive crosslinking reagent was introduced. The most potent and selective novel compound was a 1-adamantyl derivative (Ki 6.5 nM), although some of the click products had Ki values in the range of 200–400 nM. Other potent, selective derivatives (Ki at A3AR in nM) were intended as possible receptor affinity labels: 3-nitro-4-fluorophenyl (10.6), α-bromophenacyl (9.6), thiol-reactive isothiazolone (102), and arylisothiocyanate (37.5) derivatives. The maximal functional effects in inhibition of forskolin-stimulated cAMP were measured, indicating that this class of click adducts varied from partial to full A3AR agonist compared to other widely used agonists. Thus, this strategy provides a general chemical approach to linking potent and selective A3AR agonists to reporter groups of diverse structure and to carrier moieties.  相似文献

The stereoselective binding and transformation of optically pure bicyclic alcohols by human UDP-glucuronosyltransferases from subfamily 2B were investigated. The enantiomers of 1-indanol, 1-tetralol, and 1-benzosuberol were synthesized by asymmetric Corey-Bakshi-Shibata reduction and subjected to glucuronidation assays. The alcohols studied were primarily glucuronidated by UGT2B7 and UGT2B17. The catalytic transformation by UGT2B17 was highly stereoselective, favoring conjugation of the (R)-enantiomers. UGT2B7, on the other hand, did not exhibit stereoselectivity toward 1-benzosuberol, the best substrate in this series. To assess binding affinities to the enzymes, the six different compounds were tested for their efficiency as inhibitors of either UGT2B7 or UGT2B17. The results of the latter analyses indicated that the affinities of both enantiomers of each pair towards UGT2B7 and UGT2B17 were of the same order of magnitude. Therefore, the findings of this study suggest that the spatial arrangement of the hydroxy group plays an important role in the glucuronic acid transfer reaction, but not necessarily in substrate binding to the UGTs.  相似文献

Methotrexate (MTX) exerts an anti-inflammatory effect via its metabolite adenosine, which activates adenosine receptors. The A3 adenosine receptor (A3AR) was found to be highly expressed in inflammatory tissues and peripheral blood mononuclear cells (PBMCs) of rats with adjuvant-induced arthritis (AIA). CF101 (IB-MECA), an A3AR agonist, was previously found to inhibit the clinical and pathological manifestations of AIA. The aim of the present study was to examine the effect of MTX on A3AR expression level and the efficacy of combined treatment with CF101 and MTX in AIA rats. AIA rats were treated with MTX, CF101, or both agents combined. A3AR mRNA, protein expression and exhibition were tested in paw and PBMC extracts from AIA rats utilizing immunohistochemistry staining, RT-PCR and Western blot analysis. A3AR level was tested in PBMC extracts from patients chronically treated with MTX and healthy individuals. The effect of CF101, MTX and combined treatment on A3AR expression level was also tested in PHA-stimulated PBMCs from healthy individuals and from MTX-treated patients with rheumatoid arthritis (RA). Combined treatment with CF101 and MTX resulted in an additive anti-inflammatory effect in AIA rats. MTX induced A2AAR and A3AR over-expression in paw cells from treated animals. Moreover, increased A3AR expression level was detected in PBMCs from MTX-treated RA patients compared with cells from healthy individuals. MTX also increased the protein expression level of PHA-stimulated PBMCs from healthy individuals. The increase in A3AR level was counteracted in vitro by adenosine deaminase and mimicked in vivo by dipyridamole, demonstrating that receptor over-expression was mediated by adenosine. In conclusion, the data presented here indicate that MTX induces increased A3AR expression and exhibition, thereby potentiating the inhibitory effect of CF101 and supporting combined use of these drugs to treat RA.  相似文献
