Phagocytosis of extracellular organisms in the alveolar spaces of the lungs represents the first-line of host defense against pulmonary pathogens. Disruption of this process is likely to interfere with the generation of appropriate specific immune responses, and lead to a delayed or inefficient clearance of the pathogen. Pneumocystis carinii, an opportunistic pathogen in immunodeficient individuals, is cleared from the lung by alveolar macrophages. In the absence of specific anti-Pneumocystis antibodies, phagocytosis is dependent on the non-opsonic macrophage mannose receptor (MR). Recent studies have demonstrated that alveolar macrophage MR activity is downregulated in individuals infected with HIV, and that functional MR is shed from the macrophage cell surface. Here we report that P. carinii enhances the formation of soluble MR by macrophages in vitro. Soluble MR was detected in cell-free alveolar fluid from humans infected with HIV and/or P. carinii, but not in alveolar fluid from healthy controls. Soluble MR was found in association with extracellular clumps of P. carinii in the lungs of mice with P. carinii pneumonia, and was associated with P. carinii organisms purified from these mice. When purified P. carinii organisms were incubated with soluble MR-containing supernatants, they were phagocytosed less readily by alveolar macrophages than were control organisms. Our results suggest that P. carinii organisms enhance the shedding of MR from the surface of alveolar macrophages, and that the resultant soluble MR binds to intra-alveolar organisms, thereby interfering with their non-opsonic uptake via the macrophage cell surface MR. 相似文献

Benthic invertebrates affect microbial processes and communities in freshwater sediment by enhancing sediment-water solute fluxes and by grazing on bacteria. Using microcosms, the effects of larvae of the widespread midge Chironomus plumosus on the efflux of denitrification products (N2O and N2 + N2O) and the diversity and abundance of nitrate- and nitrous-oxide-reducing bacteria were investigated. Additionally, the diversity of actively nitrate- and nitrous-oxide-reducing bacteria was analyzed in the larval gut. The presence of larvae increased the total effluxes of N2O and N2 + N2O up to 8.6- and 4.2-fold, respectively, which was mostly due to stimulation of sedimentary denitrification; incomplete denitrification in the guts accounted for up to 20% of the N2O efflux. Phylotype richness of the nitrate reductase gene narG was significantly higher in sediment with than without larvae. In the gut, 47 narG phylotypes were found expressed, which may contribute to higher phylotype richness in colonized sediment. In contrast, phylotype richness of the nitrous oxide reductase gene nosZ was unaffected by the presence of larvae and very few nosZ phylotypes were expressed in the gut. Gene abundance of neither narG, nor nosZ was different in sediments with and without larvae. Hence, C. plumosus increases activity and diversity, but not overall abundance of nitrate-reducing bacteria, probably by providing additional ecological niches in its burrow and gut. 相似文献

We have measured the uptake capacity of nitrogen (N) and potassium (K) from different soil depths by injecting 15N and caesium (Cs; as an analogue to K) at 5 and 50 cm soil depth and analysing the recovery of these markers in foliage and buds. The study was performed in monocultures of 40-year-old pedunculate oak (Quercus robur), European beech (Fagus sylvatica) and Norway spruce (Picea abies (L.) Karst.) located at an experimental site in Palsgård, Denmark. The markers were injected as a solution through plastic tubes around 20 trees of each species at either 5 or 50 cm soil depth in June 2003. After 65 days foliage and buds were harvested and the concentrations of 15N and Cs analysed. The recovery of 15N in the foliage and buds tended to be higher from 5 than 50 cm soil depth in oak whereas they where similar in spruce and beech after compensation for differences in immobilization of 15N in the soil. In oak more Cs was recovered from 5 than from 50 cm soil depth whereas in beech and spruce no difference could be detected. Out of the three investigated tree species, oak was found to have the lowest capacity to take up Cs at 50 cm soil depth compared to 5 cm soil depth also after compensating for differences in discrimination against Cs by the roots. The uptake capacity from 50 cm soil depth compared with 5 cm was higher than expected from the root distribution except for K in oak, which can probably be explained by a considerable overlap of the uptake zones around the roots and mycorrhizal hyphae in the topsoil. The study also shows that fine roots at different soil depths with different physiological properties can influence the nutrient uptake of trees. Estimates of fine root distribution alone may thus not reflect the nutrient uptake capacity of trees with sufficient accuracy. Our study shows that deep-rooted trees such as oak may have lower nutrient uptake capacity at deeper soil layers than more shallow-rooted trees such as spruce, as we found no evidence that deep-rooted trees obtained proportionally more nutrients from deeper soil layers. This has implications for models of nutrient cycling in forest ecosystems that use the distribution of roots as the sole criterion for predicting uptake of nutrients from different soil depths. 相似文献

Recent research communications indicate that the adult human brain contains undifferentiated, multipotent precursors or neural stem cells. It is not known, however, whether these cells can develop into fully functional neurons. We cultured cells from the adult human ventricular wall as neurospheres and passed them at the individual cell level to secondary neurospheres. Following dissociation and plating, the cells developed the antigen profile of the three main cell types in the brain (GFAP, astrocytes; O2, oligodendrocytes; and beta-III-tubulin/NeuN, neurons). More importantly, the cells developed the electrophysiological profiles of neurons and glia. Over a period of 3 weeks, neuron-like cells went through the same phases as neurons do during development in vivo, including up-regulation of inward Na+ -currents, drop in input resistance, shortening of the action potential, and hyperpolarization of the cell membrane. The cells developed overshooting action potentials with a mature configuration. Recordings in voltage-clamp mode displayed both the fast inactivating TTX-sensitive sodium current (INa) underlying the rising phase of the action potential and the two potassium currents terminating the action potential in mature neurons (IA and IK, sensitive to 4-AP and TEA, respectively). We describe a physical simulation of natural selection in a population of legorgs, six-segment model organisms. Legorg morphology
is genetically specified by five alleles on each segment. Legorgs show a simple form of motility that could evolve in originally
sessile animals. This motility, the ability to move horizontally on a smooth surface, depends on the morphology and interaction
of the six segments that produce different patterns of locomotion. Legorgs are selected for motility and reproduce in proportion
to fitness. After just five generations, the average population motility increases 2.5 times. Additionally, we describe a
slightly less time-consuming simulation of legorg evolution, where fitness is assigned by comparison with a template. The
calculation of gene pools is precisely the same as in the previous simulation and produces very robust increases in fitness
during five generations. The simulation is designed as a classroom experiment to explore the mechanism of natural selection.
A test of its learning efficiency by evaluating the students’ conception of central aspects of evolutionary theory before
and after showed a significant improvement. The surprising power of natural selection in this very simple physical system
