Breeding at the right time is essential for animals in seasonal climates in order to ensure that the energy demands of reproduction, particularly the nutritional requirements of growing young, coincide with peak food availability. Global climate change is likely to cause shifts in the timing of peak food availability, and in order to adapt successfully to current and future climate change, animals need to be able to adjust the time at which they initiate breeding. Many animals use environmental cues available before the breeding season to predict the seasonal peak in food availability and adjust their phenology accordingly. We tested the hypothesis that regulation of breeding onset should reflect the scale at which organisms perceive their environment by comparing phenology of three seabird species at a North Sea colony. As predicted, the phenology of two dispersive species, black-legged kittiwake ( Rissa tridactyla ) and common guillemot ( Uria aalge ), correlated with a large-scale environmental cue (the North Atlantic Oscillation), whereas a resident species, European shag ( Phalacrocorax aristotelis ), was more affected by local conditions (sea surface temperature) around the colony. Annual mean breeding success was lower in late years for European shags, but not for the other two species. Since correlations among climate patterns at different scales are likely to change in the future, these findings have important implications for how migratory animals can respond to future climate change.  相似文献

Medium-chain acyl-CoA dehydrogenase (MCAD) is a homotetrameric flavoprotein which catalyses the initial step of the beta-oxidation of medium-chain fatty acids. Mutations in MCAD may cause disease in humans. A Y42H mutation is frequently found in babies identified by newborn screening with MS/MS, yet there are no reports of patients presenting clinically with this mutation. As a basis for judging its potential consequences we have examined the protein phenotype of the Y42H mutation and the common disease-associated K304E mutation. Our studies of the intracellular biogenesis of the variant proteins at different temperatures in isolated mitochondria after in vitro translation, together with studies of cultured patient cells, indicated that steady-state levels of the Y42H variant in comparison to wild-type were decreased at higher temperature though to a lesser extent than for the K304E variant. To distinguish between effects of temperature on folding/assembly and the stability of the native enzyme, the thermal stability of the variant proteins was studied after expression and purification by dye affinity chromatography. This showed that, compared with the wild-type enzyme, the thermostability of the Y42H variant was decreased, but not to the same degree as that of the K304E variant. Substrate binding, interaction with the natural electron acceptor, and the binding of the prosthetic group, FAD, were only slightly affected by the Y42H mutation. Our study suggests that Y42H is a temperature sensitive mutation, which is mild at low temperatures, but may have deleterious effects at increased temperatures.  相似文献

The aim of the study was to standardize and evaluate technically optimized food allergen extracts for use in skin prick test (SPT). The standardization procedure comprised 36 allergic histories in 32 food allergic patients with 21 healthy, non-atopic individuals serving as controls. The patients had a history of allergic symptoms upon ingestion of either cow’s milk (n=3), hen’s egg (n=9), wheat (n=4), hazelnut (n=14) or cod (n=6). They also had specific IgE in serum to the food in question and a positive SPT with a fresh preparation of the food. The diagnosis had been confirmed by a double-blind, placebo-controlled food challenge, except for the hazelnut-allergic patients. The controls were subjected to an open food challenge with all the foods to ensure tolerance. The standardization was performed by means of titrated SPT in accordance with the guidelines on biological standardization from the Nordic Council on Medicine. Regression analysis of the skin wheal areas was performed for each patient and the median protein concentration of allergen preparation (median Ch10) eliciting a wheal area of the same size as histamine 10 mg/ml was calculated. The median Ch10 was 0.56 mg/ml for milk, 0.88 mg/ml for egg, 5.4 mg/ml for wheat, 2.1 mg/ml for hazelnut and 0.017 mg/ml for the cod extract. The sensitivity of the median Ch10 estimated from the SPT data was 1 for milk, 0.98 for egg, 1 for wheat, 1 for hazelnut and 0.87 for the cod extract. The allergenic activity of the hazelnut extract was further investigated by leukocyte histamine release (HR) and immunoblotting experiments using sera from 27 hazelnut allergic patients. The clinical sensitivity of the optimized hazelnut extract evaluated by HR was 0.78 compared to 0.30 for a commercially available hazelnut extract (Soluprick). Immunoblotting results showed a stronger IgE binding capacity and additional IgE-binding bands of the optimized hazelnut extract compared with the Soluprick extract.  相似文献

An important issue in the management of zebra mussel (Dreissena polymorpha) populations is early, rapid, and accurate detection of the planktonic larvae (veliger) of the zebra mussel. The goal of this study was to explore the feasibility of developing a molecular approach for the detection of zebra mussel larvae in diverse environments. In this study a Dreissena polymorpha-specific 18S ribosomal RNA gene targeted oligonucleotide primer (ZEB-715a) and Polymerase Chain Reaction (PCR) assay was developed and compared with cross-polarized microscopy as a means to detect zebra mussel veligers in plankton samples. The design of the zebra mussel-specific primer was facilitated by sequencing nearly the complete 18S rRNA gene from the zebra mussel and three other closely related freshwater Veneroids including the quagga mussel (D. bugensis), the dark false mussel (Mytilopsis leucophaeata), and the Asian freshwater clam (Corbicula fluminea). The specificity of the primer for the zebra mussel was empirically tested by using the primer as a direct probe in a blot hybridization format. A single veliger in a plankton sample could be detected by PCR using this approach. Veliger detection sensitivity using the PCR approach was estimated to be over 300 times more sensitive than cross-polarized light microscopy based techniques. Cross-polarized light microscopy and the PCR technique were used to identify the presence of zebra mussel larvae in plankton samples that were collected from a variety of natural and industrial water sources. Detection results (presence or absence) were generally consistent between the two methods. Although additional studies will be required before routine application of molecular based veliger detection technology is available, a long-term goal of this work is the application of molecular technology to the development of a field device for the routine detection and quantification of zebra mussel veligers.  相似文献

Confined within a volcanic caldera at 2000 m a.s.l., the sub-alpine desert of Tenerife, Canary Islands, harbors a distinct biota. At this altitude the climate is harsh and the growing season short. Hence, plant and animal communities, constituting the sub-alpine plant–flower-visitor network, are clearly delimited, both spatially and temporally. We investigated species composition and interaction structure of this system. A total of 11 plant species (91% endemics) and 37 flower-visiting animal species (62% endemics) formed 108 interactions. Numbers of interactions among species varied ten-fold within both plant and animal communities. Generalization level of a species was positively correlated with its local abundance. Two separate network analyses revealed a significantly nested structure. In relation to a plant–flower-visitor system, nestedness implies that specialized species (animals or plants) interact with a subset of the species pool visiting (animals) or being visited (plants) by more generalized species. Therefore, specialized, locally rare plants tend to be visited by generalized, locally abundant animals, and specialized, locally rare animals tend to utilize generalized, locally abundant food plants. Such patterns could have implications for conservation of the sub-alpine network, and stress the importance of preserving not only rare species, but also the more abundant ones, which may be key food resources or pollinators in the plant–flower-visitor network.  相似文献

The hypothesis was tested that acute water immersion to the neck (WI) compared with 6 degrees head-down tilt (HDT) induces a more pronounced distension of the heart and lower plasma levels of vasoconstrictor hormones. Ten healthy males underwent 30 min of HDT, WI, and a seated control (randomized). During WI, left atrial diameter and stroke volume increased to the same extent as during HDT. Cardiac output increased by 1 l/min more during WI than during HDT. (P < 0.05). Plasma atrial natriuretic peptide increased during WI (P < 0.05) but not during HDT, whereas plasma norepinephrine, vasopressin, and renin activity were suppressed similarly. Mean arterial pressure decreased by 9 mmHg (P < 0.05) during HDT and was unchanged during WI, and heart rate decreased more during HDT (P < 0.05). Arterial pulse pressure increased considerably more during HDT than during WI. In conclusion, the hypothesis was not confirmed because the cardiac atria were similarly distended by acute HDT and WI and the release of vasoconstrictor hormones were suppressed to the same extent.  相似文献

The paper provides an overview of approaches to dioxin risk assessment employed by different agencies worldwide over the past 20 years. Our insights regarding understanding of the toxicity of dioxins have advanced tremendously in recent years; however, important data gaps still exist. More information on topics such as mechanism of interaction, effects at low levels of exposure, interspecies differences, and sensitive populations is needed. Some differences exist between USEPA's approach to dioxin assessment and that of other health organizations around the world. The authors conclude that USEPA's reassessment of dioxin and related compounds may place too much confidence in the ability to accurately predict cancer risks at low doses. Further, it is important to derive health-based guidance values for noncancer end points especially in accordance with emerging reports that reproductive and developmental end points are very sensitive to dioxins. A worldwide convergence on the health assessment value being around 1 to 4?pg/kg/day is noted.  相似文献

A series of NN703 analogues with lysine mimetics combined with naphthyl- or biphenylalanine in the core has been prepared and tested in vitro in a rat pituitary cell based assay and subsequently in vivo in pigs in a single dose at 50 nmol/kg. Re-introduction of certain pharmacophores in the C-terminal of NN703, which were originally removed during optimisation for oral bioavailability, led to unexpectedly potent compounds in vitro as well as in vivo.  相似文献

Summary Recombinant protein G (RPG) was conjugated to colloidal gold particles and used for immunocytochemistry. In this report, the preparation of RPG—gold conjugates (RPGG) and the application of these conjugates in spot blot tests and in double immunolabelling are described. The immunolabelling was performed on ultracryosections of pig small intestine using antibodies directed against aminopeptidase N and sucrase—isomaltase. The labelling efficiency of RPGG was compared to that of protein A—gold conjugates (PAG) in different compartments of the enterocyte. Quantification showed that the labelling intensity was dependent on the size of the marker as well as on the kind of protein used for complex formation. The distributions for RPGG and PAG were respectively: for the 12nm particles, 10.3 and 6.2 particles/µm of length of microvillar membrane, 3.5 and 1.0 particles/µm2 of Golgi profile and 5.9 and 2.0 particles/µm2 of multivesicular body profile; and for the 6nm particles, 49.6 and 15.7 particles/µm of length of microvillar membrane, 24.4 and 5.0 particles/µm2 of Golgi profile and 25.4 and 3.4. particles/µm2 of multivesicular body profile. Controls showed very little non-specific gold labelling (<0.02 gold particles/µm2 of section).  相似文献
