The timing of ovulation relative to the onset of oestrus and the preovulatory surge in luteinizing hormone (LH) was studied in red deer following treatments to synchronize oestrus and induce either a monovulatory or superovulatory response. Mature hinds (n = 36) were allocated randomly to two mating groups (n = 16 + 20), with respective treatments staggered by 4 weeks during the 1990 rut (March-April). Each hind was treated with an intravaginal controlled internal drug releasing (CIDR)-type S device for 14 days. Treatments to induce a monovulatory response included CIDR device alone (treatment A; n = 4 + 8) and additional injection of 200 iu pregnant mares' serum gonadotrophin (PMSG) at device removal (treatment B; n = 4 + 4). Treatments to induce a superovulatory response included injections of 200 iu PMSG and 0.5 units ovine follicle-stimulating hormone (FSH) at about time of removal of CIDR devices (treatment C; n = 4 + 4) and further treatment with gonadotrophin-releasing hormone (GnRH) analogue 18 h after removal of CIDR devices (treatment D; n = 4 + 4). The hinds were run with crayon-harnessed stags from insertion of CIDR devices (12 March or 9 April) and blood samples were taken every second day to determine plasma progesterone. Further blood samples were collected for determination of plasma LH and progesterone via indwelling jugular cannulae every 2 h for 72 h from removal of CIDR devices. Hinds were allocated randomly to an initial ovarian examination by laparoscopy at either 16 or 20 h (A and B), or 12 or 16 h (C and D) after the onset of oestrus, with laparoscopy repeated at intervals of 8 h until either ovulation was recorded (A and B), or for four successive occasions (C and D). All hinds received cloprostenol injections 15 days after device removal. A total of 28 hinds (78%) exhibited oestrus and a preovulatory LH surge, with mean (+/- SEM) times to onset of oestrus of 44.6 +/- 1.0 h (A; n = 7), 37.4 +/- 2.0 h (B; n = 7), 16.3 +/- 1.7 h (C; n = 6) or 14.0 +/- 1.7 h (D; n = 8). In Experiment 1, 24 suckled beef cows were assigned to 4 treatment groups (6 cows/group). Group I cows calved spontaneously. Parturition was induced in Groups 2, 3 and 4 with 20 mg dexamethasone (DEX) 8 to 12 days prior to expected calving date. Additionally, cows in Groups 3 and 4 received 8 mg triamcinalone acetonide (TA) 6 days prior to DEX treatment. Animals in Group 4 also received 10 mg estradiol benzoate (EB) with TA, and on alternate days until DEX, when 20 mg EB was given. Gonadotropin releasing hormone (GnRH, 100 mug) was given intramuscular (IM) to all cows on days 2 or 3 postpartum. Plasma LH increased (P< .05) following GnRH treatment in Groups 2, 3 and 4, but not in Group 1. LH release (area under the curve) following GnRH was greater (P< .05) for cows in Group 4 compared to cows in Groups 1, 2 or 3, and differences in LH release between Groups 1, 2 or 3 were not significant. In Experiment II, 36 mature Hereford cows were assigned to a 2 x 3 factorial experiment (6 cows/group). Groups 1 and 2, 3 and 5, and 4 and 6 received 0, 100, or 200 mug GnRH (IM) at 78 hr postpartum, respectively. In addition, cows in Groups 2, 5 and 6 received 5 mg EB at 36 hr postpartum. Plasma LH concentrations were not different (P <.05) among groups from 36 to 78 hr postpartum. A surge of LH in response to EB treatment was not detected at 54 to 62 hr (18 to 26 hr post EB), indicating a lack of response by the positive feedback mechanism at this early time postpartum. Mean plasma LH concentrations were elevated 78 to 82 hr postpartum for Groups 3 through 6. Treatment with EB at 36 hr caused a significantly greater (P< .05) response to GnRH with 200 mug of GnRH releasing more LH than 100 mug of GnRH.  相似文献

A new synthetic agent R, S-2-amino-1(2-amino-4, 5-dihydroxyphenyl) propane dihydrobromide, also referred to as α-methyl-6-aminodopamine (α-Me-6-ADA), has been found to produce acute (one day) and longer-term (seven day) depletion of norepinephrine (NE) levels in mouse brain and peripheral tissues. A 100 mg/kg dose of α-Me-6-ADA (i.v., free base) produced greater than 85% depletion of NE in the heart and spleen at one day and one week after treatment. Intracranially, α-Me-6-ADA (100 μg i.vtr.) depleted NE in the telencephalon and brain stem by 79% and 21% respectively at seven days. In addition DA was depleted by 45% in the ipsilateral striatum. The α-Me-6-ADA appears to have a relative selectivity for noradrenergic nerves, as an intracranial dose of 10 μg, which decreased NE in mouse whole brain by 52% at one day, failed to alter the DA content. These data suggest that α-Me-6-ADA may be a neurotoxin.  相似文献

The dynamic mechanical properties of purified collagen from bovine tendon were studied using a torsion pendulum in the temperature range of 120°-360°K at 0.3-1 cps. In the temperature range studied, two loss peaks were observed: a β-peak at about 200°K, and an α-peak approximately five times larger at about 280°K. The temperature of the α-transition is shown to be dependent on water content, decreasing with increasing amount of water and shifting to lower temperatures. Broad-line proton magnetic resonance results were also obtained on similar samples. A narrow nuclear magnetic resonance (NMR) line appears at about 250°C. The effects of shrinkage to form gelatin and of cross-linking on the relaxation behavior of collagen were also studied. The motions taking place in collagen over the 120°-360°K range are discussed.  相似文献

Pelvic exenteration offers the only possibility for cure in patients who have pelvic recurrence after receiving optimum amounts of irradiation. With improved radiotherapy techniques, the number of patients with isolated central failure is steadily diminishing, but there remains a significant number of patients with recurrent cancer of the cervix after radiation therapy for whom the procedure offers the only chance for life. Each patient must be assessed individually, with the risks of the procedure weighed against the possible benefits. Technical advances continue to reduce the operative mortality and ameliorate the postoperative morbidity associated with pelvic exenteration.  相似文献

Due to its hemizygous inheritance and role in sex determination, the X‐chromosome is expected to play an important role in the evolution of sexual dimorphism and to be enriched for sexually antagonistic genetic variation. By forcing the X‐chromosome to only be expressed in males over >40 generations, we changed the selection pressures on the X to become similar to those experienced by the Y. This releases the X from any constraints arising from selection in females and should lead to specialization for male fitness, which could occur either via direct effects of X‐linked loci or trans‐regulation of autosomal loci by the X. We found evidence of masculinization via up‐regulation of male‐benefit sexually antagonistic genes and down‐regulation of X‐linked female‐benefit genes. Potential artefacts of the experimental evolution protocol are discussed and cannot be wholly discounted, leading to several caveats. Interestingly, we could detect evidence of microevolutionary changes consistent with previously documented macroevolutionary patterns, such as changes in expression consistent with previously established patterns of sexual dimorphism, an increase in the expression of metabolic genes related to mito‐nuclear conflict and evidence that dosage compensation effects can be rapidly altered. These results confirm the importance of the X in the evolution of sexual dimorphism and as a source for sexually antagonistic genetic variation and demonstrate that experimental evolution can be a fruitful method for testing theories of sex chromosome evolution.  相似文献
