Summary Analysis of reconstructions, prepared from electron micrographs of successive longitudinal serial sections, has led to the conclusion that the somatic telophase chromosome of Tradescantia paludosa contains four cytologically separable chromonemata. The four represent a pair of pairs, that is, two diplospiremes — one with its two chromonemata arranged helically in dextrorse relationship, and the other with its two in sinistrorse relationship — which are associated to form a tetraspireme. During anaphase and telophase the tetraspireme constitutes the chromosome; during prophase and metaphase the tetraspireme represents one of the two chromatids of the chromosome, which is accordingly an octospireme in terms of the number of cytologically identifiable chromonemata. Photolysis of E-[ring-2-14C]urocanic acid (UA) with native or denatured calf thymus DNA leads to covalent binding of the radiolabel to the nucleic acid. A similar observation is made upon photolysis of the labeled UA with the polyribonucleotides, in which case a strong preference is observed for binding to poly[U]. DNA or poly[U], which had been reacted with UA and purified by dialysis and multiple precipitations, releases UA upon further irradiation with 254 nm light (as expected for cyclobutane adducts). Quantum efficiencies for binding of the UA to native DNA have been measured at 308 and 266 nm and are 0.30 x 10(-5) and 1.3 x 10(-4), respectively, at comparable reactant concentrations. The large increase at the shorter wavelength (where DNA absorption is more competitive) is taken as evidence for the primary role of a DNA excited state in initiating the binding reaction(s). 相似文献

Fire regimes have three inter-related components that can affect population dynamics: frequency, intensity and season. However,
there has been little effort to study the effects of any of these components on arbuscular mycorrhizas (AM). In order to examine
the long-term effects of fire frequency on AM colonization, the roots of Dillwynia retorta were examined at 32 sites supporting Hawkesbury Sandstone vegetation in the Sydney region of southeastern Australia. These
sites were representative of the broad-scale variability in fire frequencies with respect to the length and timing of inter-fire
intervals found in the Sydney region during the previous 30 years. The length of the shortest inter-fire interval was significantly
correlated with total AM colonization, and the length of the longest inter-fire interval was related to the arbuscular colonization.
The length of the most-recent inter-fire interval and the time since the shortest inter-fire interval were not related to
AM colonization in D. retorta. Furthermore, AM colonization was directly related to the local abundance of the host plant, indicating that the effects
of fire frequency on AM colonization are likely to occur indirectly via direct effects on the host plant. Canonical correspondence
analysis demonstrated that the presence and abundance of alternate potential hosts had no influence on mycotrophy in D. retorta. Thus, the impact of fire on D. retorta was probably the main factor influencing its mycorrhizal status in relation to fire history.

The dissociation of the purified human placental alpha 2 beta 2 heterotetrameric insulin receptor complex into an alpha beta heterodimeric state was found to occur in a pH- and dithiothreitol (DTT)-dependent manner. Formation of the alpha beta heterodimeric complex, under conditions which preserved tracer insulin binding and protein kinase activities (pH 8.75 for 25 min followed by 2.0 mM DTT for 5 min) occurred with an approximate 50% efficiency. The resulting nondissociated alpha 2 beta 2 heterotetrameric complexes could then be separated effectively by Bio-Gel A-1.5m gel filtration chromatography at neutral pH. The isolated DTT-treated but nondissociated alpha 2 beta 2 heterotetrameric complex was resistant to any further dissociation by a second round of DTT and alkaline pH treatment, whereas the isolated alpha beta heterodimeric complex was stable to spontaneous reassociation for at least 72 h at pH 7.60. Kinetic analyses of the insulin receptor protein kinase activity demonstrated that the insulin stimulation of glutamic acid:tyrosine (4:1) synthetic polymer phosphorylation for both the alpha 2 beta 2 heterotetrameric and alpha beta heterodimeric complexes occurred via an increase in Vmax without any significant change in Km. Examination of beta subunit autophosphorylation of the alpha beta heterodimeric complex, in the presence but not in the absence of insulin, demonstrated the appearance of the covalent 32P-labeled alpha 2 beta 2 heterotetrameric complex. Further, the initial rate of insulin-stimulated beta subunit autophosphorylation in the isolated alpha beta heterodimeric complex occurred in a dilution-dependent (intermolecular) manner. These data demonstrate that the isolated alpha beta heterodimeric insulin receptor complex is fully capable of expressing insulin-dependent activation of the beta subunit protein kinase domain with the covalent reassociation of the alpha beta heterodimeric complex into an alpha 2 beta 2 heterotetrameric disulfide-linked state. 相似文献
