This study uses survey and tagging data and cluster analysis to interpret dynamics of ontogenetic movements and seasonal migrations in snow crab (Chionoecetes opilio) along the Newfoundland and Labrador (NL) continental shelves. Most historic literature from Atlantic Canada suggests snow crab undertake small-scale ontogenetic movements while observations of seasonal migrations had been near-exclusive to the inshore. Information on both types of movement in the most spatially expansive offshore region of Atlantic Canada, in NL, was lacking. We find that that both ontogenetic movements and seasonal migrations occur in most areas of the NL offshore, with ontogenetic movements generally down-slope and seasonal migrations generally up-slope. Conservative estimates of average ontogenetic movements range from 54 to 72 km for both males and females in the largest offshore regions while seasonal migrations are slightly smaller, with two independent studies on the Grand Bank producing average estimates of 43–46 km and an adjacent tagging study in a smaller inshore bay producing an average estimate of 25 km. Ontogenetic movements appear associated with a search for warm water while seasonal migrations appear associated with both mating and molting in shallow water. On average, morphometrically mature crab of both sexes move less vertical distance than morphometrically immature crab during seasonal migrations. We investigate plausible explanations for ontogenetic movements and spring migrations and detail how bottom temperature affects crab distribution and life history dynamics. We further document movement patterns from tagging studies on the burgeoning snow crab stock in the Eastern Barents Sea toward establishing consistencies in species behaviour on the global scale. Finally, we discuss explanations for historical disparities in the literature between scales of movement for snow crab in the Eastern Bering Sea of Alaska versus Atlantic Canada and advance perspectives on life history theory for the species.  相似文献

Drugs of addiction lead to a wide range of epigenetic changes at the promoter regions of genes directly implicated in learning and memory processes. We have previously shown that the histone deactylase inhibitor, sodium butyrate (NaB), accelerates the extinction of nicotine‐seeking and provides resistance to relapse. Here, we explore the potential molecular mechanisms underlying this effect. Rats received intravenous nicotine or saline self‐administration, followed by 6 days of extinction training, with each extinction session followed immediately by treatment with NaB or vehicle. On the last day of extinction, rats were killed and the medial ventral prefrontal cortex retained for chromatin immunoprecipitation and quantitative polymerase chain reaction (qPCR). A history of nicotine exposure significantly decreased H3K14 acetylation at the brain‐derived neurotrophic factor (BDNF) exon IV promoter, and this effect was abolished with NaB treatment. In contrast, nicotine self‐administration alone, resulted in a significant decrease in histone methylation at the H3K27me3 and H3K9me2 marks in the promoter regions of BDNF exon IV and cyclin‐dependent kinase 5 (Cdk‐5). Quantitative PCR‐identified changes in several genes associated with NaB treatment that were independent of nicotine exposure; however, an interaction of nicotine history and NaB treatment was detected only in the expression of BDNF IV and BDNF IX. Together these results suggest that nicotine self‐administration leads to a number of epigenetic changes at both the BDNF and Cdk‐5 promoters, and that these changes may contribute to the enhanced extinction of nicotine‐seeking by NaB.  相似文献

Colloidal quantum dots are promising materials for flexible solar cells, as they have a large absorption coefficient at visible and infrared wavelengths, a band gap that can be tuned across the solar spectrum, and compatibility with solution processing. However, the performance of flexible solar cells can be degraded by the loss of charge carriers due to recombination pathways that exist at a junction interface as well as the strained interface of the semiconducting layers. The modulation of the charge carrier transport by the piezoelectric effect is an effective way of resolving and improving the inherent material and structural defects. By inserting a porous piezoelectric poly(vinylidenefluoride‐trifluoroethylene) layer so as to generate a converging electric field, it is possible to modulate the junction properties and consequently enhance the charge carrier behavior at the junction. This study shows that due to a reduction in the recombination and an improvement in the carrier extraction, a 38% increase in the current density along with a concomitant increase of 37% in the power conversion efficiency of flexible quantum dots solar cells can be achieved by modulating the junction properties using the piezoelectric effect.  相似文献

Climate change is increasing the threat of erosion and flooding along coastlines globally. Engineering solutions (e.g. seawalls and breakwaters) in response to protecting coastal communities and associated infrastructure are increasingly becoming economically and ecologically unsustainable. This has led to recommendations to create or restore natural habitats, such as sand dunes, saltmarsh, mangroves, seagrass and kelp beds, and coral and shellfish reefs, to provide coastal protection in place of (or to complement) artificial structures. Coastal managers are frequently faced with the problem of an eroding coastline, which requires a decision on what mitigation options are most appropriate to implement. A barrier to uptake of nature‐based coastal defence is stringent evaluation of the effectiveness in comparison to artificial protection structures. Here, we assess the current evidence for the efficacy of nature‐based vs. artificial coastal protection and discuss future research needs. Future projects should evaluate habitats created or restored for coastal defence for cost‐effectiveness in comparison to an artificial structure under the same environmental conditions. Cost‐benefit analyses should take into consideration all ecosystem services provided by nature‐based or artificial structures in addition to coastal protection. Interdisciplinary research among scientists, coastal managers and engineers is required to facilitate the experimental trials needed to test the value of these shoreline protection schemes, in order to support their use as alternatives to artificial structures. This research needs to happen now as our rapidly changing climate requires new and innovative solutions to reduce the vulnerability of coastal communities to an increasingly uncertain future.  相似文献

The most carbon (C)‐dense ecosystems of Amazonia are areas characterized by the presence of peatlands. However, Amazonian peatland ecosystems are poorly understood and are threatened by human activities. Here, we present an investigation into long‐term ecohydrological controls on C accumulation in an Amazonian peat dome. This site is the oldest peatland yet discovered in Amazonia (peat initiation ca. 8.9 ka BP), and developed in three stages: (i) peat initiated in an abandoned river channel with open water and aquatic plants; (ii) inundated forest swamp; and (iii) raised peat dome (since ca. 3.9 ka BP). Local burning occurred at least three times in the past 4,500 years. Two phases of particularly rapid C accumulation (ca. 6.6–6.1 and ca. 4.9–3.9 ka BP), potentially resulting from increased net primary productivity, were seemingly driven by drier conditions associated with widespread drought events. The association of drought phases with major ecosystem state shifts (open water wetland–forest swamp–peat dome) suggests a potential climatic control on the developmental trajectory of this tropical peatland. A third drought phase centred on ca. 1.8–1.1 ka BP led to markedly reduced C accumulation and potentially a hiatus during the peat dome stage. Our results suggest that future droughts may lead to phases of rapid C accumulation in some inundated tropical peat swamps, although this can lead ultimately to a shift to ombrotrophy and a subsequent return to slower C accumulation. Conversely, in ombrotrophic peat domes, droughts may lead to reduced C accumulation or even net loss of peat. Increased surface wetness at our site in recent decades may reflect a shift towards a wetter climate in western Amazonia. Amazonian peatlands represent important carbon stores and habitats, and are important archives of past climatic and ecological information. They should form key foci for conservation efforts.  相似文献

For many commercial potato cultivars, tuber yield is optimal at average daytime temperatures in the range of 14–22 °C. Further rises in ambient temperature can reduce or completely inhibit potato tuber production, with damaging consequences for both producer and consumer. The aim of this study was to use a genetic screen based on a model tuberization assay to identify quantitative trait loci (QTL) associated with enhanced tuber yield. A candidate gene encoding HSc70 was identified within one of the three QTL intervals associated with elevated yield in a Phureja–Tuberosum hybrid diploid potato population (06H1). A particular HSc70 allelic variant was linked to elevated yield in the 06H1 progeny. Expression of this allelic variant was much higher than other alleles, particularly on exposure to moderately elevated temperature. Transient expression of this allele in Nicotiana benthamiana resulted in significantly enhanced tolerance to elevated temperature. An TA repeat element was present in the promoter of this allele, but not in other HSc70 alleles identified in the population. Expression of the HSc70 allelic variant under its native promoter in the potato cultivar Desiree resulted in enhanced HSc70 expression at elevated temperature. This was reflected in greater tolerance to heat stress as determined by improved yield under moderately elevated temperature in a model nodal cutting tuberization system and in plants grown from stem cuttings. Our results identify HSc70 expression level as a significant factor influencing yield stability under moderately elevated temperature and identify specific allelic variants of HSc70 for the induction of thermotolerance via conventional introgression or molecular breeding approaches.  相似文献

Phospholipase D (PLD) genes are members of a superfamily that is defined by several highly conserved motifs. PLD in mammals has been proposed to play a role in membrane vesicular trafficking and signal transduction. Using site-directed mutagenesis, 25 point mutants have been made in human PLD1 (hPLD1) and characterized. We find that a motif (HxKxxxxD) and a serine/threonine conserved in all members of the PLD superfamily are critical for PLD biochemical activity, suggesting a possible catalytic mechanism. Functional analysis of catalytically inactive point mutants for yeast PLD demonstrates that the meiotic phenotype ensuing from PLD deficiency in yeast derives from a loss of enzymatic activity. Finally, mutation of an HxKxxxxD motif found in a vaccinia viral protein expressed in the Golgi complex results in loss of efficient vaccinia virus cell-to-cell spreading, implicating the viral protein as a member of the superfamily and suggesting that it encodes a lipid modifying or binding activity. The results suggest that vaccinia virus and hPLD1 may act through analogous mechanisms to effect viral cellular egress and vesicular trafficking, respectively.  相似文献

The NPM-ALK fusion gene, formed by the t(2;5)(p23;q35) translocation in non-Hodgkin's lymphoma, encodes a 75-kDa hybrid protein that contains the amino-terminal 117 amino acid residues of the nucleolar phosphoprotein nucleophosmin (NPM) joined to the entire cytoplasmic portion of the receptor tyrosine kinase ALK (anaplastic lymphoma kinase). Here, we demonstrate the transforming ability of NPM-ALK and show that oncogenesis by the chimeric protein requires the activation of its kinase function as a result of oligomerization mediated by the NPM segment. Sedimentation gradient experiments revealed that NPM-ALK forms in vivo multimeric complexes of approximately 200 kDa or greater that also contain normal NPM. Cell fractionation studies of the t(2;5) translocation-containing lymphoma cell line SUP-M2 showed NPM-ALK to be localized within both the cytoplasmic and nuclear compartments. Immunostaining performed with both polyclonal and monoclonal anti-ALK antibodies confirmed the dual location of the oncoprotein and also indicated that NPM-ALK is abundant within both the nucleoplasm and the nucleolus. An intact NPM segment is absolutely required for NPM-ALK-mediated oncogenesis, as indicated by our observation that three different NPM-ALK mutant proteins lacking nonoverlapping portions of the NPM segment were each unable to form complexes, lacked kinase activity in vivo, and failed to transform cells. However, NPM could be functionally replaced in the fusion protein with the portion of the unrelated translocated promoter region (TPR) protein that activates the TPR-MET fusion kinase by mediating dimerization through its leucine zipper motif. This engineered TPR-ALK hybrid protein, which transformed cells almost as efficiently as NPM-ALK, was localized solely within the cytoplasm of cells. These data indicate that the nuclear and nucleolar localization of NPM-ALK, which probably occur because of transport via the shuttling activity of NPM, is not required for oncogenesis. Further, the activation of the truncated ALK protein by a completely heterologous oligomerization domain suggests that the functionally important role of the NPM segment of NPM-ALK in transformation is restricted to the formation of kinase-active oligomers and does not involve the alteration of normal NPM functions.  相似文献

To help feed, clothe, house, improve land productivity, confront difficult resource necessities, and stabilize ravaged land resources through reforestation, there is need for a revitalized worldwide assessment of little-known legume species. Many legumes produce economically important organic compounds such as gums, dyes, Pharmaceuticals, pesticides, phytochemicals, and many specialty products. However, most legumes have never been or have just begun to be studied for phy-tochemical or biologically active components, and new reservoirs of commercially valuable materials remain to be discovered. Not only could these legumes provide healthier and a more secure food supply for Third World countries, but they can protect fragile soil resources where erosion now threatens tragedy.  相似文献
