The goal of this study was to examine the state of hepatocyte mitochondrial respiratory chain of rats with toxic hepatitis
induced by CCl4 and ethanol. Oxygen consumption by hepatocytes and mitochondria was determined. Endogenous oxygen consumption by pathological
hepatocytes was 1.3-fold higher compared with control. Rotenone resulted in 27% suppression of respiration by pathological
hepatocytes whereas 2,4-dinitrophenol produced a 1.4-fold increase of respiration. States 3 and 4 of mitochondrial respiration
with malate and glutamate were found to be higher as compared to control. State dinitrophenol and state 3 respirations were
similar within every group of animals when being tested with malate and glutamate or succinate. Cytochrome c oxidase activity in hepatitis was 1.8-fold higher compared with control. Simvastatin administration resulted in a decrease
in hepatocyte endogenous respiration in hepatitis. The presented data lead to the assumption that the increased oxygen consumption
by the respiratory chain of pathological mitochondria to be linked mainly with the altered function of complex I. 相似文献

Rat muscle infiltration by neutrophils after muscle activity (MA) was investigated on myeloperoxidase (MPO) concentration. MPO distribution in muscle subcellular fractions was also studied. Increase of MPO concentration in skeletal muscles was discovered after MA. Its maximum was determined within 1-5 days of the rest. This fact can be considered as an evidence of neutrophil influx in muscle tissue. The electroral MPO concentration increase in plasmalemma membrane fraction after MA was shown. In vitro MPO was able to catalyze 125I inclusion in membrane material. These results give a possibility to propose that neutrophil MPO can have a certain significance in muscle tissue damage by haloid joining to plasmalemma proteins. 相似文献

The intensity of nucleic acids and protein synthesis in the cells of the thymus and the bursa of Fabricius was studied in chickens against the background of an immunodepression induced by administration of hydrocortisone and cyclophosphamide. It was found out that hydrocortisone causes in chicken a marked lowering of the intensity of inclusion of 3H-thymidine, 3H-uridine and 14C-glycine in thymic cells, and cyclophosphamide--in the cells of the bursa of Fabricius. Under the conditions of selective immunodepression the preparations on the basis of the peptides of the thymus (thymalin) and of the bursa of Fabricius (bursilin) regulate the processes of nucleic acids and protein synthesis chiefly in the cells of organs which produce them. 相似文献

Morphometry and cytofluorometry of the kinetics of phagocytosis were applied to study the quantitative mechanisms of acidification in the area of contact between the plasma membrane of macrophages and Candida albicans yeast cells conjugated with fluorescein isothiocyanate. It was found by stereological transformation of the morphometry data that the main part of macrophages phagocytose the limiting amount of particles by minute 5-10. A good agreement was established between the cytofluorometric histograms and the stereological data. This made it possible to evaluate, using the calibration curve, the pH on the surface of the phagocytosed material based on the fluorescence quenching. As an advantage of the suggested comprehensive approach to the study of acidification, the authors stress the possibility of making structural-functional analysis of the rearrangements occurring in the intact phagocytosing cell. 相似文献

A micromethod for measurement of mass changes of glutaraldehyde treated protein crystals is presented. The method is based on analysis of transverse resonance vibration of a cantilevered tungsten micro-needle (1,5 divided by 2 mm long, 30 divided by 40 mkm in diameter) having the specimen stuck on its free sharp end. The method is accurate to within 0.1% for specimens with masses 0.1 divided by 0.01 mg. Absorption isotherms for water uptake by triclinic (P1), monoclinic (P2(1) ) and tetragonal (P4(3)2(1)2) crystals as well as by amorphous films of hen egg-white lysozyme are obtained. Masked tonal thresholds by noise in the bottlenose dolphin have been measured as a function of angle of noise direction. Sharp directional selectivity of hearing has been discovered resulting in an abrupt decrease of noise masking with the increase of the angle of noise coming. It has been shown that narrow directional characteristics of hearing is an angle 8 degrees plus or minus 2 degrees for the level of minus 3db. It is one of the main mechanisms supplying the echolocation system of the dolphin with the high level of resistance to noise. 相似文献
