Kindler Syndrome (KS), characterized by transient skin blistering followed by abnormal pigmentation, skin atrophy, and skin cancer, is caused by mutations in the FERMT1 gene. Although a few KS patients have been reported to also develop ulcerative colitis (UC), a causal link to the FERMT1 gene mutation is unknown. The FERMT1 gene product belongs to a family of focal adhesion proteins (Kindlin-1, -2, -3) that bind several β integrin cytoplasmic domains. Here, we show that deleting Kindlin-1 in mice gives rise to skin atrophy and an intestinal epithelial dysfunction with similarities to human UC. This intestinal dysfunction results in perinatal lethality and is triggered by defective intestinal epithelial cell integrin activation, leading to detachment of this barrier followed by a destructive inflammatory response.  相似文献

Evolutionary theory predicts that high genetic variation maintains plasticity in a species’ response to parasite pressure. However, higher genetic diversity might also cause easier infiltration by social parasites, because odour diversity is high and nest-mate recognition poor. Here we test if the obligate myrmecophile Lycaenid butterfly Phengaris nausithous, a parasite of colonies of the highly polygynous ant Myrmica rubra causes local adaptation by enhancing genetic variance in parasitized versus non parasitized ant populations M. rubra colonies from six infested and three uninfested sites were assayed at five microsatellite loci to quantify genetic variation. Our results reveal isolation by distance and a significantly enhanced intracolonial variance due to the parasite pressure.  相似文献

Visual arrestin specifically binds to photoactivated and phosphorylated rhodopsin and inactivates phototransduction. In contrast, the p44 splice variant can terminate phototransduction by binding to nonphosphorylated light-activated rhodopsin. Here we report the crystal structure of bovine p44 at a resolution of 1.85 Å. Compared to native arrestin, the p44 structure reveals significant differences in regions crucial for receptor binding, namely flexible loop V–VI and polar core regions. Additionally, electrostatic potential is remarkably positive on the N-domain and the C-domain. The p44 structure represents an active conformation that serves as a model to explain the ‘constitutive activity’ found in arrestin variants.  相似文献

The contribution of soil microbial residues to stable carbon pools may be of particular importance in the tropics where carbon residence times are short and any available carbon is rapidly utilized. In this study we investigated the vertical distribution of microbially-derived amino sugars in two tropical forests on contrasting meta-sedimentary and serpentinite parent materials in the lowlands of Mt. Kinabalu, Borneo. Despite their similar climate, vegetative cover, and general microbial community structure, the two soils were chemically and physically distinct. We found that both parent material and depth significantly influenced the pool sizes of microbial residues in the two soils. In particular, the soil derived from sedimentary parent material had greater amino sugar contents, glucosamine to galactosamine ratios, and percentage of total soil carbon that is amino sugar derived, than the soil derived from serpentinite substrate. We speculate that residue stabilization was linked to soil iron oxide content, with significant difference in amino sugars contribution to total soil carbon at depth in the serpentinite-derived soil versus that derived from sedimentary parent material. Based on observed patterns of amino sugar content and relative abundance we suggest that near the surface of both soils vegetation and litter input determines the composition and quantity of microbial residues. With increasing depth the influence of vegetation declines and production and stabilization of microbial amino sugars becomes driven by soil matrix characteristics. These differences in stabilization mechanism and carbon dynamics with depth may be particularly critical in deep weathered tropical soils.  相似文献

The quantity and composition of the six major mandibular gland components of young queenless workers of the Cape honeybee (Apis mellifera capensis) were determined. The total amount of the six components increased with age. The relative quantities in the mandibular gland secretion of queenless caged workers were found to change rapidly during the first 4 days after emergence and to become dominated by the queen substance, 9-keto-2(E)-decenoic acid. Also the relative amounts of 9-hydroxy-decenoic acid, a precursor of the queen substance, showed an increase of an order of magnitude within the first 4 days of imaginal life. The relative amounts of the aromatic compounds typical to the queen pheromone remained similar in this developmental time window. The increase of queenlike compounds is particularly strong between days two and three after emergence. These queen-like pheromones play a major role in the development of reproductive hierarchies among workers under queenless conditions. This may be an important factor in the socio-parasitic pathway of A. m. capensis.  相似文献

Adaptation of rainbow fish to lake and stream habitats   总被引:3,自引:0,他引:3  
Fish occupy a range of hydrological habitats that exert different demands on locomotor performance. We examined replicate natural populations of the rainbow fishes Melanotaenia eachamensis and M. duboulayi to determine if colonization of low-velocity (lake) habitats by fish from high-velocity (stream) habitats resulted in adaptation of locomotor morphology and performance. Relative to stream conspecifics, lake fish had more posteriorly positioned first dorsal and pelvic fins, and shorter second dorsal fin bases. Habitat dimorphism observed between wild-caught fish was determined to be heritable as it was retained in M. eachamensis offspring raised in a common garden. Repeated evolution of the same heritable phenotype in independently derived populations indicated body shape divergence was a consequence of natural selection. Morphological divergence between hydrological habitats did not support a priori expectations of deeper bodies and caudal peduncles in lake fish. However, observed divergence in fin positioning was consistent with a family-wide association between habitat and morphology, and with empirical studies on other fish species. As predicted, decreased demand for sustained swimming in lakes resulted in a reduction in caudal red muscle area of lake fish relative to their stream counterparts. Melanotaenia duboulayi lake fish also had slower sustained swimming speeds (Ucrit) than stream conspecifics. In M. eachamensis, habitat affected Ucrit of males and females differently. Specifically, females exhibited the pattern observed in M. duboulayi (lake fish had faster Ucrit than stream fish), but the opposite association was observed in males (stream males had slower Ucrit than lake males). Stream M. eachamensis also exhibited a reversed pattern of sexual dimorphism in Ucrit (males slower than females) relative to all other groups (males faster than females). We suggest that M. eachamensis males from streams responded to factors other than water velocity. Although replication of muscle and Ucrit phenotypes across same habitat populations within and/or among species was suggestive of adaptation, the common garden experiment did not confirm a genetic basis to these associations. Kinematic studies should consider the effect of the position and base length of dorsal fins.  相似文献

Sympatric individuals of Rattus fuscipes and Rattus leucopus, two Australian native rats from the tropical wet forests of north Queensland, are difficult to distinguish morphologically and are often confused in the field. When we started a study on fine-scale movements of these species, using microsatellite markers, we found that the species as identified in the field did not form coherent genetic groups. In this study, we examined the potential of an iterative process of genetic assignment to separate specimens from distinct (e.g. species, populations) natural groups. Five loci with extensive overlap in allele distributions between species were used for the iterative process. Samples were randomly distributed into two starting groups of equal size and then subjected to the test. At each iteration, misassigned samples switched groups, and the output groups from a given round of assignment formed the input groups for the next round. All samples were assigned correctly on the 10th iteration, in which two genetic groups were clearly separated. Mitochondrial DNA sequences were obtained from samples from each genetic group identified by assignment, together with those of museum voucher specimens, to assess which species corresponded to which genetic group. The iterative procedure was also used to resolve groups within species, adequately separating the genetically identified R. leucopus from our two sampling sites. These results show that the iterative assignment process can correctly differentiate samples into their appropriate natural groups when diagnostic genetic markers are not available, which allowed us to resolve accurately the two R. leucopus and R. fuscipes species. Our approach provides an analytical tool that may be applicable to a broad variety of situations where genetic groups need to be resolved.  相似文献

Prioritizing areas for conservation requires the use of surrogates for assessing overall patterns of biodiversity. Effective surrogates will reflect general biogeographical patterns and the evolutionary processes that have given rise to these and their efficiency is likely to be influenced by several factors, including the spatial scale of species turnover and the overall congruence of the biogeographical history. We examine patterns of surrogacy for insects, snails, one family of plants and vertebrates from rainforests of northeast Queensland, an area characterized by high endemicity and an underlying history of climate-induced vicariance. Nearly all taxa provided some level of prediction of the conservation values for others. However, despite an overall correlation of the patterns of species richness and complementarity, the efficiency of surrogacy was highly asymmetric; snails and insects were strong predictors of conservation priorities for vertebrates, but not vice versa. These results confirm predictions that taxon surrogates can be effective in highly diverse tropical systems where there is a strong history of vicariant biogeography, but also indicate that correlated patterns for species richness and/or complementarity do not guarantee that one taxon will be efficient as a surrogate for another. In our case, the highly diverse and narrowly distributed invertebrates were more efficient as predictors than the less diverse and more broadly distributed vertebrates.  相似文献
