Twenty-six pig fetuses (at 94 days gestational age) were fitted with carotid artery catheters. Eight fetuses were given 1,500 ng of IGF-1 (in 100 microliters) directly into lateral cerebral ventricle; 7 further fetuses received the IGF-1 together with 150 microliters of a potent specific anti-somatostatin serum into a ventricle, 5 other fetuses received the anti-somatostatin serum alone while 6 controls received normal sheep serum. Administration of IGF-1 caused a rapid decrease in circulating growth hormone (pGH) levels but there was no significant change in plasma levels of somatostatin immediately following the IGF-1 administration, suggesting that the decrease in pGH was not mediated by somatostatin secretion. Further evidence that somatostatin was not involved in this effect was provided by the lack of effect of concurrent antisomatostatin serum on the IGF-1-induced decrease in pGH. Thus the high circulating levels of GH in the fetus may result from a lack of feedback of IGF-1, but not through the somatostatin-pituitary axis.  相似文献

The growth characteristics of five octopine-catabolizing pseudomonads have been determined in batch and continuous cultures. All five strains belonged to rRNA homology group I and showed a more psychrotrophic growth pattern than did Agrobacterium tumefaciens B6 and ATCC 15955. In chemostats limited by octopine, either as the source of carbon and nitrogen or the sole source of nitrogen, maximum specific growth rates and substrate affinities were lower than those in chemostats limited by glutamate. These growth dynamics were similar to those observed for Agrobacterium strains B6 and ATCC 15955 even though the catabolic genes and pathways are believed to be different in the two genera. An analysis of the yields in octopine-limited chemostats indicated that the use of octopine as the sole source of carbon and nitrogen was grossly inefficient. Octopine and presumably lysopine and octopinic acid provided a better source of nitrogen than of carbon. One of the Pseudomonas fluorescens strains, E175D, was able to produce its highest yield on octopine as a nitrogen source. Competition models formulated on pure culture parameters indicated that two of the Pseudomonas spp. would dominate A. tumefaciens B6 and ATCC 15955 when in simple competition for octopine as a limiting substrate.  相似文献

An agar medium, LL-agar (lactate-lead acetate) was designed to selectively differentiate members of the genus Pectinatus (S. Y. Lee, M. S. Mabee, and N. O. Jangaard, Int. J. Syst. Bacteriol. 28:582-594, 1978; S. Y. Lee, M. S. Mabee, N. O. Jangaard, and E. K. Horiuchi, J. Inst. Brew. 86:28-30, 1980) from other brewery microorganisms. Selectivity was achieved by the use of sodium lactate as the sole source of carbon and phenylethyl alcohol as an inhibitor for aerobic gram-negative bacteria and yeast. Differentiation was established by the introduction of lead acetate into the medium, which reacted with the H2S liberated by Pectinatus and resulted in a blackening of the Pectinatus colonies while the other brewery organisms, when present, remained white. In combination with the Lee tube (J. E. Ogg, S. Y. Lee, and B. J. Ogg, Can. J. Microbiol. 25:987-990, 1979) and this medium, isolation of Pectinatus organisms from beer samples was accomplished with convenience and simplicity.  相似文献

A rapid and sensitive method has been outlined for the measurement of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) utilizing a weak cation-exchange resin and liquid chromatography with electrochemical detection. The sensitivity of the procedure allows measurement of the amine in punches of rat substantia nigra even after local injection of the neurotoxin 5,7-dihydroxytryptamine. Increases in 5-HT and decreases in 5-HIAA concentrations after pargyline, and selective increases in 5-HIAA concentrations after probenecid were detected in selected brain regions (nucleus accumbens, anterior striatum, substantia nigra). Thus, this procedure is sensitive enough to estimate 5-HT turnover in discrete nuclei of the rat brain.  相似文献

When C57BL/6 mice were injected iv with BCG in an oil-in-saline emulsion, they developed intense pulmonary granulomatous inflammation (PGI) and splenomegaly as well as chemotactic activity for macrophages and angiotensin-converting enzyme (ACE) in their lung fluids. PGI, splenomegaly, and levels of chemotactic activity and ACE were markedly reduced in T-cell-deficient “B” mice. The capacity to develop PGI was fully restored and splenomegaly was partially restored in “B” mice by the provision of syngeneic thymocytes, spleen cells, or purified T cells. These results indicate that the full expression of BCG-induced PGI is dependent upon thymus-derived cells and is associated with high levels of chemotactic activity for macrophages and ACE in the lung lavage fluid. Although BCG-induced splenomegaly appears to be T cell dependent, it did not reach its full magnitude in reconstituted “B” mice.  相似文献

Molecular recombination and the repair of DNA double-strand breaks (DSB) have been examined in the G-0 and S phase of the cell cycle using a temperature-sensitive CHO cell line to test i) if there are cell cycle restrictions on the repair of DSB's' ii) the extent to which molecular recombination can be induced between either sister chromatids or homologous chromosomes and iii) whether repair of DSB's involves recombination (3). Mitomycin C (1-2 micrograms/ml) or ionizing radiation (50 krad) followed by incubation resulted in molecular recombination (hybrid DNA) in S phase cells. Approximately 0.03 to 0.10% of the molecules (number average molecular weight: 5.6 x 10(6) Daltons after shearing) had hybrid regions for more than 75% of their length. However, no recombination was detected in G-0 cells. Since the repair of DSB was observed in both stages with more than 50% of the breaks repaired in 5 hours, it appears that DSB repair in G-0 cells does not involve recombination between homologous chromosomes. The possibility is not excluded that repair in G-0 cells involves only small regions (less than 4 x 10(6) Daltons).  相似文献

Studies were performed to investigate the effects of the immunosuppressive chemical TCDD. Fetal and neonatal rats were exposed to TCDD through maternal dosing (5 μg/Kg) at Day 18 of gestation and on Days 0, 7, and 14 of postnatal life. Another group of neonatal rats were exposed to TCDD through maternal dosing on Days 0, 7, and 14 of postnatal life only. Parameters of cell-mediated and humoral immune function were investgiated. TCDD suppressed delayed hypersensitivity responses and responses to the mitogens Con A and PHA without affecting humoral immune function. Suppression of T-cell function was selective in that helper function was not suppressed. Transfer of primed T-lymphocytes from TCDD treated and non-treated animals into neonatally thymectomized animals confirmed this. Results indicate that delayed hypersensitivity function and helper function reside in distinct T-cell subsets.  相似文献
