Furfural and 5-hydroxymethylfurfural (HMF) are representative inhibitors generated from biomass pretreatment using dilute acid hydrolysis that interfere with yeast growth and subsequent fermentation. Few yeast strains tolerant to inhibitors are available. In this study, we report a tolerant strain, Saccharomyces cerevisiae NRRL Y-50049, which has enhanced biotransformation ability to convert furfural to furan methanol (FM), HMF to furan di-methanol (FDM), and produce a normal yield of ethanol. Our recent identification of HMF and development of protocol to synthesize the HMF metabolic conversion product FDM allowed studies on fermentation metabolic kinetics in the presence of HMF and furfural. Individual gene-encoding enzymes possessing aldehyde reduction activities demonstrated cofactor preference for NADH or NADPH. However, protein extract from whole yeast cells showed equally strong aldehyde reduction activities coupled with either cofactor. Deletion of a single candidate gene did not affect yeast growth in the presence of the inhibitors. Our results suggest that detoxification of furfural and HMF by the ethanologenic yeast S. cerevisiae strain Y-50049 likely involves multiple gene mediated NAD(P)H-dependent aldehyde reduction. Conversion pathways of furfural and HMF relevant to glycolysis and ethanol production were refined based on our findings in this study. 1,3-propanediol oxidoreductase (DhaT), which catalyzes the conversion of 3-hydroxypropionaldehyde (3-HPA) to 1,3-propanediol (1,3-PD) with the oxidation of NADH to NAD+, is a key enzyme in the production of 1,3-PD from glycerol. DhaT is known to be severely inactivated by its physiological substrate, 3-HPA, due to the reaction of 3-HPA with the thiol group of the cysteine residues. In this study, using site-directed mutagenesis, four cysteine residues in Klebsiella pneumoniae J2B DhaT were substituted to alanine, the amino acid commonly found in cysteine’s positions in other DhaT, individually and in combination. Among the total of 15 mutants developed, a double mutant (C28A_C107A) and a triple mutant (C28A_C93A_C107A) exhibited approximately 50 and 16% higher activity than the wild-type counterpart, respectively, after 1 h incubation with 10 mM 3-HPA. According to detailed kinetic studies, the double mutant had slightly better kinetic properties (V max , K cat , and K m for both 3-HPA and NADH) than wild-type DhaT. This study shows that DhaT stability against 3-HPA can be increased by cysteine-residue removal, albeit to a limited extent.  相似文献

There is considerable experimental evidence that hyperactive Ras proteins promote breast cancer growth and development including invasiveness, despite the low frequency of mutated forms of Ras in breast cancer. We have previously shown that H-Ras, but not N-Ras, induces an invasive phenotype mediated by small GTPase Rac1 in MCF10A human breast epithelial cells. Epidermal growth factor (EGF) plays an important role in aberrant growth and metastasis formation of many tumor types including breast cancer. The present study aims to investigate the correlation between EGF-induced invasiveness and Ras activation in four widely used breast cancer cell lines. Upon EGF stimulation, invasive abilities and H-Ras activation were significantly increased in Hs578T and MDA-MB-231 cell lines, but not in MDA-MB-453 and T47D cell lines. Using small interfering RNA (siRNA) to target H-Ras, we showed a crucial role of H-Ras in the invasive phenotype induced by EGF in Hs578T and MDA-MB-231 cells. Moreover, siRNA-knockdown of Rac1 significantly inhibited the EGF-induced invasiveness in these cells. Taken together, this study characterized human breast cancer cell lines with regard to the relationship between H-Ras activation and the invasive phenotype induced by EGF. Our data demonstrate that the activation of H-Ras and the downstream molecule Rac1 correlates with EGF-induced breast cancer cell invasion, providing important information on the regulation of malignant progression in mammary carcinoma cells.  相似文献

Leaf senescence in a stay-green mutant of Arabidopsis thaliana, ore10, was investigated during dark-incubation of its detached leaves. During this dark-induced senescence (DIS), Chl loss was delayed in ore10 mutants, as compared with wild type, but the rate of decline in the photochemical efficiency of PSII was not delayed in mutant leaves. After 2 d of DIS, native green gel electrophoresis of ore 10 leaf proteins resulted in a significant amount of pigment remaining as aggregates on top of the stacking gel. In addition, the accumulation of aggregates coincided with the emergence of a new band near 700 nm (F(699)) in the 77 K fluorescence emission spectrum of the aggregates. At 4 d, F(699) became a major band, both in the isolated aggregates and in intact leaves. Prolonged treatment with detergents revealed that light-harvesting complex II (LHCII) remaining after 2 d was highly stable, and the accumulation of aggregates coincided with the appearance of truncated LHCII in senescing ore10 leaves. These results suggest that increased LHCII stability is due to the formation of aggregates of trimmed LHCII. Thus, the LHCII protein degradation step that follows proteolysis of its terminal peptides is a possible lesion site of the ore10 mutant.  相似文献

Picea koraiensis, called Korean spruce, is an evergreen tree and found mostly in northeast Asia. In this study, plant regeneration via somatic embryogenesis from open-pollinated immature zygotic embryos of nine genotypes of elite trees was established. Immature zygotic embryos were cultured onto RJW medium modified from 505 medium with 21.48 μM NAA, 2.22 μM BA, and 2.32 μM KT. The average frequency for all nine genotypes was 74.2%. Embryogenic calluses of the nine genotypes of elite trees were subcultured on RJW basal medium containing 8.06 μM NAA, 1.11 μM BA, and 1.16 μM kinetin. The calluses of three lines, 3#, 9#, and 2#, were actively proliferated but others were not. Somatic embryogenesis was induced from the embryogenic callus in genotypes of 3#, 9#, and 2# on RJW medium with ABA and 60 g l−1 sucrose. Cotyledonary somatic embryos were subjected to a drying process. The drying of embryos by uncapping the culture bottle for 5 days on a clean bench resulted in a high frequency of germination of somatic embryos (87% in RJW medium). However, plantlet conversion from germinated embryos was greatly reduced and the optimal medium for plant conversion was 1/2 WPM or 1/2 BMI medium. In conclusion, we have, for the first time, established a plant regeneration system via somatic embryogenesis in the Korean spruce, which can be applied for rapid micropropagation of elite trees.  相似文献

The pollen morphology of subtribe Menthinae sensu Harley et al. [In: The families and genera of vascular plants VII. Flowering plants·dicotyledons: Lamiales (except Acanthaceae including Avicenniaceae). Springer, Berlin, pp 167–275, 2004] and two genera of uncertain subtribal affinities (Heterolamium and Melissa) are documented in order to complete our palynological overview of the tribe Mentheae. Menthinae pollen is small to medium in size (13–43 μm), oblate to prolate in shape and mostly hexacolpate (sometimes pentacolpate). Perforate, microreticulate or bireticulate exine ornamentation types were observed. The exine ornamentation of Menthinae is systematically highly informative particularly at generic level. The exine stratification in all taxa studied is characterized by unbranched columellae. Orbicules are consistently absent in Menthinae. Our palynological data are interpreted in a phylogenetic context at tribal level in order to assess the systematic value of pollen characters and to evaluate the existing molecular phylogenies for this group. Pollen morphology suggests Heterolamium as a close relative of subtribe Nepetinae and supports the molecular affinity of Melissa to subtribe Salviinae.  相似文献
