The seasonal variability of sediment–water ammonium flux andoxygen uptake was studied in an estuarine bay (Alfacs Bay, Ebro Delta, NWMediterranean) influenced by temporal freshwater discharges. Three stationswith different organic loading were sampled. The relationships of benthicfluxes to bottom water (temperature, dissolved oxygen, ammonium, nitrateplus nitrite) and to sediment (porosity, chlorophyll a derivative pigments,organic carbon and nitrogen) variables were examined. Oxygen uptake rangedfrom 0.3 to 2.5 mmol m–2 h–1 and ammoniumrelease ranged from 6 to 230 µmol m–2 h–1.The lowest value was recorded at the station furthest from the freshwaterinputs, and the highest was at the littoral station nearest the freshwaterdischarge channels (for oxygen uptake) and at the deep station at the saltwedge front (for ammonium flux). Water temperature and the concentration ofchlorophyll a derivative pigments on the surface sediment were revealed asthe main variables to be taken into account to explain the variabilityfound. Changes in fluxes reflecting temperature changes were found at thestation furthest from the freshwater inputs, while at the other, fluxvariability was found to be related to the cycle of functioning offreshwater discharge channels. The different patterns of variability arediscussed in relation to the dynamics of the estuary and to the mainfeatures of benthic nitrogen cycling.  相似文献

We investigated the heat tolerance of adults of three replicated lines of Drosophila melanogaster that have been evolving independently by laboratory natural selection for 15 yr at “nonextreme” temperatures (18°C, 25°C, or 28°C). These lines are known to have diverged in body size and in the thermal dependence of several life-history traits. Here we show that they differ also in tolerance of extreme high temperature as well as in induced thermotolerance (“heat hardening”). For example, the 28°C flies had the highest probability of surviving a heat shock, whereas the 18°C flies generally had the lowest probability. A short heat pretreatment increased the heat tolerance of the 18°C and 25°C lines, and the threshold temperature necessary to induce thermotolerance was lower for the 18°C line than for the 25°C line. However, neither heat pretreatment nor acclimation to different temperatures influenced heat tolerance of the 28°C line, suggesting the loss of capacity for induced thermotolerance and for acclimation. Thus, patterns of tolerance of extreme heat, of acclimation, and of induced thermotolerance have evolved as correlated responses to natural selection at nonextreme temperatures. A genetic analysis of heat tolerance of a representative replicate population each from the 18°C and 28°C lines indicates that chromosomes 1, 2, and 3 have significant effects on heat tolerance. However, the cytoplasm has little influence, contrary to findings in an earlier study of other stocks that had been evolving for 7 yr at 14°C versus 25°C. Because genes for heat stress proteins (hsps) are concentrated on chromosome 3, the potential role of hsps in the heat tolerance and of induced thermotolerance in these naturally selected lines is currently unclear. In any case, species of Drosophila possess considerable genetic variation in thermal sensitivity and thus have the potential to evolve rapidly in response to climate change; but predicting that response may be difficult.  相似文献

Polypeptide sequences encoded by some exons of the human tropoelastin gene (EDP, elastin-derived peptide) have been analysed for their ability to coacervate and to self-assembly. The great majority of them were shown to form organized structures, but only a few were indeed able to coacervate. Negative staining and rotary shadowing transmission electron microscopy showed the polypeptides to adopt a variety of supramolecular organization, from filaments, as those typical of tropoelastin, to amyloid-like fibers. The results obtained gave significant insight to the possible roles played by specific polypeptide sequences of tropoelastin.  相似文献

The 185delAG BRCA1 deletion occurs with a high frequency in Ashkenazi Jews. We detected this mutation in two Spanish Gypsy women (the only Gypsy participants) in an extensive study of 90 high-risk families and 160 women with early-onset breast cancer. One of these Gypsy women belonged to a high-risk family and the other had had early-onset breast cancer. The mutation was also detected in 1 out of 25 Gypsy samples unrelated to breast cancer. All the samples with the mutation shared the marker alleles present in Jewish samples with 185delAG. This is the first report of this mutation in a non-Jewish well-defined ethnic population. According to these findings the carrier frequency of this mutation in Gypsy individuals could be several times higher than that of the general population, and this should be taken into consideration in genetic screening for cancer in Gypsy populations. CK2alpha is the catalytic subunit of protein kinase CK2, an acidophilic and constitutively active eukaryotic Ser/Thr kinase involved in cell proliferation. A crystal structure, at 2.1 A resolution, of recombinant maize CK2alpha (rmCK2alpha) in the presence of ATP and Mg2+, shows the enzyme in an active conformation stabilized by interactions of the N-terminal region with the activation segment and with a cluster of basic residues known as the substrate recognition site. The close interaction between the N-terminal region and the activation segment is unique among known protein kinase structures and probably contributes to the constitutively active nature of CK2. The active centre is occupied by a partially disordered ATP molecule with the adenine base attached to a novel binding site of low specificity. This finding explains the observation that CK2, unlike other protein kinases, can use both ATP and GTP as phosphorylating agents.  相似文献

A thymidine incorporation technique was used to determine the tolerance of a soil bacterial community to Cu, Cd, Zn, Ni, and Pb. An agricultural soil was artificially contaminated in our laboratory with individual metals at three different concentrations, and the results were compared with the results obtained by using the plate count technique. Thymidine incorporation was found to be a simple and rapid method for measuring tolerance. Data obtained by this technique were very reproducible. A linear relationship was found between changes in community tolerance levels obtained by the thymidine incorporation and plate count techniques (r = 0.732, P < 0.001). An increase in tolerance to the metal added to soil was observed for the bacterial community obtained from each polluted soil compared with the community obtained from unpolluted soil. The only exception was when Pb was added; no indication of Pb tolerance was found. An increase in the tolerance to metals other than the metal originally added to soil was also observed, indicating that there was multiple heavy metal tolerance at the community level. Thus, Cu pollution, in addition to increasing tolerance to Cu, also induced tolerance to Zn, Cd, and Ni. Zn and Cd pollution increased community tolerance to all five metals. Ni amendment increased tolerance to Ni the most but also increased community tolerance to Zn and, to lesser degrees, increased community tolerance to Pb and Cd. In soils polluted with Pb increased tolerance to other metals was found in the following order: Ni > Cd > Zn > Cu. We found significant positive relationships between changes in Cd, Zn, and Pb tolerance and, to a lesser degree, between changes in Pb and Ni tolerance when all metals and amendment levels were compared. The magnitude of the increase in heavy metal tolerance was found to be linearly related to the logarithm of the metal concentration added to the soil. Threshold tolerance concentrations were estimated from these linear relationships, and changes in tolerance could be detected at levels of soil contamination similar to those reported previously to result in changes in the phospholipid fatty acid pattern (Å. Human granulocyte-macrophage colony-stimulating factor (GM-CSF) stimulates the proliferation and maturation of normal myeloid progenitor cells and can also stimulate the growth of acute myelogenous leukemia (AML) blasts. GM-CSF is not normally produced by resting cells but is expressed by a variety of activated cells including T lymphocytes, macrophages, and certain cytokine-stimulated fibroblasts and endothelial cells. Production of GM-CSF by cultured AML cells has been demonstrated, and GM-CSF expression by normal myeloid progenitors has been postulated to play a role in myelopoiesis. We have investigated the regulation of expression of GM-CSF in AML cell lines, and our results demonstrate the presence of a strong constitutive promoter element contained within 53 bp upstream of the cap site. We have also identified a negative regulatory element located immediately upstream of the positive regulatory element (within 69 bp of the cap site) that is active in AML cell lines but not T cells or K562 CML cells. Competition transfection and mobility shift studies demonstrate that this activity correlates with binding of a 45-kDa protein.  相似文献

A new technique with controlled interface generation allows separation and quantitation of enzyme inactivation by both solvent/aqueous interface and dissolved solvent. This has now been used in n-butanol, isopropylether, 2-octanone, n-hexane, n-butylbenzene, and n-tridecane. Ribonuclease was stable with all the solvent/aqueous interfaces studied. Chymotrypsin was mainly inactivated by the more hydrophobic solvent/aqueous interfaces, whereas lipase was only inactivated by the less hydrophobic solvent/aqueous interfaces. Urease was inactivated by some interfaces, but not all, without an obvious trend. Thus, the commonly expected simple relationship with solvent polarity (e.g., log P) does not apply when interfacial inactivation is determined specifically. Greater dissolved solvent inactivation occurred with the more polar solvents, though only a general trend was apparent with log P. A better correlation was noted with the Hilde-brand solubility parameter. Interfacial effects are discussed with reference to enzyme molecular weight, denaturation temperature, hydrophobicity, and adiabatic compressibility. (c) 1994 John Wiley & Sons, Inc.  相似文献

Abstract. To evaluate the effects of local competition on the growth and size variability of sprouts following disturbance in a natural population of Arbutus unedo in Catalonia, plants were cut at the base and a neighbor removal experiment was performed. Removal of neighbors resulted in an increase in the number and biomass of sprouts at 2 and 7.5 months after clipping. Number and biomass of sprouts was also correlated with initial plant size (stump area). Inter-genet competition appeared to be symmetric, and acted to delay the onset of interference among sprouts within a genet. Size variability of sprouts on an individual was positively correlated with their density on the stump, supporting the hypothesis that competition among ramets (sprouts) within a genet is asymmetric. Reduced inter-genet competition from neighbor removal resulted in an increase in the number and biomass of sprouts growing from a stump. This resulted in an increase in the asymmetry of competition among sprouts, and therefore an increase in the size variability of these sprouts.  相似文献
