Summary Transformation of plant tissues into crown gall tumors has been associated with the transfer of a portion of a tumor-inducing
plasmid (Ti-plasmid) into plant DNA. Various laboratories have regenerated normal-appearing plants from a number of crown
gall tumors. This study investigates the fate of the foreign DNA in a series of tissues derived from various parts of a plant
regenerated from the tumor BT-37 by Braun and his coworkers. It was found that all the foreign DNA sequences were lost from
tissues that had lost all their tumorous traits; whereas the plasmid DNA sequences were still present in tissues that appeared
normal but still exhibited tumorous traits when returned to tissue culture media. From these studies it would appear that
the presence of the Ti-plasmid sequences in the plant DNA is required for the maintenance of the transformed state.

Four dengue virus serotypes (DENV1-4) circulate globally, causing more human illness than any other arthropod-borne virus. Dengue can present as a range of clinical manifestations from undifferentiated fever to Dengue Fever to severe, life-threatening syndromes. However, most DENV infections are inapparent. Yet, little is known about determinants of inapparent versus symptomatic DENV infection outcome. Here, we analyzed over 2,000 DENV infections from 2004 to 2011 in a prospective pediatric cohort study in Managua, Nicaragua. Symptomatic cases were captured at the study health center, and paired healthy annual samples were examined on a yearly basis using serological methods to identify inapparent DENV infections. Overall, inapparent and symptomatic DENV infections were equally distributed by sex. The mean age of infection was 1.2 years higher for symptomatic DENV infections as compared to inapparent infections. Although inapparent versus symptomatic outcome did not differ by infection number (first, second or third/post-second DENV infections), substantial variation in the proportion of symptomatic DENV infections among all DENV infections was observed across study years. In participants with repeat DENV infections, the time interval between a first inapparent DENV infection and a second inapparent infection was significantly shorter than the interval between a first inapparent and a second symptomatic infection. This difference was not observed in subsequent infections. This result was confirmed using two different serological techniques that measure total anti-DENV antibodies and serotype-specific neutralizing antibodies, respectively. Taken together, these findings show that, in this study, age, study year and time interval between consecutive DENV infections influence inapparent versus symptomatic infection outcome, while sex and infection number had no significant effect. Moreover, these results suggest that the window of cross-protection induced by a first infection with DENV against a second symptomatic infection is approximately 2 years. An increasing number of human cases of gnathostomiasis have been reported in Sinaloa, Mexico, most of whom have a custom of eating of raw fish dishes such as 'cebiche'. Here we report five adult patients, three women and two men, having an acute episode of vomiting and abdominal pain a few minutes after eating a dish of cebiche prepared from a spotted sleeper perch (Eleotris picta) fished from a nearby lake in southern Sinaloa. All five patients experienced acute throat pain, chest and joint pains, headache and fever. One patient, a 55-year-old male, was hospitalized with suspected pancreatitis and pneumonia. By 8-9 days later, all five patients developed between three and 12 edematous, migrating skin lesions on the back, abdomen, upper and lower extremities, face, eye and scalp. By ELISA, all of them were sero-positive to Gnathostoma doloresi antigen and had elevated IgE levels. Eosinophilia was found in two patients. These patients lived in an agricultural and fishing community. In this community we carried out a sero-epidemiological survey and study of living conditions in a random sample of 309 individuals distributed in 74 households. Frequent consumption of raw fish was reported in 36% of households, and 12 individuals had a clinical history of migrating skin lesions. The sero-prevalence to Gnathostoma antigens was 34.95%. Five fish species and four species of ichthyophagous birds collected from three lakes in the village and a nearby estuary were infected with the advanced third-stage larvae of G. binucleatum, a species found in Ecuador and Mexico. The results describe the first known outbreak of acute gnathostomiasis on the American continent. 相似文献

We evaluated under semi-field conditions the intrinsic competition between Diachasmimorpha longicaudata (Ashmead), as an invasive parasitoid, and Doryctobracon areolatus (Szépligeti) and Utetes anastrephae (Viereck) (all Hymenoptera: Braconidae) as resident parasitoids, as well as that between U. anastrephae as an invader and D. areolatus as a resident. The percentage of live D. areolatus larvae decreased by 39.8% and 29.4% following attack by D. longicaudata and U. anastrephae as invasive parasitoids, respectively. Likewise, the parasitism percentage of D. areolatus decreased by 31.5% and 60.8% under competition with the invasive parasitoids D. longicaudata and U. anastrephae, respectively. Parasitism by D. longicaudata decreased by 44.6% and 41.6% in the presence of the residents D. areolatus and U. anastrephae, respectively, while parasitism of U. anastrephae was only affected when this species was a resident. We concluded that D. areolatus is an inferior intrinsic competitor and that U. anastrephae resists the competitive presence of D. longicaudata. 相似文献

Classifying glycerol dehydratase by its functional residues and purifying selection in its evolution
Glycerol dehydratase (GD) catalyses glycerol reductive conversion to 3-hydroxypropanaldehyde (3-HPA), this being the first step required for the microbial conversion of glycerol to 1, 3 -propanodiol. GD has been functionally characterised to date and two main groups have been determined, one of them being vitamin B(12)-dependent and the other B(12)-independent. GD evolutionary history has been described and an exhaustive analysis made for detecting the functional residues responsible for type I divergence. GD phylogenetic tree topology was seen to be statistically robust and the data indicated strong purifying selection operating on the GD proteins within it. Two clades were indentified, one for vitamin B(12)-dependent and the other for B(12)- independent classes. The ancient hot-pot residues responsible for protein divergency for each clade were also identified. The basic evolutionary biology for GD proteins has been described, thereby opening the way forward for developing rational mutagenesis studies. 相似文献
