The frequency of recovery of atypical mycobacteria was estimated in two treatment plants providing drinking water to Paris, France, at some intermediate stages of treatment. The two plants use two different filtration processes, rapid and slow sand filtration. Our results suggest that slow sand filtration is more efficient for removing mycobacteria than rapid sand filtration. In addition, our results show that mycobacteria can colonize and grow on granular activated carbon and are able to enter distribution systems. We also investigated the frequency of recovery of mycobacteria in the water distribution system of Paris (outside buildings). The mycobacterial species isolated from the Paris drinking water distribution system are different from those isolated from the water leaving the treatment plants. Saprophytic mycobacteria (present in 41.3% of positive samples), potentially pathogenic mycobacteria (16.3%), and unidentifiable mycobacteria (54.8%) were isolated from 12 sites within the Paris water distribution system. Mycobacterium gordonae was preferentially recovered from treated surface water, whereas Mycobacterium nonchromogenicum was preferentially recovered from groundwater. No significant correlations were found among the presence of mycobacteria, the origin of water, and water temperature.  相似文献

An endo-beta-mannanase (EC 3.2.1.78) from Streptomyces ipomoea CECT 3341 was purified and applied to the biobleaching of pine kraft pulps. The maximum level of endo-beta-mannanase activity (0.6 units ml(-1)) was achieved after 4 days of growth in a medium containing locust bean gum and yeast extract. Zymograms revealed mannanase bands (Man) with high and low electrophoretic mobility on the second and seventh days of incubation (Man1, Man3) and three bands of high, medium and low mobility from the third to sixth days of growth (Man1, Man2, Man3). Although these exhibited different molecular masses, their amino-terminal sequences were identical. The action of proteases detected in the culture supernatant could be responsible for such events, suggesting that only one endo-beta-mannanase is produced by S. ipomoea. The purified Man3 exhibited a molecular mass of 40 kDa, an isoelectric point of 4.0 and an optimal temperature and pH reaction of 55 degrees C and 7.5, respectively. It was strongly inhibited by Ag+, Hg2+, Al3+ and Fe3+, and was strongly activated by Mn2+. The ability of the purified endo-beta-mannanase to improve the bleachability of pine kraft pulp, when applied with alkaline extraction, was demonstrated by an increase in the pulp brightness (1.7%, using the International Standards Organisation's test) and an absence of variations in the viscosity values. A relationship between the increase in pulp brightness and the presence of manganese in the pulps could be established.  相似文献

We studied the resistance of various mycobacteria isolated from a water distribution system to chlorine. Chlorine disinfection efficiency is expressed as the coefficient of lethality (liters per minute per milligram) as follows: Mycobacterium fortuitum (0.02) > M. chelonae (0.03) > M. gordonae (0.09) > M. aurum (0.19). For a C·t value (product of the disinfectant concentration and contact time) of 60 mg·min·liter−1, frequently used in water treatment lines, chlorine disinfection inactivates over 4 log units of M. gordonae and 1.5 log units of M. fortuitum or M. chelonae. C·t values determined under similar conditions show that even the most susceptible species, M. aurum and M. gordonae, are 100 and 330 times more resistant to chlorine than Escherichia coli. We also investigated the effects of different parameters (medium, pH, and temperature) on chlorine disinfection in a chlorine-resistant M. gordonae model. Our experimental results follow the Arrhenius equation, allowing the inactivation rate to be predicted at different temperatures. Our results show that M. gordonae is more resistant to chlorine in low-nutrient media, such as those encountered in water, and that an increase in temperature (from 4°C to 25°C) and a decrease in pH result in better inactivation.  相似文献

The existence of seasonal changes in concentrations of water-soluble carbohydrates in arthropods (both freezing-tolerant and intolerant species) from Signy Island was demonstrated. Seasonal patterns of variation, imposed by seasonality of the maritime Antarctic environment, in the production of soluble carbohydrates in response to low temperatures and/or dehydration for a range of terrestrial arthropods were confirmed. The freshwater copepod Pseudoboeckellapoppei exhibited much lower levels of soluble carbohydrates, with glycerol as the main component, and smaller seasonal fluctuations relative to the four terrestrial species. The two Antarctic mites (Alaskozetes antarcticus and Gamasellus racovitzai) accumulated glycerol (as a single-component cryoprotective system), in agreement with previous work reporting increased glycerol levels and lowering of the supercooling point in A. antarcticus. In the case of G. racovitzai, increased levels of glycerol may function in a different manner. The larval dipteran Eretmoptera murphyi and the collembolan Cryptopygus antarcticus have complex multi-component cryoprotective systems involving trehalose that may be related to low temperature acclimation and dehydration. These findings are discussed in relation to published work on single and multiple cryoprotective systems, supercooling points and the involvement of dehydration as a complementary stress in overwintering insects. 我们用玉米黑粉菌(Ustilago maydis)热休克基因(Heat shock gene,hsp 70)的启动子和终止子与新霉素磷酸转移酶基因相连,构建成了有效的双功能质粒pDL1(在E。coli中用Amp作为选择标记,在玉米黑粉菌中用新霉素作为选择标记)。以分离的一株新霉素敏感的玉米黑粉菌作为受体菌,用构建的pDL1质粒作为供体DNA,对影响受体菌原生质球的形成条件(培养基、菌龄、各种酶、酶的浓度、作用时间和渗透压稳定剂)、再生条件和DNA转化条件进行了初步研究。对数前中期收集的菌体,以5mg/ml真菌溶壁酶处理30分钟左右,90%都形成原生质球,其再生率为60—80%,转化率为300—1000转化子/μg DNA。随机选出25个转化子,DNA杂交都为阳性。分析dot-blot和Southern blot DNA杂交结果,发现质粒在细胞中以整合形式存在,一个细胞可以有多拷贝的整合质粒,质粒可能以非完全同源性的重组形式,参入性地整合进染色体中。  相似文献

The use of ancient DNA techniques in human studies has been hampered by problems of contamination with modern human DNA. The main problem has been that the object of study belongs to the same species as the observer, and the complete elimination of the contamination risk is seemingly unlikely. Contamination has even been detected in the most specialized laboratories in this field. In these kinds of studies it is therefore very important to detect contamination and to distinguish contaminants from authentic results. Here, we report the use of a strategy to authenticate the identity of ancient mitochondrial DNA (mtDNA), based on the previously established relationship between D-loop sequence substitutions and haplogroup-specific restriction site changes. Forty-four individuals from a 16th-century necropolis were analyzed, from which 28 control region sequences were obtained. These sequences were preclassified into haplogroups, according to the observed motifs. Subsequently, the DNA extracts from which the sequences were obtained, along with independent extracts of subsets of the same individuals, were subjected to restriction fragment length polymorphism (RFLP) analysis to compare and corroborate the results. Using this approach, 24 sequences were authenticated, while two were discarded because of result mismatches. The final distribution of the haplogroups in the sample, and the differences in the sequences, are two additional criteria of authentication.  相似文献

The yeast Debaryomyces hansenii is usually found in salty environments such as the sea and salted food. It is capable of accumulating sodium without being intoxicated even when potassium is present at low concentration in the environment. In addition, sodium improves growth and protects D. hansenii in the presence of additional stress factors such as high temperature and extreme pH. An array of advantageous factors, as compared with Saccharomyces cerevisiae, is putatively involved in the increased halotolerance of D. hansenii: glycerol, the main compatible solute, is kept inside the cell by an active glycerol-Na+ symporter; potassium uptake is not inhibited by sodium; sodium protein targets in D. hansenii seem to be more resistant. The whole genome of D. hansenii has been sequenced and is now available at http://cbi.labri.fr/Genolevures/ and, so far, no genes specifically responsible for the halotolerant behaviour of D. hansenii have been found.  相似文献

Respiratory burst oxidase homologs (RBOHs) catalyze the reduction of oxygen to generate superoxide anion, a kind of reactive oxygen species (ROS). The ROS produced by RBOHs play essential roles in diverse processes, such as root hair development, stomata closure and signaling mechanisms in response to abiotic stimuli and during plant-pathogen interactions. Recently, we found that PvRbohB silencing in transgenic Phaseolus vulgaris roots had a negative impact on lateral root density. In this work, we show that the downregulation of PvRbohB affects both the growth and ROS levels in recently emerged lateral roots. In addition, we found that the PvRbohB promoter was activated during lateral root primordium initiation in the pericycle, and remained active throughout lateral root development. This study identifies RBOHs as potentially important players in lateral root development in P. vulgaris.  相似文献

Nuclear insertions of mitochondrial origin (NUMTs) can be useful tools in evolution and population studies. However, due to their similarity to mitochondrial DNA (mtDNA), NUMTs may also be a source of contamination in mtDNA studies. The main goal of this work is to present a database of NUMTs, based on the latest version of the human genome—GRCh37 draft. A total of 755 insertions were identified. There are 33 paralogous sequences with over 80% sequence similarity and of a greater length than 500 bp. The non-identical positions between paralogous sequences are listed for the first time. As an application example, the described database is used to evaluate the impact of NUMT contamination in cancer studies. The evaluation reveals that 220 positions from 256 with zero hits in the current mtDNA phylogeny could in fact be traced to one or more nuclear insertions of mtDNA. This is due to they are located in non-identical positions between mtDNA and nuclear DNA (nDNA). After in silico primer validation of each revised cancer study, risk of co-amplification between mtDNA and nDNA was detected in some cases, whereas in others no risk of amplification was identified. This approach to cancer studies clearly proves the potential of our NUMT database as a valuable new tool to validate mtDNA mutations described in different contexts. Moreover, due to the amount of information provided for each nuclear insertion, this database should play an important role in designing evolutionary, phylogenetic and epidemiological studies.  相似文献
