Migration of IgA-bearing lymphocytes into salivary glands 总被引:4,自引:0,他引:4
The relative migratory properties of [125I]iododeoxyuridine-labeled dividing mesenteric lymph node (MLN) and peripheral lymph node (PLN) cells were assessed in mice using the adoptive transfer system. MLN cells from virgin donors showed a greater tendency to localize in MLN, small intestine, and mammary glands (lactating recipients only) of virgin and lactating recipients. In addition, MLN cells were shown to selectively localize in the salivary (submandibular and sublingual) glands. The relative migratory properties of MLN and PLN cell populations were identical when donor cells were obtained from virgin or lactating animals. Selective depletion of IgA-bearing cells from the MLN transfer cell population abrogated the preferential localization in control mucosal tissues and salivary glands. These data show that the salivary glands can be included as an additional mucosal area populated by gut-derived IgA-bearing cells and provides direct evidence that the common mucosal immune sytstem extends to secretory tissue in the oral cavity. 相似文献

Netrins, axon guidance cues in the CNS, have also been detected in epithelial tissues. In this study, using the embryonic pancreas as a model system, we show that Netrin-1 is expressed in a discrete population of epithelial cells, localizes to basal membranes, and specifically associates with elements of the extracellular matrix. We demonstrate that alpha6beta4 integrin mediates pancreatic epithelial cell adhesion to Netrin-1, whereas recruitment of alpha6beta4 and alpha3beta1 regulate the migration of CK19+/PDX1+ putative pancreatic progenitors on Netrin-1. These results provide evidence for the activation of epithelial cell adhesion and migration by a neural chemoattractant, and identify Netrin-1/integrin interactions as adhesive/guidance cues for epithelial cells. 相似文献

The morphogenesis of cells dissociated from fetal rat intestine at 18 days of gestation was compared in vitro on two different substrates, tissue culture plastic and collagen gel. During the first 2 days in culture growth on both substrates was similar, resulting in the formation of layers of epithelial cells and of small lumina within them. On plastic, the cell layers contracted over time, resulting in the formation of small mounds of cells bearing on their surface small protrusions covered with epithelial cells which had densely packed microvilli on their apical surfaces. When the cultures on collagen gels were overlaid with more collagen gel, vesicles lined with epithelial cells developed. These cells were joined by junctional complexes and displayed an apical brush border which was periodic acid-Schiff and alkaline phosphatase positive. After 1 week in culture, when the vesicles reached their maximum extent, numerous epithelial cells were actively incorporating labeled thymidine and cells were being extruded into the lumen. These results demonstrate that the dissociated intestinal cells have the capacity to form intestine-like organoids in vitro and that surrounding the cells with the collagen gel allows expression of this potential. The collagen gel cultures thus undergo morphogenetic processes which can be modified by the surrounding environment and should provide a useful in vitro model system for the study of intestinal development. 相似文献

Microsatellite variation was analyzed in five Chinese indigenous goat breeds, which include four Cashmere breeds (Tibetan, Neimonggol, Liaoning, Taihang) and one Hubei local breed (Matou) used for meat production. Five ovine and one bovine microsatellites, selected from eight ovine microsatellites and five bovine microsatellites were suitable for use in this study. With these six microsatellites, allele frequencies, heterozygosity, polymorphism information content (PIC) and effective allele number were calculated. A neighbor-joining tree was constructed using Nei's standard genetic distance (1978). In the tree, Neimonggol and Liaoning were grouped together, then with Taihang; while Tibetan and Matou individually had their own branch. The genetic relationship of five breeds corresponds to their history and geographic origins. 相似文献

The dopamine transporter (DAT) is a crucial regulator of dopaminergic neurotransmission, controlling the length and brevity of dopaminergic signaling. DAT is also the primary target of psychostimulant drugs such as cocaine and amphetamines. Conversely, methylphenidate and amphetamine are both used clinically in the treatment of attention-deficit hyperactivity disorder and narcolepsy. The action of amphetamines, which induce transport reversal, relies primarily on the ionic composition of the intra- and extracellular milieus. Recent findings suggest that DAT interacting proteins may also play a significant role in the modulation of reverse dopamine transport. The pharmacological inhibition of the serine/threonine kinase αCaMKII attenuates amphetamine-triggered DAT-mediated 1-methyl-4-phenylpyridinium (MPP(+)) efflux. More importantly, αCaMKII has also been shown to bind DAT in vitro and is therefore believed to be an important player within the DAT interactome. Herein, we show that αCaMKII co-immunoprecipitates with DAT in mouse striatal synaptosomes. Mice, which lack αCaMKII or which express a permanently self-inhibited αCaMKII (αCaMKII(T305D)), exhibit significantly reduced amphetamine-triggered DAT-mediated MPP(+) efflux. Additionally, we investigated mice that mimic a neurogenetic disease known as Angelman syndrome. These mice possess reduced αCaMKII activity. Angelman syndrome mice demonstrated an impaired DAT efflux function, which was comparable with that of the αCaMKII mutant mice, indicating that DAT-mediated dopaminergic signaling is affected in Angelman syndrome. 相似文献

A mathematical foundation is developed for the computation of pressures exerted on portions of the anatomy constrained by elastic fabric support. General formulae are derived for curvatures of an ellipsoidal model corresponding to orthogonal tension components at the point of measurement. It is found that the pressure contributions from both tension components are significant to first order in the longitudinal variation of circumference over an axial test zone, because while the transverse curvature approaches the zeroth order cylindrical approximation, its longitudinal counterpart does not. 相似文献

Representative energy data for a human population can be produced by combining randomly sampled time allocation observations with activity-specific energy expenditure measurements. Research to produce representative energy data for adults of a population of Machiguenga Indians has recently been conducted in lowland, southeastern Peru. Marked contrast was found between the sexes for average married adults in energy expended on an average day. Men spent about 3,200 kcals and women, about 1,925; ratio: 1.66 to 1. In general, men tended to work at somewhat more energetic activities and for longer periods than did women. In addition to sex-role-related task differences were contrasts in uses of technological items and in respective work settings. These representative behavior data permit direct estimates of population-level energy requirements for average days, seasons, or for 1 year. 相似文献

Dihydrotestosterone glucuronide (DHTG), a series of 5 alpha-bile acids, or allo-bile acids (3 alpha-hydroxy-5 alpha-cholanic acid, 3-keto-5 alpha-cholanic acid and 3 beta-hydroxy-5 alpha-cholanic acid) and their normal bile acid analogues (3 alpha-hydroxy-5 beta-cholanic acid or lithocholate, 3-keto-5 beta-cholanic acid and 3 beta-hydroxy-5 beta-cholanic acid) were administered intravenously to female rats in order to determine their effects on bile flow. All agents caused a rapid and profound inhibition of bile flow which was dose-dependent. The logarithm of the dose vs the cholestatic response curve for DHTG, the allo-bile acids and lithocholate were all parallel. DHTG was the most potent congener and was two times more potent than 3-keto-5 alpha-cholanic acid and 5 times more potent than lithocholate. These data indicate that the glucuronic acid moiety and the trans configuration of the A and B rings of the steroid nucleus confer the greatest cholestatic potency. 相似文献

A system for culturing human fetal liver cells in monolayers is described and the effects of various conditions of growth on the morphology and function of the cultured cells are presented. The addition of 10% calf serum or 1% human serum to the growth medium accelerated the proliferation of the liver cells, with subsequent loss of characteristic morphology and specific functional activity. In the absence of serum, the cultured liver cells retained their morphology and their function for at least 4 wk, as evidenced by secretion of serum albumin and storage of glycogen and iron. 相似文献
