Zusammenfassung Bei den Teleostiern Mugil und Gobius wurden die Veränderungen des Zwischenhirn-Hypophysensystems in veränderten Außenmedien (hypotonisches Seewasser) histologisch untersucht.Beim langsamen Überführen aus Seewasser von 38,4 S in Seewasser von 25,4 und 20,8 S nahm das Sekret nicht, wie erwartet, zu. Im Nucleus praeopticus und im Hinterlappen der Hypophyse erfolgt eine starke Verringerung des Sekretbestandes (Mugil). Erst in Seewasser von weit niedrigerem Salzgehalt (6,3 und 1, 4) nimmt das Sekret im Hypophysenhinterlappen wieder zu (Gobius). Bei 6 S ist eine intensive Sekretableitung im N.p. und Tractus praeopticohypophyseus zu beobachten. Eine Deutung der Ergebnisse wird versucht, indem das weitgehend konstante Innenmedium mit den wechselnden Außenfaktoren in Beziehung gebracht wird.Die Befunde weisen darauf hin, daß das Zwischenhirn-Hypophysensystem auch bei Teleostiern an der Regulation des Wasserhaushaltes beteiligt ist.Der Deutschen Forschungsgemeinschaft danke ich für die Gewährung eines Stipendiums, das mir den Aufenthalt in Neapel ermöglichte. Zusammenfassung In einleitenden terminologischen Erwägungen wird vorgeschlagen, den Ausdruck Schlüpfrhythmus zugunsten von tageszeitlich gebundenem Schlüpfen aufzugeben. Pseudosmittia arenaria zeigt im normalen Tag-Nachtwechsel ein deutliches tageszeitlich gebundenes Schlüpfen mit einem Maximum 6–8h nach DL (Beginn der Beleuchtung).In Dauerdunkel schlüpfen keine Imagines. In Dauerlicht erscheinen die Tiere gleichmäßig über den ganzen Tag verteilt.Der ganze Bereich des sichtbaren Lichtes (geprüft von 476–641 m) ist wirksam. Auch die Lichtintensität in Licht-Dunkelbedingungen spielt im untersuchten Bereich (18–350 Lux) keine Rolle.Das Schlüpfmaximum von Ps. arenaria zeigt eine deutliche Beziehung zu DL. Es wandert bei konstanter Tageslänge um so dichter an DL heran, je kürzer die relative Länge der Lichtzeit ist.Bei kürzeren Tageslängen wandert das Maximum von DL fort, bei längeren an DL heran. Bei Tagen, die kürzer als 18h sind, liegt es in der folgenden Dunkelzeit, bei solchen, die länger als 36h sind, vor DL. Sein Weg beschreibt dabei eine kubische Parabel.Beim Eintritt des Gipfels in die folgende Dunkelzeit erscheint ein Maximum nur an jedem 2. Tag. Ein Maximum vor DL ruft ein zusätzliches Maximum in der gleichen Entfernung von LD (Beginn der Dunkelzeit) hervor. Bei extremen Tageslängen ist also dennoch ein Maximum ungefähr alle 24h zu beobachten. Dies ist exogen bedingt. Verlagert man durch schwache Beleuchtung während der Dunkelzeit das Maximum im 12h-Tag in die Lichtzeit, so erhält man ein Maximum an jedem Tag. Der Eintritt des Maximums in eine Dunkelzeit hat also das Umspringen auf einen Schlüpfgipfel an jedem 2. Tag bzw. auf 2 je Tag zur Folge.Im 12h-Tag mit absoluter Dunkelzeit während der Dunkelzeit kann man auch ein Maximum alle 12h erreichen, und zwar durch Umstimmung eines Teils der Population. Man hat dann 2 Gruppen, deren Maxima um 12h gegeneinander verschoben sind. Hemmend und fördernd auf den Einfluß des Lichtes wirken Temperatur und Substratfeuchtigkeit. Beide Faktoren können ohne Licht kein Schlüpfen hervorrufen. Beziehungen zwischen Lebensweise, Ökologie oder systematischer Stellung und tageszeitlich gebundenem Schlüpfen lassen sich bisher nicht feststellen. 相似文献

Through use of commercially available DnaK proteins and anti-DnaK monoclonal antibodies, a competitive enzyme-linked immunosorbent assay was developed to quantify this heat shock protein in Escherichia coli ATCC 25922 subjected to various heating regimens. For a given process lethality (F7010 of 1, 3, and 5 min), the intracellular concentration of DnaK in E. coli varied with the heating temperature (50 or 55°C). In fact, the highest DnaK concentrations were found after treatments at the lower temperature (50°C) applied for a longer time. Residual DnaK after heating was found to be necessary for cell recovery, and additional DnaK was produced during the recovery process. Overall, higher intracellular concentrations of DnaK tended to enhance cell resistance to a subsequent lethal stress. Indeed, E. coli cells that had undergone a sublethal heat shock (105 min at 55°C, F7010 = 3 min) accompanied by a 12-h recovery (containing 76,786 ± 25,230 molecules/cell) resisted better than exponentially growing cells (38,500 ± 6,056 molecules/cell) when later heated to 60°C for 50 min (F7010 = 5 min). Results reported here suggest that using stress protein to determine cell adaptation and survival, rather than cell counts alone, may lead to more efficient heat treatment. 相似文献

To investigate the physiological roles of translation initiation factor IF3 and ribosomal protein L20 inEscherichia coli, theinfC, rpmI andrpIT genes encoding IF3, L35 and L20, respectively, were placed under the control oflac promoter/operator sequences. Thus, their expression is dependent upon the amount of inducer isopropyl thiogalactoside (IPTG) in the medium. Lysogenic strains were constructed with recombinant lambda phages that express eitherrpmI andrplT orinfC andrpmI in trans, thereby allowing depletion of only IF3 or L20 at low IPTG concentrations. At low IPTG concentrations in the IF3-limited strain, the cellular concentration of IF3, but not L20, decreases and the growth rate slows. Furthermore, ribosomes run off polysomes, indicating that IF3 functions during the initiation phase of protein synthesis in vivo. During slow growth, the ratio of RNA to protein increases rather than decreases as occurs with control strains, indicating that IF3 limitation disrupts feedback inhibition of rRNA synthesis. As IF3 levels drop, expression from an AUU-infC-lacZ fusion increases, whereas expression decreases from an AUG-infC-lacZ fusion, thereby confirming the model of autogenous regulation ofinfC. The effects of L20 limitation are similar; cells grown in low concentrations of IPTG exhibited a decrease in the rate of growth, a decrease in cellular L20 concentration, no change in IF3 concentration, and a small increase in the ratio of RNA to protein. In addition, a decrease in 50S subunits and the appearance of an aberrant ribosome peak at approximately 41–43S is seen. Previous studies have shown that the L20 protein negatively controls its own gene expression. Reduction of the cellular concentration of L20 derepresses the expression of anrplT-lacZ gene fusion, thus confirming autogenous regulation by L20. 相似文献

Palynological studies on late Quaternary lake sediments from the region of the Amazon estuary, 100 km north-east of Belém, Pará State, Brazil, enable reconstruction of lowland Amazonian rain forest during the Late-glacial and Holocene periods. Late-glacial forests included populations of Podocarpus which suggests a distinct climatic cooling. Ilex was abundant in the early Holocene. Records of the mangrove taxon, Rhizophora, indicate rapid Atlantic sea-level rise in the beginning of the Holocene. High charcoal representation may reflect the first arrival of Amerindians in the Amazon coastal area, probably about 10 800 B.P. 相似文献

Nearly 7000 Arabidopsis thaliana -expressed sequence tags (ESTs) from 10 cDNA libraries have been sequenced, of which almost 5000 non-redundant tags have been submitted to the EMBL data bank. The quality of the cDNA libraries used is analysed. Similarity searches in international protein data banks have allowed the detection of significant similarities to a wide range of proteins from many organisms. Alignment with ESTs from the rice systematic sequencing project has allowed the detection of amino acid motifs which are conserved between the two organisms, thus identifying tags to genes encoding highly conserved proteins. These genes are candidates for a common framework in genome mapping projects in different plants. 相似文献

Abstract: Using a cell culture model of the blood-brain barrier (BBB), we investigated the brain capillary endothelial cell (EC) response to hypoxia. The activities of antioxidant enzymes such as glutathione peroxidase, glutathione reductase, catalase, and superoxide dismutase and the GSH level of brain capillary ECs alone or in coculture with astrocytes, as well as those of pericytes, were compared with those obtained with freshly isolated microvessels. These results demonstrated that brain capillary ECs cocultured with astrocytes and used in the presence of a coculture-conditioned medium provided a relevant in vitro model for studying the effect of hypoxia-reoxygenation at the BBB level. The effect of hypoxia on antioxidant enzymes, GSH, and ATP levels was studied, as well as the modification of the permeability to small weight molecules. A decrease in all enzymes and the GSH level could explain an increase in the susceptibility of the brain capillary ECs to further oxidant injury. Second, profound rearrangements of F-actin filaments of the ECs and a decrease in the ATP level could be associated with an increase in the permeability of the monolayer. Furthermore, an apoptotic process was detected by in situ end labeling of DNA. These results indicate that hypoxia distorts the function of ECs and that these cells in culture provide a valuable tool for exploring mechanisms after hypoxia-reoxygenation. 相似文献

Gene rearrangements in Chlamydomonas chloroplast DNAs are accounted for by inversions and by the expansion/contraction of the inverted repeat 总被引:1,自引:0,他引:1
To gain insight into the mutational events responsible for the extensive variation of chloroplast DNA (cpDNA) within the green algal genus Chlamydomonas, we have investigated the chloroplast gene organization of Chlamydomonas pitschmannii, a close relative of the interfertile species C. eugametos and C. moewusii whose cpDNAs have been well characterized. At 187 kb, the circular cpDNA of C. pitschmannii is the smallest Chlamydomonas cpDNA yet reported; it is 56 and 105 kb smaller than those of its C. eugametos and C. moewusii counterparts, respectively. Despite this substantial size difference, the arrangement of 77 genes on the C. pitschmannii cpDNA displays only three noticeable differences from the organization of the corresponding genes on the collinear C. eugametos and C. moewusii cpDNAs. These changes in gene order are accounted for by the expansion/contraction of the inverted repeat and one or two inversions in a single-copy region. In land plant cpDNAs, these kinds of events are also responsible for gene rearrangements. The large size difference between the C. pitschmannii and C. eugametos/C. moewusii cpDNAs is mainly attributed to multiple events of deletions/additions as opposed to the usually observed expansion/contraction of the inverted repeat in land plant cpDNAs. We also found that the mitochondrial genome of C. pitschmannii is a circular DNA molecule of 16.5 kb which is 5.5 and 7.5 kb smaller than its C. moewusii and C. eugametos counterparts, respectively. 相似文献
