Myxococcus xanthus RppA and MmrA are homologous to methyl-accepting chemotaxis proteins (MCPs) and to multidrug transporters, respectively. We reported previously that rppA-mmrA double mutant exhibited reduced colony expansion, agglutination, and polysaccharide levels. We have demonstrated here that the rppA-mmrA mutant also exhibited reduced amino acid uptake. Furthermore, the double mutant appeared to be more susceptible to some antimicrobial agents, such as streptomycin, ethidium bromide and norfloxacin, than the wild-type. These phenotypes were not shown in the rppA or mmrA single mutant. These results indicate that M. xanthus RppA and MmrA are also involved in the uptake of amino acids and efflux of some antimicrobial agents.  相似文献

CD4(+)CD25(+) T cells have immunoregulatory and suppressive functions and are responsible for suppressing self-reactive cells and maintaining self-tolerance. In addition to CD4(+)CD25(+) T cells, there is some evidence that a fraction of CD4(+)CD25(-) T cells exhibit suppressive activity in vitro or in vivo. We have shown, using aged mice, that aging not only leads to a decline in the ability to mount CD4(+)CD25(-) T cell responses, but, at the same time, renders aged CD4(+)CD25(-) T cells suppressive. In this study we report two newly established mAbs that could abrogate the suppressive function of aged CD4(+)CD25(-) T cells. These mAbs recognized the same protein, the transmembrane phosphatase CD45. Cross-linking of CD45 on aged CD4(+)CD25(-) T cells was required for the disruption of their suppressive activity. Surprisingly, these mAbs also abrogated the suppressive action of CD4(+)CD25(+) T cells in vitro. Our results demonstrate an unexpected function of CD45 as a negative regulator neutralizing the suppressive activity of aged CD4(+)CD25(-) and young CD4(+)CD25(+) T cells.  相似文献

We have shown that a 4-phenyl-1,2,3,6-tetrahydropyridine fragment plays an important role in improving inhibitory potency against poly(ADP-ribose) polymerase-1 (PARP-1). Various benzamide analogues linked with this fragment via alkyl spacers have been prepared and evaluated. As a result, some of them have been found to be highly potent PARP-1 inhibitors.  相似文献

A new species of chlorarachniophyte alga, Lotharella vacuolata Ota et Ishida sp. nov., is described. This alga has been maintained as strain CCMP240 at the Provasoli‐Guillard National Center for Culture of Marine Phytoplankton at Bigelow Laboratory for Ocean Sciences. We examined in detail its morphology, ultrastructure and life cycle, using light microscopy, transmission electron microscopy and time‐lapse videomicroscopy. The dominant stage in the life cycle was represented by coccoid cells; however, amoeboid and flagellated stages were also observed. This alga showed unique post‐cell division behavior: one of the two daughter cells became amoeboid and escaped through a pore on the parental cell wall; the other daughter cell remained within the parental cell wall. Pyrenoid ultrastructure and nucleomorph location, which are used as the main generic criteria of chlorarachniophytes, confirmed that the strain CCMP240 is a member of Lotharella. This alga, however, was clearly distinguished from other known Lotharella species by the presence of large vacuoles, unusual post‐cell division behavior and some unique ultrastructural characters.  相似文献

Intraspecific variation among 36 strains of the freshwater alga Gonium pectorale Müller (Chlorophyceae) isolated from three geographically different locations in Tibet, Nepal, and Japan was investigated by isozyme analysis. Variation in isozyme patterns of eight enzyme systems (malate dehydrogenase, glutamate dehydrogenase, tetrazolium oxidase, lactate dehydrogenase, octanol dehydrogenase, xanthine dehydrogenase, phosphoglucomutase, and malic enzyme) of axenic and clonal cultures was revealed by polyacrylamide gel electrophoresis. Unweighted average linkage clustering, based on Jaccard's similarity coefficient, illustrated the high similarity between most strains from Nepal and all strains from the Ryukyu Islands (Japan). However, there was relatively low similarity between strains from Tibet and those from Nepal and Japan. Strains from the Ryukyu Islands (Japan) grouped into two clusters, and most Nepalese strains formed a single cluster, but Tibetan strains were heterogenous.  相似文献

Chloroplasts in heterokont algae probably originated from a red algal endosymbiont which was engulfed and retained by a eukaryotic host, and are surrounded by four envelope membranes. The outermost of these membranes is called chloroplast ER (CER) and usually connects with the nuclear envelope. This information, however, is based mainly on studies on single‐plastid heterokont algae. In multi‐plastid heterokont algae, it is still unclear whether CER is continuous with the nuclear envelope. Since nuclear‐encoded chloroplast proteins are synthesized by ribosomes on the ER membrane, clarifying the ER‐CER structure in the heterokont algae is important in order to know the targeting pathway of those proteins. We did a detailed ultrastructural observation of endomembrane systems in a multi‐plastid heterokont alga: Heterosigma akashiwo, and confirmed that the CER membrane was continuous with the ER membrane. However, unlike the CER membranes in other heterokont algae, it seemed to have very few ribosome attached. We also performed experiments for protein targeting into canine microsomes using a precursor for a nuclear‐encoded chloroplast protein, a fucoxanthin‐chlorophyll protein (FCP), of H. akashiwo, to see if the protein is targeted to the ER. It demonstrated that the precursor has a functional signal sequence for ER targeting, and is co‐translationally translocated into the microsomes. Based on these data, we propose a hypothesis that, in H. akashiwo, nuclear‐encoded chloroplast protein precursors that have been co‐translationally inserted into the ER lumen are sorted in the ER and transported to the chloroplasts through the ER.  相似文献

The phosphocreatine content of smooth muscle is of similar magnitude to ATP. Thus the function of the creatine kinase system in this tissue cannot simply be regarded as an energy buffer. Thus an understanding of its role in smooth muscle behavior can point to CK function in other systems. From our perspective CK function in smooth muscle is one example of a more general phenomenon, that of the co-localization of ATP synthesis and utilization. In an interesting and analogous fashion distinct glycolytic cascades are also localized in regions of the cell with specialized energy requirements. Similar to CK, glycolytic enzymes are known to be localized on thin filaments, sarcoplasmic reticulum and plasma membrane. In this chapter we will describe the relations between glycolysis and smooth muscle function and compare and contrast to that of the CK system. Our goal is to more fully understand the significance of the compartmentation of distinct pathways for ATP synthesis with specific functions in smooth muscle. This organization of metabolism and function seen most clearly in smooth muscle is likely representative of many other cell types.  相似文献

Field litterbag studies were conducted in the 2000 rainy season and the 2000/2001 dry season along the transect of West African major agroecological zones (agroeco-zones) to measure the decomposition of, and N and P release from 5 plant residues (leaves of woody species) with increasing quality: Dactyladenia barteri, Pterocarpus santalinoides, Alchornea cordifolia, Senna siamea and Gliricidia sepium. The decomposition rate constant (wk−1) ranged from 0.034 (Dactyladenia, subhumid zone) to 0.49 (Gliricidia, humid zone) in the rainy season, and from 0.01 (Dactyladenia, subhumid zone) to 0.235 (Pterocarpus, arid zone) in the dry season. The direct correlation between the decomposition rate of plant residues and their quality was only valid in agroeco-zones where there is not moisture stress. Similarly, the direct correlation between the decomposition rate of plant residues and moisture availability was only valid for plant residues with high quality. The decomposition rate of the low quality plant residue could increase from humid to arid zone in West Africa. In the arid zone, the low quality plant residue could also decompose faster than high quality plant residue. The climate-residue quality interactive effects on plant residue decomposition in West Africa were attributed to the feedback of low quality plant residue’s mulching effect, soil fauna and appreciable photodegradation in dry regions. A decomposition equation that could be used to predict the decomposition rate of plant residues with various qualities across agroeco-zones in West Africa was obtained from this study. The equation was expressed as follow: k = 0.122 − 0.000747*PRQI2− 0.0233*PRQI*CI + 0.00337*CI* PRQI2, in which k is the decomposition rate constant (wk−1), PRQI the plant residue quality index, and CI the climate index (ratio of rainfall to sunshine hours cumulative during the entire decomposition). The response of N and P release from plant residues to residue quality and climate was similar to that of residue decomposition. At the late stage of the dry season decomposition, the high C/N and C/P ratio plant residue (Dactyladenia leaves) that immobilized N and P in wet zones showed a release of N and P in the dry zone.
