Summary The in vitro production of the important regulatory of erythropoiesis, erythropoietin (Epo), is reviewed. It is concluded that it is possible to produce almost routinely small quantities of Epo in tissue culture. Although such procedures offer the potential to provide large quantities of the hormone for clinical use, the optimum culture conditions and mechanisms for triggering Epo production have yet to be resolved. These studies examine the trophic effects of progesterone (P) on the progesterone receptor (Rp) and growth of the decidua basalis (DB) and junctional zone (JZ) in the rat placenta. Pregnant rats were ovariectomized (Ovx) in mid-pregnancy and received steroid replacement therapy consisting of implantation of P pellets (25 mg) and injections of estradiol (E), 2 micrograms s.c., daily. Placental protein synthesis, measured by 3H-leucine incorporation in vitro, decreased more than 99% within 24 h of Ovx. However, treatment with P immediately after castration maintained control levels of synthesis. Delay of P treatment for 4 h caused a 60% decline in protein production measured 20 h later (p less than 0.01). Intraperitoneal implantation of a 50-mg pellet of the antiprogestin, RU-38486, in intact pregnant rats decreased protein synthesis by 50% within 6 h and by more than 90% 12 h and 24 h post-implantation (p less than 0.01). Growth of DB and JZ in Ovx rats treated for 48 h with P and/or E was studied both histologically and by changes in protein and DNA content. Rp binding activity was also measured by exchange assay under equilibrium conditions. Only P was able to reverse the effects of Ovx on growth of the DB and JZ. P also maintained Rp levels in the DB above those observed in Ovx and Ovx + E-treated groups (p less than 0.01). The Rp may be a constitutive product in the JZ since binding activity was not altered by Ovx or by steroid treatments. This study shows that P is clearly a trophic hormone of the maternal and chorioallantoic placenta and is essential for placental growth, cellular differentiation, and histological integrity.  相似文献

Roadways may pose barriers to long-distance migrators such as some ungulates. Highway underpasses mitigate wildlife-vehicle collisions and can be an important management tool for protecting migration corridors. In northern California, 3 underpasses were built on United States Route 395 (Route 395) in Hallelujah Junction Wildlife Area (HJWA) in the 1970s for a migratory mule deer (Odocoileus hemionus) herd that had been negatively affected by highway traffic. To determine whether these underpasses were still reducing mule deer mortalities >40 years after construction, we investigated deer use of the underpasses from 2006–2019 using cameras, global positioning system (GPS) collars, and roadkill records. We used occupancy models, approximations of GPS-collared mule deer movement paths, and roadkill locations to estimate the highway crossing patterns of deer. From camera data, there was higher use of the underpasses by deer during migration (spring [Mar–Jun], fall [Oct–Dec]) than in summer (Jul–Sep), when only resident deer were present. Higher underpass usage occurred in the spring compared to fall migrations. Eleven of 21 GPS-collared migrating mule deer crossed Route 395. We estimated 30% of the crossings (by 7 of the 11 deer) occurred south of the underpasses where deer could easily access the highway because of short (1-m high) and deteriorating highway fencing. Roadkill data confirmed that deer-vehicle collisions were occurring south of the underpasses and at the underpasses. This was likely due to deteriorating infrastructure at the underpasses that allows wildlife access to the highway. Overall, our study indicated that although underpasses can provide safe passage for migratory deer decades (>40 yr) after their construction, deteriorating infrastructure such as fencing and gates can lead to wildlife mortalities on highways near underpasses. © 2021 The Wildlife Society.  相似文献

Professional workers and consumers frequently use hard-surface cleaning products and these products may contain glycol ethers (GEs), such as 2-Butoxyethanol (2-BE). Governmental agencies have set exposure limits for some chemicals used in cleaning products and these exposure limits have been used as guides to protect human health. The study objectives were to determine realistic inhalation exposures for professional workers performing multiple, sequential cleaning tasks and compare the exposures to the acute reference exposure level (REL), which California established for 2-BE. The ConsExpo model was acceptable for evaluating exposure based on a comparison of its predictions to experimentally measured 2-BE vapor concentrations from hard-surface cleaning. The typical worker exposure was predicted for a cleaning scenario consisting of three bathrooms and three kitchens (or three bedrooms) in a 1-h period. This exposure scenario would not be expected to result in significant health consequences because the predicted exposure was much lower than the REL. The predicted chronic and aggregate exposures were also acceptable. This analysis identified two important variables that affect inhalation exposure: cleaners should be used with adequate ventilation and wet wiping towels should be properly disposed so that they are not a source of continuing exposure.  相似文献

An intensive regional research campaign was conducted by the North American Carbon Program (NACP) in 2007 to study the carbon cycle of the highly productive agricultural regions of the Midwestern United States. Forty‐five different associated projects were conducted across five US agencies over the course of nearly a decade involving hundreds of researchers. One of the primary objectives of the intensive campaign was to investigate the ability of atmospheric inversion techniques to use highly calibrated CO2 mixing ratio data to estimate CO2 flux over the major croplands of the United States by comparing the results to an inventory of CO2 fluxes. Statistics from densely monitored crop production, consisting primarily of corn and soybeans, provided the backbone of a well studied bottom‐up inventory flux estimate that was used to evaluate the atmospheric inversion results. Estimates were compared to the inventory from three different inversion systems, representing spatial scales varying from high resolution mesoscale (PSU), to continental (CSU) and global (CarbonTracker), coupled to different transport models and optimization techniques. The inversion‐based mean CO2‐C sink estimates were generally slightly larger, 8–20% for PSU, 10–20% for CSU, and 21% for CarbonTracker, but statistically indistinguishable, from the inventory estimate of 135 TgC. While the comparisons show that the MCI region‐wide C sink is robust across inversion system and spatial scale, only the continental and mesoscale inversions were able to reproduce the spatial patterns within the region. In general, the results demonstrate that inversions can recover CO2 fluxes at sub‐regional scales with a relatively high density of CO2 observations and adequate information on atmospheric transport in the region.  相似文献

Elevated atmospheric CO2 concentrations ([CO2]) generally increase primary production of terrestrial ecosystems. Production responses to elevated [CO2] may be particularly large in deserts, but information on their long‐term response is unknown. We evaluated the cumulative effects of elevated [CO2] on primary production at the Nevada Desert FACE (free‐air carbon dioxide enrichment) Facility. Aboveground and belowground perennial plant biomass was harvested in an intact Mojave Desert ecosystem at the end of a 10‐year elevated [CO2] experiment. We measured community standing biomass, biomass allocation, canopy cover, leaf area index (LAI), carbon and nitrogen content, and isotopic composition of plant tissues for five to eight dominant species. We provide the first long‐term results of elevated [CO2] on biomass components of a desert ecosystem and offer information on understudied Mojave Desert species. In contrast to initial expectations, 10 years of elevated [CO2] had no significant effect on standing biomass, biomass allocation, canopy cover, and C : N ratios of above‐ and belowground components. However, elevated [CO2] increased short‐term responses, including leaf water‐use efficiency (WUE) as measured by carbon isotope discrimination and increased plot‐level LAI. Standing biomass, biomass allocation, canopy cover, and C : N ratios of above‐ and belowground pools significantly differed among dominant species, but responses to elevated [CO2] did not vary among species, photosynthetic pathway (C3 vs. C4), or growth form (drought‐deciduous shrub vs. evergreen shrub vs. grass). Thus, even though previous and current results occasionally show increased leaf‐level photosynthetic rates, WUE, LAI, and plant growth under elevated [CO2] during the 10‐year experiment, most responses were in wet years and did not lead to sustained increases in community biomass. We presume that the lack of sustained biomass responses to elevated [CO2] is explained by inter‐annual differences in water availability. Therefore, the high frequency of low precipitation years may constrain cumulative biomass responses to elevated [CO2] in desert environments.  相似文献

The actin-associated protein Pdlim7 is essential for heart and fin development in zebrafish; however, the expression and function of this PDZ-LIM family member in the mammal has remained unclear. Here, we show that Pdlim7 predominantly localizes to actin-rich structures in mice including the heart, vascular smooth muscle, and platelets. To test the requirement for Pdlim7 in mammalian development and function, we analyzed a mouse strain with global genetic inactivation of Pdlim7. We demonstrate that Pdlim7 loss-of-function leads to significant postnatal mortality. Inactivation of Pdlim7 does not disrupt cardiac development, but causes mild cardiac dysfunction in adult mice. Adult Pdlim7 -/- mice displayed increased mitral and tricuspid valve annulus to body weight ratios. These structural aberrations in Pdlim7 -/- mice were supported by three-dimensional reconstructions of adult cardiac valves, which revealed increased surface area to volume ratios for the mitral and tricuspid valve leaflets. Unexpectedly, we found that loss of Pdlim7 triggers systemic venous and arterial thrombosis, leading to significant mortality shortly after birth in Pdlim7 +/- (11/60) and Pdlim7 -/- (19/35) mice. In line with a prothrombotic phenotype, adult Pdlim7 -/- mice exhibit dramatically decreased tail bleed times compared to controls. These findings reveal a novel and unexpected function for Pdlim7 in maintaining proper hemostasis in neonatal and adult mice.  相似文献

The catalyst layer of the cathode is arguably the most critical component of low‐temperature fuel cells and carbon dioxide (CO2) electrolysis cells because their performance is typically limited by slow oxygen (O2) and CO2 reduction kinetics. While significant efforts have focused on developing cathode catalysts with improved activity and stability, fewer efforts have focused on engineering the catalyst layer structure to maximize catalyst utilization and overall electrode and system performance. Here, we study the performance of cathodes for O2 reduction and CO2 reduction as a function of three common catalyst layer preparation methods: hand‐painting, air‐brushing, and screen‐printing. We employed ex‐situ X‐ray micro‐computed tomography (MicroCT) to visualize the catalyst layer structure and established data processing procedures to quantify catalyst uniformity. By coupling structural analysis with in‐situ electrochemical characterization, we directly correlate variation in catalyst layer morphology to electrode performance. MicroCT and SEM analyses indicate that, as expected, more uniform catalyst distribution and less particle agglomeration, lead to better performance. Most importantly, the analyses reported here allow for the observed differences over a large geometric volume as a function of preparation methods to be quantified and explained for the first time. Depositing catalyst layers via a fully‐automated air‐brushing method led to a 56% improvement in fuel cell performance and a significant reduction in electrode‐to‐electrode variability. Furthermore, air‐brushing catalyst layers for CO2 reduction led to a 3‐fold increase in partial CO current density and enhanced product selectivity (94% CO) at similar cathode potential but a 10‐fold decrease in catalyst loading as compared to previous reports.  相似文献
