Bacterial glucokinase (GK) binds to purified, human erythrocyte glucose transporter (GT) reconstituted in vesicles. The binding is largely abolished if GT is predigested with trypsin, indicating that GK binds to the cytoplasmic domain of GT. The binding is a saturable function of GK concentration showing two distinct affinities with apparent KD of 0.33 and 5.1 μM. The binding is stimulated by an increasing concentration of ADP with the 50% maximal effect at 5 mM. Glucose-6-phosphate (G6P) also stimulates the binding with a distinct optimum at 25 mM. The binding is stimulated only slightly by ATP. D-glucose has no affect on the binding. KCl enhances the binding with the maximal effect at physiological intracellular concentrations. The binding is sensitive to changes in pH with an optimum at pH 4. The binding causes no detectable functional change in GT. However, the enzymatic activity of GK measured at nanomolar concentrations of GK is significantly greater in the presence of GT vesicles than in its absence or in the presence of protein-free vesicles, indicating that GK interacts with GT at this low concentration range with an apparent KD of 10 mM. Although its physiological significance is not known, the GK-GT interaction in vitro described here suggests that these two proteins may also interact in the cell and regulate carbohydrate metabolism. © 1993 Wiley-Liss, Inc.  相似文献

Sickle cell anaemia (SCA) exhibits significant variations in clinical presentation in different populations for which several genetic factors including SCA-associated -and -thalassaemias, G-6-PD deficiency and elevated Hb F level have been implicated as possible ameliorating factors. Saudi Arabia is unique in that mild and severe forms of the disease occur at a high frequency. We investigated the G/A ratio and Hb F level and correlated these values with the severity of SCA. The results showed that Hb F level varies significantly in both groups of patients with no evident correlation with the mild clinical manifestations. However, G/A ratio correlated significantly with the disease severity where a high ratio was observed in patients with the mild and a low ratio in patients with the severe disease. The results are evaluated and discussed in the light of correlation studies and regression analysis.  相似文献

Primary structure of β-chain of pigeon is presented. It was determined by amino acid sequence analysis of intact β-chain and its peptides obtained by the enzymatic and chemical cleavage. Comparison of amino acid sequence of the chain with other available data shows β 14 Ile, β61 Lys, and β113 Ile as residues specific to pigeon. This report describes the cases of three mentally and physically well-developed siblings, 12, 10, and 8 years of age, with varying degrees of isolated valvular pulmonic stenosis not related to age. The severest lesion occurred in the middle patient and was associated with a right-to-left shunt through a patent foramen ovale. The three children had no other siblings, and there was no history of congenital heart disease among close relatives. These cases support the conclusion that genetic factors play a significant role in the development of certain congenital cardiac disorders.  相似文献

The population abundance, infestation, and harmful effects of the aphid Aphis craccivora Koch (Hemiptera: Aphididae) were studied on four bean plant species, namely the country bean (Lablab purpureus var. BARI Seem 1), the yard‐long bean (Vigna sesquipedalis var. BARI Borboti 1), the hyacinth bean (Dolichos lablab var. BARI Seem 6), and the bush bean (Phaseolus vulgaris var. BARI Jar Seem 3). Aphid abundance and infestation on the leaves, inflorescences, flowers, and pods differed significantly among the bean plant species, with P. vulgaris and V. sesquipedalis having the lowest and highest results, respectively. Aphid severity grade and the number of trichomes of the bean plant species were negatively correlated. The duration of the growth stages among the bean plant species were significantly different, with V. sesquipedalis having the shortest durations. Aphid abundance and infestation significantly affected the physical and phytochemical characteristics of the bean plant species. The highest reduction of number of leaves, flower inflorescences, and pod inflorescences per plant, and moisture and chlorophyll content in the leaves was found in L. purpureus. The results for V. sesquipedalis revealed the highest reduction in plant height, seed weight, and pH, while those of D. lablab showed the highest reduction in leaf area.  相似文献
