The aim of this investigation was to determine the persistence of biofilm-associated antibiotic resistance developed by methicillin-sensitive Staphylococcus aureus (MSSA), of different capsular types, during biofilm formation. Because of superiority of the tissue culture plate (TCP) over the Congo Red Agar (CRA) method for measuring biofilm formation, it was used to determine the persistence of the antibiotic resistance developed by the isolates in biofilms. The antibiotic resistance was found to persist for 3–4 wk post-propagation as planktonic subcultures. Interestingly, some strains even developed resistance to vancomycin and/or teicoplanin. However, no association of either biofilm formation or persistent antibiotic resistance with the major capsular phenotype was observed. These observations highlight the potential significance of (a) determining the antibiograms of S. aureus subcultured from biofilms developed in vitro using the TCP method as well as from planktonic cultures for formulation of an optimal therapeutic strategy, and (b) continuing to identify predominant non-capsular antigens contributing to biofilm formation, regardless of the capsular phenotype for the development of an effective potentially broad-spectrum vaccine for prevention of bovine mastitis caused by S. aureus.  相似文献

Summary Responses from four generations of index selection for egg production to 280 days of age in four White Leghorn populations have been presented. A pedigreed randombred population derived from one of the lines was reared with the selected lines to measure the environmental trend. The magnitude of total as well as average response although varying from population to population was positive in all the lines studied. Close correspondence between predicted and realized gains indicated that natural selection, genotype environmental interactions and environmental fluctuations were unimportant during the course of selection. Realized heritabilities agreed fairly well with the estimated heritabilities in at least three out of four populations studied. Probable reasons for variable and insufficient response were investigated.  相似文献

The photosynthetic and physiological performances of Oryza sativa L. (rice) were evaluated in organic and conventional rice–rice agroecosystems for 120 days after transplantation by measuring net photosynthesis (P N), transpiration (E), stomatal conductance (g s), intercellular CO2 concentration (C i), chlorophyll content (SPAD) and JIP fluorescence rise. The soil health was measured as soil bacterial and fungi density and activities of soil microbial enzymes (amylase, invertase, cellulase, protease, alkaline phosphatase and dehydrogenase). The conventionally managed fields showed lower microbial density and activity than of organic fields especially after 60 days of transplantation. The crop grown in the conventional fields has significantly low level of P N and chlorophyll, but E, g s and C i did not differ significantly till 105 days after transplantation. The JIP rise was low in conventional fields than in organic fields during 90–120 days. The efficiency and plant performance parameters (φP 0, Ψ 0, φE 0, PIφ, PI Ψ , PIABS, and PItotal) showed a rapid rate of decrease in the conventional than of organic fields. Significant positive correlation could be established between the performance and soil microbial activities, whereas the stress indicating fluorescence parameters (V J, M 0, φD 0, DI0/RC) showed significant negative correlation with the soil parameters in both the farming systems. The result showed that JIP analysis can be used as an early indicator of soil fertility and plant performance.  相似文献

Transient receptor potential, melastatin-like 7 (Trpm7) is a combined ion channel and kinase implicated in the differentiation or function of many cell types. Early lethality in mice and frogs depleted of the corresponding gene impedes investigation of the functions of this protein particularly during later stages of development. By contrast, zebrafish trpm7 mutant larvae undergo early morphogenesis normally and thus do not have this limitation. The mutant larvae are characterized by multiple defects including melanocyte cell death, transient paralysis, and an ion imbalance that leads to the development of kidney stones. Here we report a requirement for Trpm7 in differentiation or function of dopaminergic neurons in vivo. First, trpm7 mutant larvae are hypomotile and fail to make a dopamine-dependent developmental transition in swim-bout length. Both of these deficits are partially rescued by the application of levodopa or dopamine. Second, histological analysis reveals that in trpm7 mutants a significant fraction of dopaminergic neurons lack expression of tyrosine hydroxylase, the rate-limiting enzyme in dopamine synthesis. Third, trpm7 mutants are unusually sensitive to the neurotoxin 1-methyl-4-phenylpyridinium, an oxidative stressor, and their motility is partially rescued by application of the iron chelator deferoxamine, an anti-oxidant. Finally, in SH-SY5Y cells, which model aspects of human dopaminergic neurons, forced expression of a channel-dead variant of TRPM7 causes cell death. In summary, a forward genetic screen in zebrafish has revealed that both melanocytes and dopaminergic neurons depend on the ion channel Trpm7. The mechanistic underpinning of this dependence requires further investigation.  相似文献

The improved accessibility to data that can be used in human health risk assessment (HHRA) necessitates advanced methods to optimally incorporate them in HHRA analyses. This article investigates the application of data fusion methods to handling multiple sources of data in HHRA and its components. This application can be performed at two levels, first, as an integrative framework that incorporates various pieces of information with knowledge bases to build an improved knowledge about an entity and its behavior, and second, in a more specific manner, to combine multiple values for a state of a certain feature or variable (e.g., toxicity) into a single estimation. This work first reviews data fusion formalisms in terms of architectures and techniques that correspond to each of the two mentioned levels. Then, by handling several data fusion problems related to HHRA components, it illustrates the benefits and challenges in their application.  相似文献

Simple sequence repeats (SSR) are the DNA markers of choice for genetic analysis in rice (Oryza sativa L.) due to their abundance, high polymorphism and simple assays using agarose gel electrophoresis. In an attempt to find most variable SSR loci for the agarose gel system, the relationship between SSR length and level of polymorphism was evaluated in a set of eight diverse rice genotypes using 201 random SSR loci of different repeat motifs and lengths, representing both genic and intergenic sequences from the 12 rice chromosomes. There was a positive correlation between SSR length and average number of alleles per locus but linearity of this relationship was limited to the SSR length range of 10–70 bp. The highest level of polymorphism was in the SSR length range of 51–70 bp, beyond which there was stabilization and then decline of polymorphism in SSRs longer than 70 bp. Proportion of polymorphic loci in the different SSR length groups also followed similar pattern with even sharper decline of polymorphism in the highest size range. Here we describe a genome wide set of 436 validated highly variable SSR (HvSSR) markers with repeat lengths of 51–70 bp for their consistent amplification and high polymorphism. In the parental lines of three different mapping populations, the HvSSR loci showed more than twice the level of polymorphism than random SSR markers with average repeat length of 34 bp, and therefore are suitable for QTL mapping and fingerprinting studies in rice employing agarose gels.  相似文献
