Insect-derived cell lines are used extensively to produce recombinant proteins because they are capable of performing a range of post-translational modifications. Due to their significance in biotechnological applications, various methods have been developed to transfect them. In this study, we introduce a virosome constructed from vesicular stomatitis virus (VSV) as a new delivery system for sf9 cells. We labeled these VSV virosomes by fluorescent probe Rhodamine B chloride (R18). By fluorescence microscope observation and conducting a fusion assay, we confirmed the uptake of VSV virosomes via endocytosis by sf9 cells and their fusion with the endosomal membrane. Moreover, we incubated cationic VSV virosomes with a GFP-expressing bacmid and transfected sf9 cells, after 24?h some cells expressed GFP indicating the ability of VSV virosomes to deliver heterologous DNA to these cells. This is the first report of a virosome-based delivery system introduced for an insect cell line.  相似文献

The sawfly, Arge rosae Linnaeus (Hymenoptera: Argidae) is a serious pest of rose plants in a vast area of the world including Iran. Pesticides are applied in order to control in areas where infestation is high. So, there is a need for more environmentally benign alternatives to control this insect pest in urban areas. Therefore, the aim of the current study was to study the effect of cereal seed proteinaceous extracts including wheat and triticale on the insect α-amylase. Wheat and triticale seed proteins as well as fourth instars larvae α-amylase were extracted. The results showed that there are two different α-amylase isoenzymes in the insect gut, one major band and the other minor band, which had optimal activity at alkaline pH (pH 8.0). The effect of five concentrations including 0.42, 0.21, 0.105, 0.05 and 0.025?mg?ml?1 of each seed extract was tested on α-amylase activity of the larvae gut. At the highest concentration (0.42?mg?ml?1), wheat seed extract caused 74% inhibition of the enzyme activity while triticale seed extract inhibited 62% enzyme activity. Experiments proved that seed proteinaceous extract affected the enzyme activity in a pH-dependant manner.  相似文献

It is hypothesized that Capoeta fusca might display specific life‐history traits that differ from other species of this genus. To test this hypothesis a total of 354 specimens of C. fusca (listed by ICUN in the category DD = data deficient) were caught in the Qanat of Shadmehr (a well‐known active man‐made water well system in eastern Iran) on a monthly basis to cover fully the reproductive season from April to October 2007. Based on opercula readings the maximum ages of the population were 5+ years for both sexes. Sizes ranged from 57 to 190 mm total length (weight 2.14–84.76 g). Length‐weight relationship implied that the growth was negatively allometric for males and isometric for females. The von Bertalanffy growth model was estimated as Lt=18.74(1‐e?0.33(t+0.473)) and Lt=22.35(1‐e?0.32(t+0.333)) for males and females, respectively. Sex ratio was 1 : 2.42 in favour of females. The GSI indicated that reproduction of the fish in the qanat system occurred between May and August, with the highest average value of 6.12 for males in June and 9.55 for females in July. Oocyte diameters ranged from 0.30 to 2.05 mm, with a mean value of 0.92 mm. Absolute fecundity ranged between 506 and 22 800 eggs, with a mean of 4961 eggs. Fecundity relative to total weight fluctuated from 34 to 583, with a mean value of 133 eggs per g. Absolute fecundity and oocyte diameter to fish size (length and weight) were significantly correlated.  相似文献

The spider mite, Tetranychus urticae Koch is a serious pest of economically important plants in closed and open area worldwide. The spider mite resistance to acaricide plays a major role in the failure of the chemical control method. Thus, the aim of the current study was to evaluate the efficacy of two acaricides, abamectin and propargite, against two populations (strains) of the spider mite. Results showed that LC50s of the abamectin against susceptible and resistant strains of the spider mite were 0.1 and 2730?ppm, respectively. Whilst LC50s of the propargite against susceptible and resistant strains of the spider mite were 55 and 7199?ppm, respectively. Resistance ratio (RR) calculated as the ratio of resistance LC50/susceptible LC50 showed that RR for abamectin and propargite was 20285 and 130, respectively. The enzyme assay results showed that three mechanisms of MFO, GST and EST are involved in the abamectin resistance of the spider mite. In gel assays, when α-naphthyl acetate was used as substrate, three bands appeared in the gel in which bands E2 and E3 were major bands and E1 was a minor band confirming that α-naphthyl acetate was a better substrate for general esterase activity in the spider mite whereas β-acetate when used for esterase activity, only two faint bands (E1 and E2) were observed. The order of their involvement in the abamectin resistance is EST?>?MFO?>?GST.  相似文献
