A series of aqueous solutions of poly(vinyl alcohol) of various commercial products were prepared and applied onto the surfaces of cotton and blends of cotton/polyester fabrics. Fourier transform infrared spectrophotometer was used to confirm the molecular structure of the polyvinyl alcohol used. Performance tests such as colorfastness to rubbing (dry and wet) and colorfastness to washing were determined. The controlling variables affecting the performance properties of the finished substrate such as post-treatment with poly(vinyl alcohol) of various commercial trades, concentration and dilutions were studied. Crocking, washing and hue change of the treated dyed and printed fabrics is accompanied by the formation of semi-inter-penetrated network structure due to the presence of the hydroxyl (-OH) groups which make feasible to a number of grafting and physical cross linking reactions of polymer backbone. 相似文献
Arsenical compounds exhibit a differential toxicity to cancer cells. Microtubules are a primary target of a number of anticancer drugs, such as arsenical compounds. The interaction of 1-NAA (1-naphthylarsonic acid) has been investigated on microtubule polymerization under in vitro and cellular conditions. Microtubules were extracted from sheep brain. Transmission electron microscopy was used to show microtubule structure in the presence of 1-NAA. Computational docking method was applied for the discovery of ligand-binding sites on the microtubular proteins. Proliferation of HeLa cells and HF2 (human foreskin fibroblasts) was measured by the MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide] assay method following their incubation with 1-NAA. Fluorescence microscopic labelling was done with the help of α-tubulin monoclonal antibody and Tunel kit was used to investigate the apoptotic effects of 1-NAA on the HeLa cells. 1-NAA inhibits the tubulin polymerization by the formation of abnormal polymers having high affinity to the inner cell wall. 相似文献
Calcitonin (CT) is one of the factors affecting the embryo implantation, but its effects on the implantation window have not been fully investigated. The current study investigated the effects of CT on the endometrium receptivity by morphological study and evaluation of leukemia inhibitory factor (LIF), mucin 1 (Muc-1), and microRNA (miRNA) Let-7a in the ovarian stimulation and the normal ovarian cycle. Then the mechanism of the CT effects through the mammalian target of rapamycin (mTOR) signaling pathway was studied by using PP242. A total of 64 BALB/c mice were divided into the normal ovarian cycle and ovarian stimulation groups. Each group consisted of four subgroups: control, calcitonin, PP242, and calcitonin+PP242. CT and PP242 were injected on the fourth of pregnancy into the mice and 24 hr later all the mice were killed. The uterine tissue samples were used for morphological analysis, and endometrial cells were mechanically isolated for evaluation of gene and protein expression. The results showed that ovarian stimulation induced mTOR phosphorylation as well as increased expression of the Let-7a miRNA. In addition, CT injection increased the expression of LIF and miRNA Let-7a in ovarian stimulation similar to that in normal ovarian cycles. However, injection of PP242 reduced expression of miRNA Let-7a and increased Muc-1 expression in ovarian stimulation group. In conclusion, the administration of CT improved endometrial receptivity in mice. This phenomenon occurred by upregulation of LIF, miRNA Let-7a and downregulation of Muc-1 via mTOR signaling pathway. 相似文献
The human epidermal growth factor 2 (HER2) gene undergoes various mutations that could alter its activity or respond to the antibody therapies. Cetuximab, a known anti-EGFR monoclonal antibody (mAB), is widely administered in metastatic colorectal cancer (mCRC) cases. Here we identified mCRC patients who did not respond to cetuximab (500 mg/m2, q2w) after fluoropyrimidine/oxaliplatin regimen failure. Tumor samples were examined with immunohistochemistry for protein distribution, polymerase chain reaction (PCR) sequencing for mutation detection and real-time PCR for mRNA expression pattern analysis between cetuximab sensitive and resistance patients. The conformational differences of normal and mutated protein structures were predicted by bioinformatics analysis. The 5-year survival rates of target groups were estimated using the Kaplan–Meier method. Immunohistochemistry showed that all cases had high level of HER2 protein. No K-Ras or B-Raf mutation was observed among the study population; however, cetuximab resistance patients harbored a somatic mutation R784G at the exon 20 region of HER2 coding sequence. According to bioinformatics analysis, this mutation caused a notable misfold in protein conformation. Meanwhile, survival analysis showed R784G mutated mCRC patients had shortened survival rate compared with the mCRC cases with wild-type HER2. Collectively, these data report a new mechanism of resistance to cetuximab and might be applicable in modifying new therapeutic strategies for HER2 involved cancers. 相似文献
Cylindrospermopsis (Raphidiopsis) raciborskii is an invasive, filamentous, nitrogen-fixing cyanobacterium that forms frequent blooms in freshwater habitats. While viruses play key roles in regulating the abundance, production and diversity of their hosts in aquatic ecosystems, the role(s) of viruses in the ecology of C. raciborskii is almost unexplored. Progress in this field has been hindered by the absence of a characterized virus–host system in C. raciborskii. To bridge this gap, we sequenced the genome of CrV-01T, a previously isolated cyanosiphovirus, and its host, C. raciborskii strain Cr2010. Analyses suggest that CrV-01T represents a distinct clade of siphoviruses infecting, and perhaps lysogenizing, filamentous cyanobacteria. Its genome contains unique features that include an intact CRISPR array and a 12 kb inverted duplication. Evidence suggests CrV-01T recently gained the ability to infect Cr2010 and recently lost the ability to form lysogens. The cyanobacterial host contains a CRISPR-Cas system with CRISPR spacers matching protospacers within the inverted duplication of the CrV-01T genome. Examination of metagenomes demonstrates that viruses with high genetic identity to CrV-01T, but lacking the inverted duplication, are present in C. raciborskii blooms in Australia. The unique genomic features of the CrV/Cr2010 system offers opportunities to investigate in more detail virus–host interactions in an ecologically important bloom-forming cyanobacterium. 相似文献
The acyl carrier proteins (ACPs) of fatty acid synthase and polyketide synthase as well as peptidyl carrier proteins (PCPs) of nonribosomal peptide synthetases are modified by 4'-phosphopantetheinyl transferases from inactive apo-enzymes to their active holo forms by transferring the 4'-phosphopantetheinyl moiety of coenzyme A to a conserved serine residue of the carrier protein. 4'-Phosphopantetheinyl transferases have been classified into two types; the AcpS type accepts ACPs of fatty acid synthase and some ACPs of type II polyketide synthase as substrates, whereas the Sfp type exhibits an extraordinarily broad substrate specificity. Based on the previously published co-crystal structure of Bacillus subtilis AcpS and ACP that provided detailed information about the interacting residues of the two proteins, we designed a novel hybrid PCP by replacing the Bacillus brevis TycC3-PCP helix 2 with the corresponding helix of B. subtilis ACP that contains the interacting residues. This was performed for the PCP domain as a single protein as well as for the TycA-PCP domain within the nonribosomal peptide synthetase module TycA from B. brevis. Both resulting proteins, designated hybrid PCP (hPCP) and hybrid TycA (hTycA), were modified in vivo during heterologous expression in Escherichia coli (hPCP, 51%; hTycA, 75%) and in vitro with AcpS as well as Sfp to 100%. The designated hTycA module contains two other domains: an adenylation domain (activating phenylalanine to Phe-AMP and afterward transferring the Phe to the PCP domain) and an epimerization domain (converting the PCP-bound l-Phe to d-Phe). We show here that the modified PCP domain of hTycA communicates with the adenylation domain and that the co-factor of holo-hPCP is loaded with Phe. However, communication between the hybrid PCP and the epimerization domain seems to be disabled. Nevertheless, hTycA is recognized by the next proline-activating elongation module TycB1 in vitro, and the dipeptide is formed and released as diketopiperazine. 相似文献
We are proposing graphene (G)-based multilayered plasmonic spatial switch, operating at 10 THz. It is composed of hBN/Ag/hBN/G/hBN/G/hBN/SiO2/p+-Si multilayers. When a 10-THz transverse magnetic (TM)-polarized signal is normally incident upon the structure top surface, the nanoaperture devised in the Ag nanolayer, acting as a grating, excites surface plasmons at the top graphene micro-ribbons/hBN interface. These surface plasmons depending on the graphenes chemical potentials can be coupled to the lower-right or left graphene micro-ribbons and continue to propagate laterally towards the corresponding output port. Numerical simulations show that a change of ∆VG ≈ ± 2.7 V in the voltage, applied to the gated micro-ribbons, can modulate their chemical potentials sufficiently to switch the right (left) output port from ON (OFF) to OFF(ON) and vice versa. Besides its low power consumption, the switch ultra-small dimensions make it a potential spatial router suitable for THz-integrated circuit applications.
The popularity of lemon balm in conventional medicine is suggested by the existence of two groups of compounds, namely essential oil and phenylpropanoids pathway derivatives. One of the promising approaches to improve tolerance to drought stress induced oxidative damage in seedlings grown in greenhouses and plant growth chambers is replacing the traditional and high-cost light technologies by recently developed light emitting diodes (LED). In this experiment, we analyzed the role of various LED lights including red (R), blue (B), red (70%) + blue (30%) (RB), and white (W) as well as normal greenhouse light (as control) to stimulate defense mechanisms against drought stress in two genotypes of Melissa officinalis L. The present study demonstrates that pre-treatment with LEDs with high-intensity output for 4 weeks alleviated harmful effects of drought stress in the two genotypes. Under drought stress, RB LED pre-treated plantlets of the two genotypes exhibited the highest relative growth index of shoot and root and total phenolic and anthocyanin content compared to those irradiated with other LEDs and greenhouse lights. The highest amount of malondialdehyde level was detected in R LED exposed plants. In response to drought, LED-exposed plants especially RB light-irradiated plants of the two genotypes maintained significantly higher antioxidant and phenylalanine ammonia-lyase (PAL) enzyme activities and higher expression level of the PAL1 and 4CL-1 genes compared to those irradiated with greenhouse light. We concluded that RB LED light provides a better growth condition and resistance to drought stress for the two genotypes of lemon balm by the highest antioxidant activity and the least amount of damage to the cell membranes. Our data suggest that LED light pre-treatments as moderate stress activate antioxidant systems, enhance the scavenging of ROS and induce drought stress tolerance in the two genotypes of lemon balm plants.
