We present a model and semiparametric estimation procedures for analysis of survival data with cross-effects (CE) of survival functions. Finite sample properties of the estimators are analyzed by simulation. A goodness-of-fit test for the proportional hazards model against the CE model is proposed. The well known data concerning effects of chemotherapy and radiotherapy on the survival times of gastric cancer patients is analyzed as an example.  相似文献

Genotoxicity of goniothalamin in CHO cell line   总被引:2,自引:0,他引:2  
Goniothalamin (GTN) is a styrylpyrrone derivative from Goniothalamus umbrosus and other Annonaceae species. It has been shown to have anti-cancer and apoptosis-inducing properties against various human tumour and animal cell lines. The compound has also been shown to be active in vivo against DMBA-induced rat mammary tumours and was reported as an anti-fertility agent in rats. The aim of our study was to assess the genotoxicity of GTN in CHO cells using the UKEMS guidelines. A metabolic activation fraction (S9) was prepared according to standard methods. The methylthiazoletetrazolium (MTT) screening assay was then carried out to determine the cytotoxicity index (IC50) of GTN. The average IC50 value was 12.45 (+/- 3.63)microM. The mitotic index (MI) assay was then performed to determine the clastogenicity indices (MI(C25), MI(C50) and MI(C100)) of GTN. The chromosome aberration (CA) induction assay using air-dried metaphase spread was then performed to investigate the clastogenic effects of goniothalamin. Benzo[a]pyrene (BaP) and ethylmethanesulphonate (EMS) were used as positive controls in the presence and absence of S9 metabolic activation, respectively. The anti-genotoxicity effect of GTN was also assessed using a combination of GTN and EMS, and GTN and BaP. Dose-responses of CA frequencies were determined for both, the genotoxicity and anti-genotoxicity effects. GTN on its own and when combined with positive controls, was found to induce and enhance CA, respectively. Chromatid and whole chromosome breaks/gaps, as well as interchanges, endoreduplications and ring chromosomes were the main types of aberration induced by GTN. The overall clastogenic effect of GTN was statistically significant. In conclusion, GTN is potentially a genotoxic or clastogenic substance without any anti-genotoxic properties.  相似文献

Homo- and heterodimers of AZT and d4T, possessing carbonate and carbamate linkers, have been synthesized with the aim to enhance the antiviral activity of their components. Homo- and heterodimer carbamates showed weak anti-HIV activity. On the other hand, dinucleoside carbonates showed marked antiviral activity.  相似文献

Summary This paper elaborates on the notion of homogeneity in plant populations and its quantification. It describes a method for testing homogeneity without the need to make assumptions about generalized distributions. Inoculation with the rhizosphere bacterium Azospirillum brasilense NH, originally isolated from salt-affected soil in northern Algeria, greatly enhanced growth of durum wheat (Triticum durum var. waha) under saline soil conditions. Important plant parameters like the rate of germination, stem height, spike length, dry weight of roots and shoots, chlorophyll a and b content, proline and total sugar contents, 1000-seed weight, seed number per spike, and weight of seeds per spike were measured. At salt stress conditions (160 and 200 mM NaCl) A. brasilense NH restored almost completely vegetative growth and seed production. The combination with extracts of the marine alga Ulva lactuca resulted in even more improved salt tolerance of durum wheat. Proline and total sugar accumulation, a sign of physiological plant stress under inhibitory salt conditions, was reduced in plants inoculated with A. brasilense NH with and without addition of algal extracts. Inoculation with the salt-sensitive A. brasilense strain Sp7 could not restore salt-affected plant growth at 200 mM NaCl. Furthermore, it could be demonstrated by fluorescence in situ hybridization and confocal laser scanning microscopy that A. brasilense NH is able to colonize roots of durum wheat endophytically under salt-stressed conditions. Thus, the salt-tolerant rhizobacterium A. brasilense NH could effectively provide alone or in combination with extracts of U. lactuca a promising solution to overcome salt inhibition which is a major threat hindering productive wheat cultivation in arid saline soils.  相似文献

Kernel shattering (KS) can cause severe grain yield loss in wheat (Triticum aestivum L.). The introduction of genotypes with Fusarium head blight (FHB) resistance has elevated the KS importance. ‘Sumai3,’ the most commonly used FHB-resistant germplasm worldwide, is reported to be KS susceptible. The objectives of this study were to detect quantitative trait loci (QTLs) for KS and to determine the relationship between KS and FHB. A recombinant inbred line population derived from a cross between Sumai3 and ‘Stoa’ was evaluated for KS in five environments and FHB in two field trials, separately. Four genomic regions on chromosomes 2B, 3B, and 7A were associated with KS. Of them, two major KS QTLs were detected consistently over three environments and each located proximal to the centromere on chromosomes 3B and 7A. The resistant alleles at these two QTLs together can reduce KS by 66.1% relative to the reciprocal alleles and by 41.1% compared to the population mean. The field FHB data revealed four QTLs on chromosomes 2B, 3B, and 7A. Three of these FHB QTLs coincided with and/or linked to the KS QTLs with opposite allele effects in the corresponding genomic regions, which may explain the negative correlation (r = −0.29 and P < 0.01) between the KS and FHB infection found in this study. The results in this study indicate that KS and FHB in Sumai3 are, in part, inherited dependently. However, the correlation between KS and FHB is not strong, and the major FHB resistance QTL on chromosome arm 3BS was not linked to any KS QTL. Our results showed that pyramiding of the two major KS-resistant alleles and the unlinked major FHB-resistant allele could produce lines with both low values of KS and FHB infection.  相似文献

Mauritanian date palm cultivars and progenies of two controlled crosses were analyzed according to the identity of mitochondrial plasmid-like DNAs. Starting from total genomic DNA and appropriate primers, polymerase chain reaction was designed to amplify either a 373-bp or a 265-bp fragments corresponding to the S and the R-plasmid respectively. Data proved that 5 cultivars out of 10 studied have exhibited the R-plasmid suggesting their resistance to the fusariosis. The existence of intra-cultivar variability has also been revealed in the cv. Ahmar. In addition, analysis throughout progenies of two controlled crosses suggested the strict maternal transmission of the date palms’ mitochondrial genome.  相似文献

Disaccharidases (maltase, cellobiase, lactase, and sucrase), alpha-amylase, and glucoamylase in the camel small intestine were investigated to integrate the enzymatic digestion profile in camel. High activities were detected for maltase and glucoamylase, followed by moderate levels of sucrase and alpha-amylase. Very low activity levels were detected for lactase and cellobiase. Camel intestinal maltase-glucoamylase (MG) was purified by DEAE-Sepharose and Sephacryl S-200 columns. The molecular weight of camel small intestinal MG4 and MG6 were estimated to be 140,000 and 180,000 using Sephacryl S-200. These values were confirmed by SDS-PAGE, where the two enzymes migrated as single subunits. This study encompassed characterization of MGs from camel intestine. The Km values of MG4 and MG6 were estimated to be 13.3 mM and 20 mM maltose, respectively. Substrate specificity for MG4 and MG6 indicated that the two enzymes are maltase-glucoamylases because they catalysed the hydrolysis of maltose and starch with alpha-1,4 and alpha-1,6 glycosidic bonds, but not sucrose with alpha-1,2 glycosidic bond which was hydrolyzed by sucrase-isomaltase. Camel intestinal MG4 and MG6 had the same optimum pH at 7.0 and temperature optimum at 50 degrees C and 40 degrees C, respectively. The two enzymes were stable up to 50 degrees C and 40 degrees C, followed by strong decrease in activity at 60 degrees C and 50 degrees C, respectively. The effect of divalent cations on the activity of camel intestinal MG4 and MG6 was studied. All the examined divalent cations Ca(2+), Mn(2+), Mg(2+), Co(2+) and Fe(3+) had slight effects on the two enzymes except Hg(2+) which had a strong inhibitory effect. The effect of different inhibitors on MG4 and MG6 indicated that the two enzymes had a cysteine residue.  相似文献

Germination studies of Cymbopogon schoenanthus (Poaceae) distributed along southern Tunisia were carried out to assess the effects of salinity. A preliminary experiment showed 30°C as the optimum germination temperature for seeds of this species. After that, seed germination was studied at different salinity levels. Our results revealed a decrease in germination percentage with increasing salinity. Germination rate, however, was maintained up to 200 mM NaCl and drastically declined at 300 mM NaCl.  相似文献
