BACKGROUND: Recent studies indicate marginal biotin deficiency is more common than previously thought. That conclusion's validity rests on two indicators of biotin status that depend on renal function.OBJECTIVE: Assessing the validity of two indicators of biotin status that do not depend upon renal function: 1) activity of the biotin-dependent enzyme propionyl-CoA carboxylase (PCC) in lymphocytes and 2) accumulation of odd-chain fatty acids in the lipids of plasma and erythrocytes.DESIGN: Marginal biotin deficiency was induced in 11 healthy adults by egg-white feeding for 28 days. Blood and 24-h urine samples were collected before commencing the diet and twice weekly thereafter. After depletion, biotin status was restored with a general diet with or without 80 &mgr;g/day or 328 nmol/day biotin supplement. Activity of PCC was determined by an optimized NaH 14CO(3) incorporation assay. Fatty acid composition was determined by gas chromatography.RESULTS: With time on the egg-white diet, lymphocyte PCC activity decreased significantly (P <0.0001); C15:0 and C17:0 content increased significantly in the lipids of plasma and erythrocytes (P <0.015). In eight of 11 subjects, lymphocyte PCC activity returned to normal within three weeks of resuming general diets with or without biotin supplement. With repletion, C15:0 and C17:0 in plasma lipids decreased (P <0.02), but odd-chain content of erythrocytes did not decrease significantly.CONCLUSIONS: Lymphocyte PCC activity is an early and sensitive indicator of marginal biotin deficiency. Odd-chain fatty acids accumulate in blood lipids more gradually during marginal deficiency and return to normal more gradually after biotin repletion.  相似文献

Tobacco transformants that express an antisense RBCS construct were used to investigate the consequences of a lesion in photosynthetic carbon metabolism for nitrogen metabolism and secondary metabolism. The results show that an inhibition of photosynthesis and decrease in sugar levels leads to a general inhibition of nitrogen metabolism, and dramatic changes in the levels of secondary metabolites. The response was particularly clear in plants that received excess nitrogen. In these conditions, a decrease of Rubisco activity led to an inhibition of nitrate reductase activity, accumulation of nitrate, a decrease of amino acid levels that was larger than the decrease of sugars, and a large decrease of chlorogenic acid and of nicotine, which are the major carbon- and nitrogen-rich secondary metabolites in tobacco leaves, respectively. Similar changes were seen when nitrogen-replete wild-type tobacco was grown in low light. The inhibition of nitrogen metabolism was partly masked when wild-type plants and antisense RBCS transformants were compared in marginal or in limiting nitrogen, because the lower growth rate of the transformants alleviated the nitrogen deficiency, leading to an increase of amino acids. In these conditions, chlorogenic acid always decreased but the decrease of nicotine was ameliorated or reversed. When the changes in internal pools are compared across all the genotypes and growth conditions, two conclusions emerge. First, decreased levels of primary metabolites lead to a dramatic decrease in the levels of secondary metabolites. Second, changes of the amino acid : sugar ratio are accompanied by changes of the nicotine:chlorogenic acid ratio.  相似文献

This article examines the effect of access to health infrastructure, personnel and services on children's nutritional status in rural Nepal. Data for the study come from the 1996 Nepal Living Standards Survey, which includes individual- and household-level information on children's nutritional status and its environmental and socioeconomic determinants, and community-level information on the availability of health care infrastructure, personnel and services. The study uses a structural modelling approach to assess the relative contributions of the health care supply environment on children's anthropometric status via the pathway of maternal and child health (MCH) service use. The findings suggest that improvements in the availability of outreach clinics and the structural quality of the closest public facility would be expected to have statistically significant and large effects on the use of MCH services, and that increases in MCH service use would have a statistically significant impact on weight-for-age, but not weight-for-height or height-for-age. The overall impact of the heath care supply environment on nutritional status is assessed through a series of policy simulations.  相似文献

The availability of dissolved nutrients such as nitrate under extreme low temperatures is a strong determinant in the development and growth of ice diatoms. Consequently we investigated regulation of photosynthesis in a mixed culture of three diatom species, which grew in chemostats at -1 degrees C, 15 micromol photons m(-2) s(-1) under N-limitation. When nitrogen is limiting, pigment-protein complexes are one of the most affected structures under low-light conditions. The loss of integral polar thylakoid components destabilized the bilayer structure of the membrane with consequences for lipid composition and the degree of fatty acid desaturation. N-Limitation caused a decrease in monogalactosydiacylglycerol (MGDG) and a simultaneous increase in bilayer forming digalactosyldiacylglycerol (DGDG). Their ratio MGDG:DGDG decreased from 3.4 +/- 0.1 to 1.1 +/- 0.4, while 20:5 n-3 fatty acids of chloroplast related phospholipid classes such as phosphatidylglycerol (PG) increased under N-limitation. These data reveal that lipids are important components, required to sustain membrane structure under a deficiency of integral membrane bound proteins and pigments. Nonetheless, energy conversion at photosystem II is still affected by N-limitation despite this structural regulation. Photosynthetic quantum yield (F(v)/F(m)) and electron transport rates decreased under N-limitation caused by an increasing amount of electron acceptors (second stable electron acceptor = Q(B)) which had slower reoxidation kinetics. The energy surplus under these conditions is stored in triacylglycerols, the main energy sink in Antarctic sea ice diatoms under N-limitation.  相似文献

A simple and rapid method is presented for the preparation of RNA from various cyanobacteria. Unlike other methods that require a lysis solution, lysozymes, or proteinase K, the proposed method, called the bead–phenol–chloroform (BPC) method, uses silica/zirconia beads, phenol, and chloroform to break the cells and extract RNA more efficiently. Experiments confirm that the BPC method can successfully isolate total RNA from various cyanobacterial strains without DNA contamination, and the extracted RNA samples have a relatively high purity, concentration, and yield. Furthermore, the BPC method is more rapid, simple, and economical when compared with previously reported methods.  相似文献

Active oxygen (AO) production and a K+/H+ exchange response (XR) are two concurrent early events associated with incompatible plant-bacteria interactions that result in a hypersensitive response (HR). Recently, a protein, termed harpin, produced by Erwinia amylovora has been reported to be the elicitor responsible for the HR caused by this pathogen. Although both the bacterium and harpin are reported to induce XR in tobacco (Nicotiana tabacum) cell suspensions, there have been no reports regarding the concurrent production of AO in this system. Here we report that E. amylovora stimulates the AO response, whereas an E. amylovora mutant that does not produce harpin does not elicit the AO response. In addition, a cell-free preparation of harpin induces AO production. This study indicates that harpin may be the bacterial elicitor of the XR and AO responses during the development of E. amylovora-induced HR.  相似文献

Cytochrome P-450cam, the bacterial hemeprotein which catalyzes the 5-exo-hydroxylation of d-camphor, requires two electrons to activate molecular oxygen for this monooxygenase reaction. These two electrons are transferred to cytochrome P-450cam in two one-electron steps by the physiological reductant, putidaredoxin. The present study of the kinetics of reduction of cytochrome P-450cam by reduced putidaredoxin has shown that the reaction obeys first order kinetics with a rate constant of 33 s-1 at 25 degrees C with respect to: 1) the appearance of the carbon monoxide complex of Fe(II) cytochrome P-450cam; 2) the disappearance of the 645 nm absorbance band of high-spin Fe(III) cytochrome P-450cam; and 3) the disappearance of the g = 1.94 EPR signal of reduced putidaredoxin. This data was interpreted as indicative of the rapid formation of a bimolecular complex between reduced putidaredoxin Fe(III) cytochrome P-450cam. The existence of the complex was first shown indirectly by kinetic analysis and secondly directly by electron paramagnetic resonance spectroscopic analysis of samples which were freeze-quenched approximately 16 ms after mixing. The direct evidence for complex formation was the loss of the EPR signal of Fe(III) cytochrome P-450cam upon formation of the complex while the EPR signal of reduced putidaredoxin decays with the same kinetics as the appearance of Fe(II) cytochrome P-450. The mechanism of the loss of the EPR signal of cytochrome P-450 upon formation of the complex is not apparent at this time but may involve a conformational change of cytochrome P-450cam following complex formation.  相似文献

1-O-Sinapoyl--glucose:l-malate O-sinapoyltransferase (SMT; EC 2.3.1.) from cotyledons of red radish (Raphanus sativus L. var. sativus) was purified to apparent homogeneity with a 2100-fold enrichment and a 4% recovery. Apparent Mrs of 52 and 51, respectively, were determined by gel filtration and by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). On isoelectric focusing, the SMT resolved into two isoforms which, on SDS-PAGE, showed, slightly different Mrs (SMT I: Mr/isoelectric point = 51/5.75; SMT II: Mr/isoelectric point = 51.5/5.9). The highest activity of SMT was found at pH 6.0 (50% at pH 5.5 and pH 6.5). The temperature maxima in the presence of 10, 50, 100 and 250 mM malate were 22, 30, 35 and 37° C, respectively, with energies of activation of 55, 81, 96 and 121 kJ · mol-1. The enzyme accepted all the hydroxycinnamic acid-glucose esters tested with relative ratios of initial velocity values of 1008545262.6 of 1-O-sinapoyl-, 1-O-feruloyl-, 1-O-caffeoyl-, 1,2-di-O-sinapoyl-, and 1-O-(4-coumaroyl)--glucose. It showed an absolute acceptor specificity for l-malate. d-Malate as second acceptor molecule in standard assays with l-malate inhibited the reaction velocity noncompetitively (K i = 215 mM). The substrate saturation curves were not hyperbolic. The data for sinapoylglucose indicated substrate activation; those for l-malate, substrate inhibition. Kinetic analysis suggests a random bi bi mechanism within two ranges of substrate concentrations, with a kinetically preferred pathway via the enzyme-sinapoylglucose complex indicating a slow-transition mechanism. Basic cellular processes such as electron transport in photosynthesis and respiration require the precise control of iron homeostasis. To mobilize iron, plants have evolved at least two different strategies. The nonproteinogenous amino acid nicotianamine which is synthesized from three molecules of S-adenosyl-L-methionine, is an essential component of both pathways. This compound is missing in the tomato mutant chloronerva, which exhibits severe defects in the regulation of iron metabolism. We report the purification and partial characterization of the nicotianamine synthase from barley roots as well as the cloning of two corresponding gene sequences. The function of the gene sequence has been verified by overexpression in Escherichia coli. Further confirmation comes from reduction of the nicotianamine content and the exhibition of a chloronerva-like phenotype due to the expression of heterologous antisense constructs in transgenic tobacco plants. The native enzyme with an apparent Mr of approximately 105 000 probably represents a trimer of S-adenosyl-L-methionine-binding subunits. A comparison with the recently cloned chloronerva gene of tomato reveals striking sequence homology, providing support for the suggestion that the destruction of the nicotianamine synthase encoding gene is the molecular basis of the tomato mutation.  相似文献
