Integration of viral DNA into host cell DNA is an essential step in retroviral (HIV-1) replication and is catalyzed by HIV-1 integrase. HIV-1 integrase is a novel therapeutic target and is the focus of efforts to identify effective inhibitors that will prevent/or cure HIV infections. Four novel naphtho-gamma-pyrones, belonging to the chaetochromin and ustilaginoidin family, were discovered as inhibitors of HIV-1 integrase from the screening of fungal extracts using a recombinant in vitro assay. These compounds inhibit both the coupled and strand transfer activity of HIV-1 integrase with IC(50) values of 1-3 and 4-12 microM, respectively. The discovery, structure elucidation, chemical modification and the structure-activity relationship of these compounds are described. 相似文献

Subunit composition and architectural structure of the 26S proteasome lid is strictly conserved between all eukaryotes. This eight-subunit complex bears high similarity to the eukaryotic translation initiation factor 3 and to the COP9 signalosome (CSN), which together define the proteasome CSN/COP9/initiation factor (PCI) troika. In some unicellular eukaryotes, the latter two complexes lack key subunits, encouraging questions about the conservation of their structural design. Here we demonstrate that, in Saccharomyces cerevisiae, Rpn5 plays dual roles by stabilizing proteasome and CSN structures independently. Proteasome and CSN complexes are easily dissected, with Rpn5 the only subunit in common. Together with Rpn5, we identified a total of six bona fide subunits at roughly stoichiometric ratios in isolated, affinity-purified CSN. Moreover, the copy of Rpn5 associated with the CSN is required for enzymatic hydrolysis of Rub1/Nedd8 conjugated to cullins. We propose that multitasking by a single subunit, Rpn5 in this case, allows it to function in different complexes simultaneously. These observations demonstrate that functional substitution of subunits by paralogues is feasible, implying that the canonical composition of the three PCI complexes in S. cerevisiae is more robust than hitherto appreciated. 相似文献

We describe a female patient aged 43, who at the age of five was diagnosed with polyostotic fibrous dysplasia (FD). The patient was intermittently treated in our department since the age 33, for approximately 10 years, with intravenous bisphosphonates. At the age of 42 acromegaly was diagnosed incidentally, since clinical manifestations were poor, and, if present earlier, they had been related to FD. Only retrospectively, having biochemical confirmation of GH excess, we could relate them to acromegaly. Because of the involvement of the base of the skull there was no possibility of transphenoidal surgery. Long-acting somatostatin analogues were started, but no response was observed, with IGF-1 and GH being even higher during than before treatment. After the 37-year-history of FD, the occurrence of additional endocrine disorder enabled to make diagnosis of McCune-Albright syndrome (MAS) even in the absence of two out of three classical manifestations such as café-au-lait skin pigmentation and peripheral precocious puberty in the past medical history. 相似文献

Functional reach (FR) significantly increases with additional sensory information from the fifth metacarpal surface of the dominant hand. The purpose of this study was to determine the relationship between the innervation density of the skin above the fifth metacarpal surface of the palm and the increase of FR with an additional light touch. Monocelis lineata consists of a complex of sibling species, widespread in the Mediterranean and Atlantic Ocean. Previous genetic analysis placed in evidence at least four sibling species. Nevertheless, this research was not conclusive enough to fully resolve the complex or to infer the phylogeny/phylogeography of the group. We designed specific primers aiming at obtaining partial sequences of the mtDNA gene Cytochrome c Oxidase subunit I (COI) of M. lineata, and have identified 25 different haplotypes in 32 analyzed individuals. The dendrogram generated by Neighbor-Joining analysis confirmed the differentiation between Atlantic and Mediterranean siblings, as well as the occurrence of at least two Mediterranean sibling species. Thus validated, the method here presented appears as a valuable tool in population genetics and biodiversity surveys on the Monocelis lineata complex. 相似文献

We have studied the seasonal dynamics of abundance and feeding characteristics of three species of calanoid copepods (Acartia spp., Centropages hamatus and Temora longicornis) in the White Sea from the surface water layer (0–10 m), in order to assess their role in the pelagic food web and to determine
the major factors governing their population dynamics during the productive season. These species dominated in the upper water
layer (0–10 m) from June through September, producing up to 3 generations per year. Data on the food spectra revealed all
species to be omnivorous; but some inter- and intraspecific differences were observed. Generally, copepods consumed diatoms,
dinoflagellates and microzooplankton. The omnivory index ‘UC’ (i.e., fatty acid unsaturation coefficient) varied from 0.2
to 0.6, which implied ingestion of phytoplankton. The different degree of selectivity on the same food items by the studied
species was observed, and therefore, successful surviving strategy with minimal overlapping could be assumed. In total, the
populations of the three studied copepod species grazed up to 2.15 g C m−2 day−1 and released up to 0.68 g C m−2 day−1 in faecal pellets. They consumed up to 50% of particulate organic carbon, or up to 85% of phytoplankton standing stock (in
terms of Chl. a), and thus played a significant role in the transformation of particulate organic matter. Seasonal changes in abundance of
the studied species depended mostly on water temperature in the early summer, but were also affected by food availability
