哮喘患者外周血单核细胞血红素氧合酶-1表达水平的研究   总被引:1,自引:0,他引:1  
目的探讨血红素氧合酶-1(HO-1)在哮喘患者外周血单核细胞(PBMC)的表达及与肺通气功能的关系.方法应用免疫组织化学和逆转录聚合酶链反应技术分析18例哮喘患者PBMC的 HO-1蛋白及mRNA水平的表达,测定全血一氧化碳血红蛋白(COHb)的百分比含量、血清总IgE含量、肺通气功能,并与18名健康正常者的结果进行比较.结果哮喘组PBMC中HO-1表达阳性的细胞百分比41.7%±7.44%与正常对照组10.5%±4.36%比较,差异有显著性(P<0.01),哮喘组PBMC HO-1 mRNA表达的平均吸光度值(26.05±4.14)与正常对照组(10.82±4.26)比较,差异亦有显著性(P<0.01),HO-1染色阳性细胞的百分比与FEV1占预计值%、PEFR和MEFR50%均呈显著负相关(r值分别为-0.89,-0.56,-0.51,均P<0.01),与全血COHb的百分比含量及血清总IgE含量呈显著正相关(r值分别为0.80, 0.48, 均P<0.05).HO-1 mRNA的表达水平与1s用力呼气容积(FEV1)占预计值%、顶峰呼气流速(PEFR)和50%肺活量时的最大呼气流速(MEFR50%)均呈显著负相关(r 值分别为-0.89,-0.65,-0.67, 均P<0.01),与全血COHb的百分比含量和血清总IgE含量呈显著正相关(r分别为 0.85和0.62, 均P<0.01).结论哮喘患者PBMC 的HO-1表达水平显著增加,提示HO-1可能参与了哮喘的发病过程,HO-1表达的变化与哮喘患者的病情程度有一定关系.  相似文献

BACKGROUND: Glutamine modulates cytokine production in various tissues but its effects on the production of other inflammatory mediators such as eicosanoids and nitric oxide have not been investigated in human gut. AIM: To evaluate the influence of glutamine on interleukin (IL)-8, IL-6, nitric oxide and prostaglandin E(2) production by human gut. METHODS: Ten fasted volunteers received either enteral glutamine or isonitrogenous amino acids over 6 h in a cross-over design. Series of duodenal biopsies were frozen or cultured for 24 h with 0.5 or 5 mM of glutamine or amino acids. IL-6, IL-8 and PGE(2) were measured in culture media by ELISA and nitrites by Griess assay. mRNA levels for IL-6, IL-8, Cyclooxygenase-2 and NO synthase-2 were assessed in biopsies by RT-PCR. Results in percent, (median [range]) were compared by Wilcoxon test. RESULTS: Glutamine decreased IL-8 and IL-6 in-vitro production: 63 [2-173] vs 100 [19-177] and 37 [5-489] vs 100 [33-431], both P<0.05. IL-8 mRNA level also decreased in biopsies cultured with 5 mM glutamine: 26 [13-142] vs 92 [34-215], P<0.05. Nitrites and PGE(2) concentrations were not significantly affected by glutamine. CONCLUSION: Glutamine has a specific inhibitory effect on pro-inflammatory cytokine production in the gut and may contribution to the modulation of intestinal inflammation.  相似文献

Sampling studies were conducted in coffee plantations in South Yunan to assess the incidence, symptoms, and intensity of damage by three stem borers: Xylotrechus quadripes (Chevrolat), Acalolepta cervina (Hope), and Bacchisa sp. near pallidiventris (Thomson). Of 5,690 plants sampled in eight plantations, 440 were infested with A. cervina, 63 with X. quadripes, and three with B. pallidiventris. Plants 5-7 yr old were 10 times more heavily infested with X. quadripes than 3- to 4-yr-old plants, whereas both age groups of plants had similar levels of infestation with A. cervina. Larval galleries of the three borer species markedly differ: A. cervina and B. pallidiventris larvae develop in subcortical galleries in the main stem (A. cervina) and lateral branches (B. pallidiventris), whereas larval galleries of X. quadripes intermittently punctuate and transverse the xylem of main stems or lateral branches. Significantly more plant tissue was damaged in stems infested with X. quadripes than in those infested with A. cervina or B. pallidiventris. Stems infested with A. cervina or B. pallidiventris generally had only one or a few pupation chambers, whereas stems infested with X. quadripes contained numerous chambers. Quantitative and qualitative data collected through this study provide farmers with diagnostic tools to determine which borer species infested coffee plants. Comparison of life history traits and intensity of damage for the three borer species indicates that X. quadripes is the most severe pest of coffee in Yunan, and suggests that populations of X. quadripes have the greatest potential to steadily increase with time.  相似文献

A stable cell line (GT2-LPk) derived from LLC-Pk was created in which endogenous DNA topoisomerase II alpha (topoII alpha) protein was downregulated and replaced by the expression of topoII alpha fused with enhanced green fluorescent protein (EGFP-topoII alpha). The EGFP-topoII alpha faithfully mimicked the distribution of the endogenous protein in both interphase and mitosis. In early stages of mitosis, EGFP-topoII alpha accumulated at kinetochores and in axial lines extending along the chromosome arms. During anaphase, EGFP-topoII alpha diminished at kinetochores and increased in the cytoplasm with a portion accumulating into large circular foci that were mobile and appeared to fuse with the reforming nuclei. These cytoplasmic foci appearing at anaphase were coincident with precursor organelles of the reforming nucleolus called nucleolus-derived foci (NDF). Photobleaching of EGFP-topoII alpha associated with kinetochores and chromosome arms showed that the majority of the protein rapidly exchanges (t1/2 of 16 s). Catalytic activity of topoII alpha was essential for rapid dynamics, as ICRF-187, an inhibitor of topoII alpha, blocked recovery after photobleaching. Although some topoII alpha may be stably associated with chromosomes, these studies indicate that the majority undergoes rapid dynamic exchange. Rapid mobility of topoII alpha in chromosomes may be essential to resolve strain imparted during chromosome condensation and segregation.  相似文献

Beta-mannosidase, a high-salt-soluble enzyme, increases in activity in seeds of tomato prior to the completion of germination. This increase occurs in both the lateral and micropylar endosperm and becomes more evident during post-germinative seedling growth. The beta-mannosidase activity profile is similar to that of endo beta-mannanase although it is the first to increase in the lateral endosperm. Tomato seed beta-mannosidase was purified to homogeneity and its cDNA (LeMside1) obtained by 3'-RACE PCR using oligonucleotide sequences based on four peptide sequences obtained from the purified enzyme. The derived amino acid sequence of the tomato beta-mannosidase shows the enzyme is a member of the Glycosyl Hydrolases Family 1 (GHF1) but has a very low sequence identity with that of beta-mannosidases from non-plant sources; no other plant sequence for the enzyme is known. There appears to be only one gene encoding beta-mannosidase in tomato, the sequence of which has been determined (LeMSide2). Its expression occurs first in the micropylar endosperm, and then declines after germination. This is followed by an increase in its expression in the lateral endosperm, which precedes that of the gene for endo beta-mannanase. Expression of the beta-mannosidase gene increases appreciably in the growing seedling embryo. With this report, the cloning of all three of the enzymes involved in galactomannan mobilization (endo beta-mannanase, alpha-galactosidase and beta-mannosidase) in tomato seeds has now been achieved.  相似文献

Serum and urine chromium concentrations in elderly diabetics   总被引:5,自引:0,他引:5  
The serum and urine chromium concentrations of 57 diabetics and 55 normal fasting subjects were determined by atomic absorption spectrometry (AAS). Our results indicate that the chromium concentration ranges of serum and urine for diabetics are 0.22–0.36 and 4.54–5.90 μg/L, respectively, significantly lower than 0.66–0.84 7.80–9.68 μg/L for the normal (P<0.001), which implies that the elderly diabetics probably lack chromium. Further, it was found that the urine chromium level of the female diabetics was substantially higher than that of the male in the same age group (P<0.01), whereas the serum chromium level was almost the same. However, the urine chromium concentration increases with aging, no matter who the diabetics or the controls are. The serum chromium concentrations of the 24 cases patients with 2-h oral glucose tolerance test (OGTT) were significantly lower than that of those with empty stomach, whereas the urine chromium exhibits a contrary tendency. Our data indicate that the chromium lost and excreted from human body increases with aging and is related to the diabetics. Thus, it is recommended to supplement a certain amount of chromium to the elderly diabetics according to their nutritional level.  相似文献
