Isolates ofTyphula incarnata, a snow mold fungus, were collected from four localities with different winter climates. Their ecological traits such as mycelial growth rate, sclerotium size, carpogenic germination of sclerotia, and aggressiveness were compared between populations in order to reveal infraspecific differentiation associated with climatic differences. Population variability was evident only in sclerotium germination: isolates from more snowy localities germinated faster than those from less snowy localities.T. incarnata is regarded as a versatile pathogen with no specialized forms in contrast withT. ishikariensis. The genetic diversity of bumblebees can be adversely affected by habitat degradation. An overabundance of deer has altered
the composition and diversity of herbaceous plants in many places of the world, resulting in decreases of herbaceous flowers.
Populations of Bombus diversus may be strongly affected by this degradation of habitat in the Ashiu primary beech forest in Kyoto, Japan. To estimate the
effects of deer browsing on B. diversus populations, we analyzed and compared the genetic diversity of the extant population in Ashiu to museum specimens collected
prior to heavy deer browsing in Ashiu (1980s) and the extant population in Hyonosen primary beech forest in Tottori, Japan,
which has not been as severely degraded by deer. We successfully amplified DNA from ~20-year-old museum specimens and determined
the genetic diversity of B. diversus in Ashiu populations from the 1980s. Results were analyzed for indications of a bottleneck as well as estimates of Ne, allelic richness, rare allelic richness, expected heterozygosity, and the effective number of alleles. Our findings did
not reveal clear evidence of degradation in genetic diversity of the extant Ashiu population compared to the museum specimens
or to the Hyonosen population. Thus, the Ashiu population of B. diversus appears to have maintained a level of genetic diversity during 20 years irrespective of habitat degradation and the levels

Protein kinase C (PKC) is a family of serine/threonine kinases involved in various signal transduction pathways. We investigated the roles of PKC in the regulation of group IIA secreted phospholipase A2 (sPLA2-IIA) expression in cytokine-stimulated rat fibroblastic 3Y1 cells. Here we show that the induction of sPLA2-IIA by proinflammatory cytokines was under the control of both classical cPKCα and atypical aPKCλ/ι pathways by using PKC inhibitors, a PKC activator, and PKC knockdowns. Treatment of 3Y1 cells with PKC selective inhibitors having broad specificity, such as chelerythrine chloride and GF109203X, blocked IL-1β/TNFα-dependent induction of sPLA2-IIA protein in a dose-dependent manner. Treatment with the PKC activator phorbol 12-myristate 13-acetate (PMA), which activates cPKC and novel nPKC isoforms, markedly attenuated the cytokine-dependent induction of sPLA2-IIA expression. In comparison, 24-h pretreatment with PMA, which down-regulates these PKC isoforms, markedly enhanced sPLA2-IIA expression. Results with short hairpin RNA (shRNA)-mediated knockdown of PKC isoforms revealed that the cytokine-induced sPLA2-IIA expression was markedly enhanced in cPKCα knockdown cells compared to those in replicate control cells. In contrast, knockdown of the aPKCλ/ι isoform reduced the cytokine-induced expression of sPLA2-IIA. These results suggest that the aPKCλ/ι pathway is required for the induction of sPLA2-IIA expression and that the cPKCα pathway acts as a negative regulator of sPLA2-IIA expression in cytokine-stimulated rat fibroblasts. 相似文献
To elucidate population-increasing factors in the diving beetle Cybister tripunctatus lateralis (Fabricius) (Coleoptera: Dytiscidae) in Japan in recent years, life histories and oviposition patterns were compared among three endangered diving beetle species, Cybister brevis Aubé (qualified by the Japanese Red Data List as ‘near threatened’), Cybister chinensis Motschulsky (vulnerable), and C. tripunctatus lateralis (vulnerable). Oviposition in C. brevis, C. chinensis, and C. tripunctatus lateralis was observed from late April to mid-June, from late April to early July, and from late May to mid-August, respectively, under semi-outdoor conditions. There were no interspecies differences in total hatchling production during the reproductive season. In rearing experiments at various temperatures (20, 23, 25, 28, and 30 °C), the mortality of C. tripunctatus lateralis larvae was higher at 20 °C, and gradually lower with increasing temperature up to 30 °C. Adult body size of females in C. tripunctatus lateralis is larger than that of males but there were no significant differences among temperatures (25–30 °C). Cybister brevis had a higher emergence rate at 23–28 °C than at 20 and 30 °C. In C. brevis, the body size of adults reared at 25 or 28 °C was significantly larger than at other temperatures. Cybister chinensis did not differ in emergence rate and adult body size among the five temperature conditions. The developmental zero (i.e., the lower developmental threshold) from the first instar to adult emergence was 11.1 °C for C. brevis, 8.7 °C for C. chinensis, and 16.8 °C for C. tripunctatus lateralis. We speculate how the influence of global warming may have a positive impact on the growth and survival of C. tripunctatus lateralis. 相似文献
Understanding species composition and other characteristics of remnant trees that survive large-scale low-frequency disturbances such as tsunamis is an important step in monitoring and managing the process of recovery. This research was implemented in a study area along the shores of Sendai Bay, a region that was heavily damaged by the tsunami following the Great East Japan Earthquake of 2011. Field surveys of all the remnant patches of trees in the study area were conducted in 2014, three years after the disturbance. A total of 202 remnant patches were identified, and all the tree species, including both canopy trees and newly established seedlings, were recorded for each patch. The identified trees totaled 88 species, which were classified into three indicator-species types by a hierarchical cluster analysis. Comparisons based on species composition and similarity showed that species composition and other characteristics of the remnant patches were influenced by vegetation histories and other factors that only become visible at the regional landscape scale. The number of tree and seedling species per unit area was highest in patches that originated from pine plantations. Seven of the 25 seedling species identified were not present in any of the canopy layers in the study area, indicating that they dispersed from outside this area. These results show that, despite the immense scale of the tsunami, many diverse patches of trees managed to survive the disturbance. These patches function as hubs for species dispersion and accelerate recovery of species diversity in the disturbed area.
Processionary caterpillars of Thaumetopoea pityocampa (in Europe) and Ochrogaster lunifer (in Australia) (Lepidoptera: Notodontidae) form single files of larvae crawling head-to-tail when moving to feeding and pupation sites. We investigated if the processions are guided by polarization vision. The heading orientation of processions could be manipulated with linear polarizing filters held above the leading caterpillar. Exposure to changes in the angle of polarization around the caterpillars resulted in corresponding changes in heading angles. Anatomical analysis indicated specializations for polarization vision of stemma I in both species. Stemma I has a rhabdom with orthogonal and aligned microvilli, and an opaque and rugged surface, which are optimizations for skylight polarization vision, similar to the dorsal rim of adult insects. Stemmata II-VI have a smooth and shiny surface and lobed rhabdoms with non-orthogonal and non-aligned microvilli; they are thus optimized for general vision with minimal polarization sensitivity. Behavioural and anatomical evidence reveal that polarized light cues are important for larval orientation and can be robustly detected with a simple visual system. 相似文献
Current sampling methods for estimating infestation rates of tracheal mites in colonies of the honey bee,Apis mellifera, assume that infested bees are randomly distributed and that temporal fluctuations in infestation rates occur homogeneously throughout the colony. We examined these assumptions. Samples of bees were collected from up to five locations in each of eight colonies, and colonies were sampled several times throughout the year. Estimates of infestation rates varied, depending on the location in the colony from which a sample was obtained. Temporal fluctuations in infestation rates did not always occur homogeneously with respect to sampling location. These results demonstrate that assumptions of current sampling protocols for estimating tracheal-mite infestation rates are often violated. Consequently, estimates derived using these methods may not be accurate, and conclusions based on such estimates may not be valid. 相似文献
