Dideoxy-epigallocatechin gallate (DO-EGCG) (2), a simplified analog of naturally occurring EGCG (1), was efficiently prepared by directly introducing a ketone group at C3 and successive reduction to the sec-alcohol with 2,3-cis stereochemistry. Compound 2 showed potent anti-influenza virus activity, indicating that the hydroxyl substituents on the A-ring are not crucial for anti-influenza virus activity.  相似文献

Biomineralization on porous polyester membranes was examined using an improved alternate soaking process (ASP). The effect of ion migration for the formation of hydroxyapatite (HAp) was shown to be crucial. Ion migration was improved by reducing the surface tension by mixing ethanol into an aqueous solution. The resulting hybrid materials were evaluated in terms of calcium content; structure using scanning electron microscopy (SEM), X-ray diffraction (XRD), and infrared spectroscopy (IR); and protein adsorption. The amount of formed HAp was controlled by the number of ASP cycles and also through the ethanol content of the mixed solvent. As the formation of HAp increased, the formed structure could be verified using SEM, IR, and XRD. Protein adsorption was investigated using albumin, gamma-globulin, and fibrinogen, and the amount of adsorbed protein was well-correlated with that of the formed HAp. This result shows that the total amount of the adsorbed proteins can be regulated by the HAp content. In summary, a tunable protein reservoir was formed on a porous polyester membrane.  相似文献

The formation of thioester-linked glutathione (GSH) conjugates of bile acids (BAs) is presumed to occur via trans-acylation reactions between GSH and reactive acyl-linked metabolites of BAs. The present study examines the chemical reactivity of cholyl-adenylate and cholyl-CoA thioester, acyl-linked metabolites of cholic acid (CA), with GSH to form CA-GSH conjugate in vitro. The authentic specimen of CA-GSH was synthesized along with GSH conjugates of four common BAs found in the human body. Their structures were confirmed by proton-nuclear magnetic resonance spectroscopy and electrospray ionization (ESI)-tandem mass spectrometry in positive- and negative-ion modes. Incubation of cholyl-adenylate or cholyl-CoA thioester with GSH was carried out at pH 7.5 and 37 degrees C for 30 min, with analysis of the reaction mixture by liquid chromatography/ESI-tandem mass spectrometry, where CA-GSH was detected on the product ion mass chromatograms monitored with stable and abundant dehydrated positive-ion [M+HH(2)O](+) at m/z 680.3 and fragmented negative-ion [GSHH](-) at m/z 306.0, and was definitely identified by CID spectra by comparison with those of the authentic sample. The results show that both cholyl-adenylate and cholyl-CoA thioester are able to acylate GSH in vitro.  相似文献

This protocol describes a procedure for the synthesis of syn-beta-amino alpha-substituted aldehydes, versatile intermediates in synthetic organic chemistry, via asymmetric, direct, one-pot, three-component, cross-Mannich reaction of two different aldehydes. The reaction consists of two steps; one is the formation of imine by the reaction of aldehyde and p-anisidine in the presence of Pro, and the second step is the enantioselective addition reaction of enamine generated from the other aldehyde and Pro with the imine generated in the first step. As the aldehyde easily racemizes, gamma-amino alcohol was isolated and characterized after reduction. The yield and diastereo- and enantioselectivities are generally excellent. It will take approximately 26 h to complete the protocol: 0.5 h to set up the reaction, 20.5 h for the reaction and 5 h for the isolation and purification.  相似文献

High aluminum (Al) concentration in soil solution is the most important factor restricting plant growth in acidic soils. However, various plant species naturally grow in such soils. Generally, they are highly tolerant to Al, but organic acid exudation, the most common Al tolerance mechanism, cannot explain their tolerance. Lower phospholipid and higher sterol proportions in root plasma membrane enhance Al tolerance. Other cellular components, such as cell walls and phenolics, may also be involved in Al tolerance mechanisms. In this study, the relationships between these cellular components and the Al tolerance mechanisms in Melastoma malabathricum and Melaleuca cajuputi, both highly Al‐tolerant species growing in strongly acidic soils, were investigated. Both species contained lower proportions of phospholipids and higher proportions of sterols in roots, respectively. Concentrations of phenolics in roots of both species were higher than that of rice; their phenolics could form chelates with Al. In these species, phenolic concentrations and composition were the same irrespective of the presence or absence of Al in the medium, suggesting that a higher concentration of phenolics is not a physiological response to Al but a constitutive characteristic. These characteristics of cellular components in roots may be cooperatively involved in their high Al tolerance.  相似文献

The phosphatidylinositol 3-kinase signaling pathway in vascular endothelial cells is important for systemic angiogenesis and glucose metabolism. In this study, we addressed the precise role of the 3-phosphoinositide-dependent protein kinase 1 (PDK1)-regulated signaling network in endothelial cells in vivo, using vascular endothelial PDK1 knockout (VEPDK1KO) mice. Surprisingly, VEPDK1KO mice manifested enhanced glucose tolerance and whole-body insulin sensitivity due to suppression of their hepatic glucose production with no change in either peripheral glucose disposal or even impaired vascular endothelial function at 6 months of age. When mice were fed a standard diet at 6 months of age and a high-fat diet at 3 months of age, hypertrophy of epididymal adipose tissues was inhibited, adiponectin mRNA was significantly increased, and mRNA of MCP1, leptin, and TNFα was decreased in the white adipose tissue of VEPDK1KO mice in comparison with controls. Consequently, both the circulating adiponectin levels and the activity of hepatic AMP-activated protein kinase were significantly increased, subsequently enhancing whole-body insulin sensitivity and energy expenditure with increased hepatic fatty acid oxidation in VEPDK1KO mice. These results provide the first in vivo evidence that lowered angiogenesis through the deletion of PDK1 signaling not only interferes with the growth of adipose tissue but also induces increased energy expenditure due to amelioration of the adipocytokine profile. This demonstrates an unexpected role of PDK1 signaling in endothelial cells on the maintenance of proper glucose homeostasis through the regulation of adipocyte development.  相似文献

During epithelial junctional development, both vesicle transport and reorganization of the actin cytoskeleton must be spatiotemporally regulated. Coordination of these cellular functions is especially important, but the precise mechanism remains elusive. Previously, we identified junctional Rab13-binding protein (JRAB)/molecules interacting with CasL-like 2 (MICAL-L2) as an effector of the Rab13 small G protein, and we found that the Rab13-JRAB system may be involved in the formation of cell-cell adhesions via transport of adhesion molecules. Here, we showed that JRAB interacts with two actin-binding proteins, actinin-1 and -4, and filamentous actin via different domains and regulates actin cross-linking and stabilization through these interactions. During epithelial junctional development, JRAB is prominently enriched in the actin bundle at the free border; subsequently, JRAB undergoes a Rab13-dependent conformational change that is required for maturation of cell-cell adhesion sites. These results suggest that Rab13 and JRAB regulate reorganization of the actin cytoskeleton throughout epithelial junctional development from establishment to maturation of cell-cell adhesion.  相似文献

Disruption of cellular adhesion is an essential pathobiologic step leading to tumor dissemination. Mucin 1 (MUC1) is a mucinous glycoprotein expressed at the surfaces of epithelial cells in many tissues and their carcinomas. MUC1 plays crucial roles in tumor invasion and metastasis, especially in opposing cell adhesion. We have shown that virus infection, specifically by the human tumor virus Epstein-Barr virus (EBV) induces a spectrum of cellular invasiveness and metastasis factors. Here we show that expression of MUC1 is increased in diverse latently EBV-infected cell lines that express latent membrane protein 1 (LMP1), the main viral oncoprotein, and that the level of MUC1 was suppressed by expression of a dominant-negative mutant of LMP1. Expression of LMP1 in EBV-negative nasopharyngeal cell lines induces expression of MUC1 through activation of the MUC1 promoter via binding of STAT1 and STAT3. Finally, LMP1 reduces cell adhesion ability, which is restored by inhibition of MUC1 expression with MUC1 small interfering RNA (siRNA). In addition, LMP1 increases cell invasiveness, which is suppressed by MUC1 siRNA. Thus, LMP1 induces MUC1, a factor important in an early step of detachment and release of tumor cells, which along with induction of other invasiveness and angiogenic factors may combine to act in a complex sequential process that culminates in metastasis of EBV-infected tumor cells.  相似文献

This study examines pressurized hot water extraction of anthraquinones from dried roots of Morinda citrifolia (Noni). The effects of various operating conditions such as water temperature (110, 170, and 220 degrees C), and water flow rate (2, 4, and 6 mL min(-1)) on extraction yield and extraction rate were determined. At 220 degrees C, the extraction yield was the highest and was approximately 43.6 mg g(-1). Subcritical water extraction at 4 mL min(-1) or higher was found to be able to recover all the antraquinones present in the roots within 3 h of extraction, even at 170 degrees C. Pressure had no significant effect on the results for the range of temperatures studied. The flow rate of 6 mL min(-1) resulted in the highest extraction rate, but the extraction efficiency, as measured by the amount of anthraquinones extracted per unit volume of water was lower than that of 4 mL min(-1). Anthraquinones solubility in pressurized hot water was determined. The results indicate a presence of a mass transfer limitation in the extraction process from solid matrix.  相似文献

The sugar-binding specificities of C-type lectins isolated from marine invertebrates were investigated by frontal affinity chromatography (FAC) using 100 oligosaccharides. The lectins included BRA-2 and BRA-3, multiple lectins from the hemolymph of the acorn barnacle, Megabalanus rosa, and BRL from the acorn barnacle, Balanus rostatus. The diverse sugar-binding specificities of the C-type lectins were determined by FAC analysis. BRA-2 recognized alpha2-6 sialylation but not alpha2-3 sialylation on glycans. On the other hand, BRA-3 showed high affinity for oligosaccharides with alpha-linked non-reducing terminal galactose, but not for sialylated forms, and BRL showed enhanced recognition activity towards Lewis(x) and Lewis(a) epitopes.  相似文献
